AB - Lakes and reservoirs are the main water resources for household, irrigation, and industrial use. Hence, it is necessary to control water pollution and have reliable information on water quality, which refers to its suitability for a particular purpose. The main objective of this study was to evaluate the suitability of water for drinking and irrigation purposes from ten artificial lakes on the territory of the Republic of Serbia. The physicochemical parameters were determined using standard analytical methods, whereas the optical emission spectrometry with inductively coupled plasma, ICP-OES, was used to determine the concentrations of 23 macro-, micro-, and trace elements in water. Health risk indices of drinking water and its suitability for irrigation purposes were evaluated. The results of physicochemical parameters, except for total organic matter, indicated that most samples were within the recommended values. Among analyzed elements, the concentration of Mg was elevated in the water of the lakes Srebrno and Prvonek. The results of health risk assessment clearly showed that there was no acute health risk (HQA < 1), whereas long-term risk (HQL) in the studied lakes was observed with respect to As (lakes Grlište and Srebrno), Mn (lakes Srebrno and Prvonek), Ni (Lake Vrutci), and Sr (Lake Grlište). The irrigation criteria indicated that the water from the examined lakes had satisfactory quality for this purpose, except for residual sodium carbonates (RSC). This criterion indicated low quality or unsuitability of the water in 7 lakes. The main outcomes provided valuable information on the suitability of water from the artificial lakes for intended uses.

AB - Ever since ancient times, Allium species have played a significant role in the human diet, in traditional medicine for the treatment of many ailments, and in officinal medicine as a supplemental ingredient. The major metabolites of alliums, as well as their antioxidant and antimicrobial properties, with an emphasis on the species most represented in the Balkan region, are discussed in this review. Due to its richness in endemic species, the Balkan region is considered the genocenter of alliums. There are 56 recorded Allium species in the Balkans, and 17 of them are endemic. The most common and well-studied Allium species in the Balkans are A. cepa (onion), A. sativum (garlic), A. ampeloprasum (leek), A. schoenoprasum (chives), A. fistulosum (Welsh onion), and A. ursinum (wild garlic or bear’s garlic), which are known for their pungent taste and smell, especially noticeable in garlic and onion, and attributed to various organosulfur compounds. These plants are valued for their macronutrients and are used as desirable vegetables and spices. Additionally, phytochemicals such as organosulfur compounds, phenolics, fatty acids, and saponins are associated with the antioxidant and antimicrobial properties of these species, among many other bioactivities. All parts of the plant including the bulb, peel, clove, leaf, pseudostem, root, flower, and seed exhibit antioxidant properties in different in vitro assays. The characteristic phytocompounds that contribute to the antimicrobial activity of alliums include allicin, ajoene, allyl alcohol, and some diallyl sulfides. Nanoparticles synthesized using Allium species are also recognized for their notable antimicrobial properties. © 2023 by the authors.

AB - The demand for ready-to-use functional foods is high, which encourages manufacturers to develop new, nutritionally valuable products. As an excellent source of biologically active compounds, beetroot (Beta vulgaris L.) is considered to have highly beneficial effects on health. This research aimed to evaluate the impact of replacing spelt flour (SF) with 15%, 20% and 25% beetroot powder (BP). The physicochemical and functional properties of biscuits baked at different temperatures (150 and 170 °C) were followed at the beginning, and after 3 and 6 months of storage as standard conditions. Moisture content and water activity (aw) gave insight into the biscuits’ shelf life. The value of aw from 0.35 to 0.56 indicated appropriate storability. Dietary fiber content in fresh biscuits ranged from 6.1% to 7.6%, protein from 9.2% to 8.9% and sugar from 30.6% to 35.9%. The content of betalain, total polyphenols and flavonoids, and antioxidant activity (DPPH, FRAP) increased with beetroot powder content incorporated. A slight decrease of all the mentioned parameters during the storage indicated satisfied retention of bioactive molecules. The content of prevalent phenolic compounds gallic and protocatechuic acid, identified by HPLC, decreased from 22.2–32.0 and 21.1–24.9 in fresh biscuits to 18.3–23.4 and 17.3–20.3 mg/100 g upon six months of storage, respectively. An increase of the L* and a* and a decrease of the b* coordinate values, compared with the control sample without beetroot values, was noticed as well as the expected level of their change during the storage. The obtained results indicated that biscuits enriched with beetroot powder showed a significantly improved functional, nutritional and antioxidant potential during storage. © 2023 by the authors.

AB - Sorghum grain (Sorghum bicolor L. Moench) is a gluten-free cereal with excellent nutritional value and is a good source of antioxidants, including polyphenols, as well as minerals with proven health benefits. Herein, the phenolic composition, elemental profile, and antioxidant activity of sixteen food-grade sorghum grains (S1–S16) grown under agroecological conditions in Serbia were determined. Nine phenolic compounds characteristic of sorghum grains, such as luteolinidin, 5-methoxyluteolinidin, luteolidin derivative, luteolidin glucoside, apigeninidin, 7-methoxyapigeninidin, apigeninidin glucoside, and cyanidin derivative, were quantified. The antioxidant potential of the analyzed sorghum grains was evaluated by UV/Vis (DPPH, ABTS, and FRAP) and Electron Paramagnetic Resonance spectroscopy (hydroxyl and ascorbyl radical scavenging assays). The content of macro- and microelements was determined by Inductively Coupled Plasma Optical Emission spectroscopy. Theoretical daily intakes of selected major and trace elements were assessed and compared with the Recommended Daily Allowance or Adequate Intake. Sample S8 had the highest amount of phenolic compounds, while S4, S6, and S8 exhibited the strongest antioxidative potential. The sorghum studied could completely satisfy the daily needs of macro- (K, Mg, and P) and microelements (Se, Zn, Fe). Pattern recognition techniques confirmed the discrimination of samples based on phenolic profile and elemental analysis and recognized the main markers responsible for differences between the investigated samples. The reaction between hydroxyl radicals and luteolinidin/apigeninidin was investigated by Density Functional Theory and thermodynamically preferred mechanism was determined.

AB - Cocoa beans are part of the cocoa plant fruit (Theobroma cacao L.) used to prepare various products such as chocolate, cocoa butter, jelly, liqueurs, cosmetics, etc. Dark chocolate is consumed worldwide by different populations and is known for its good taste, making it one of the most favoured food products. This work aimed to determine the content of total polyphenols (TPC), total flavonoids (TFC), and the antioxidant potential measured through the ability to scavenge DPPH free radicals (DPPH), ferric reducing power (FRAP), and total antioxidant capacity (TAC), as well as major and trace elements contained in twelve commercially available dark chocolate samples, with cocoa content ranging from 40% to 99%. The total polyphenols content ranged between 10.55 and 39.82 mg/g GAE, while the total flavonoid content was from 10.04 to 37.85 mg/g CE. All applied antioxidant assays indicate that the sample with the highest cocoa percentage shows the greatest antioxidant activity (DPPH: 48.34% of inhibition; FRAP: 89.00 mg/g GAE; TAC: 83.86 mg/g AAE). Statistical methods were applied to establish the differences between the samples concerning TPC, TFC, DPPH, FRAP and TAC, as well as to differentiate the samples according to the mineral content. The results indicated that the differences in TPC and TFC between different samples depended on the cocoa content and the addition of dried fruit pieces. A good correlation between antioxidant potency composite index (ACI) and declared cocoa content was noticed (R2 = 0.8034), indicating that the declared percentage of cocoa is a reliable indicator for antioxidant activity of analysed dark chocolate samples. The nutritional evaluation proved that the studied chocolate samples were an excellent source of Mg, Fe, Mn and Cu.

AB - The aim of this study was to examine the quality of real samples of drinking water, collected from the territory of the city of Požega and the surrounding rural areas. An analytical technique of inductively coupled plasma with optical emission spectrometry (ICP-OES) was used to determine the concentrations of macro-and microelements in water samples. The obtained results were compared with the maximum allowed concentrations of elements specified by the national Regulation on hygienic quality of drinking water, as well as the recommendations of the World Health Organization (WHO) and the US Environmental Protection Agency (EPA). The analysed waters were low-mineralized (<1000 mg/L), calcium waters, with low sodium content. The concentrations of all tested elements in the water samples were within the allowed values. There were obvious differences in chemical composition between surface water samples and groundwater samples. All tested waters can be safely used for irrigation, because the risk of soil salinization, sodium accumulation and harmful effects of boron on crops was assessed as insignificant.

AB - Even though platinum(II/IV) complexes are found to be very active as anticancer agents, they have many well-known limitations. These drawbacks reflect through severe side-effects due to damaging healthy tissue and resistance. Exploring non-platinum metal complexes emerged as essential since they might exhibit different mechanism of action as well as reduced side effects, than platinum(II)-based ones. In this review, the progress in the field of anticancer chemistry of palladium(II)-based complexes will be given highlighting the close relationship between their structural preferences and cytotoxic ability. Activity and structure of a significant number of palladium complexes described in literature will be conjoint, and probable in vivo mechanism of palladium(II) species discussed. It is shown that bidentate chelating amines (e.g. (S,S)-R2edda-type ligands; R2edda = O,O’-dialkyl-ethylenediamine-N,N’-diacete esters) as ligands could play a crucial role in the stability of complexes and their ability to express biological activity. Complexes with general formulae [PdCl2{(S,S)-R2edda-type}], demonstrate good to moderate antiproliferative activity versus various cancer cell lines with emphasizing of O,O’-dipropyl-(S,S)-ethylenediamine-N,N’-di-2-(4-methyl)pentanoate, [PdCl2(S,S)-(n-Pr)2eddl] (most powerful against chronic lymphocytic leukemia cells, CLL). Structural features and diastereoisomers arising from nitrogen coordination in square-planar palladium(II) complexes with bidentate κ2N,N’ or tridentate κ2N,N’κO, ligands will be summarized and discussed. In vitro cytotoxicity of these complexes on a number of tumor cell lines will give an insight in structure–activity relationships which may lead to the divergence from platinum based drugs. Additionally, an overview of antimicrobial activity is presented.

AB - Xanthohumol (XN) and isoxanthohumol (IXN), prenylated flavonoids from Humulus lupulus, have been shown to possess antitumor/cancerprotective, antioxidant, antiinflammatory, and antiangiogenic properties. In this study, mesoporous silica (SBA-15) was loaded with different amounts of xanthohumol and isoxanthohumol and characterized by standard analytical methods. The anticancer potential of XN and IXN loaded into SBA-15 has been evaluated against malignant mouse melanoma B16F10 cells. When these cells were treated with SBA-15 containing xanthohumol, an increase of the activity correlated with a higher immobilization rate of XN was observed. Considering the amount of XN loaded into SBA-15 (calculated from TGA), an improved antitumor potential of XN was observed (IC50 = 10.8 ± 0.4 and 11.8 ± 0.5 µM for SBA-15|XN2 and SBA-15|XN3, respectively; vs. IC50 = 18.5 ± 1.5 µM for free XN). The main mechanism against tumor cells of immobilized XN includes inhibition of proliferation and autophagic cell death. The MC50 values for SBA-15 loaded with isoxanthohumol were over 300 µg/mL in all cases investigated. © 2022 by the authors.

The main mechanism against tumor cells of immobilized XN includes inhibition of proliferation and autophagic cell death. The MC50 values for SBA-15 loaded with isoxanthohumol were over 300 µg/mL in all cases investigated. © 2022 by the authors.",

AB - The bisphenol A (BPA) concentration was determined in 12 surface water samples of the Vrbas River and its five tributaries. The samples were taken in the area that belongs to the city of Banja Luka (Bosnia and Herzegovina). BPA was isolated using micro liquid-liquid extraction followed by derivatization and gas chromatography-mass spectrometry analysis (GC-MS). Silylation was used as a derivatization method to increase volatility and allow the GC-MS determination of BPA. The limits of detection (LOD) and quantification (LOQ), obtained by validating the procedure, were determined at 4 and 10 ng L-1, respectively. The concentrations of BPA were ranged between 33 and 354 ng L-1, and all were above the LOQ value. The lowest amount of BPA was found in the sample collected in the river Vrbas, near Švrakava estuary upstream from the city of Banja Luka. The highest concentration of BPA was recorded at the confluence of the Crkvena and Vrbas rivers, which is located in the city center. This study shows that population and human activity could affect the level of BPA in the environment.

AB - New Ni (II) and Cu (II) complexes with pyridoxal-semicarbazone were synthesized and their structures were solved by X-ray crystallography. This analysis showed the bis-ligand octahedral structure of [Ni(PLSC-H)2]·H2O and the dimer octahedral structure of [Cu(PLSC)(SO4)(H2O)]2·2H2O. Hirshfeld surface analysis was employed to determine the most important intermolecular interactions in the crystallographic structures. The structures of both complexes were further examined using density functional theory and natural bond orbital analysis. The photocatalytic decomposition of methylene blue in the presence of both compounds was investigated. Both compounds were active toward E. coli and S. aureus, with a minimum inhibition concentration similar to that of chloramphenicol. The obtained complexes led to the formation of free radical species, as was demonstrated in an experiment with dichlorofluorescein-diacetate. It is postulated that this is the mechanistic pathway of the antibacterial and photocatalytic activities. Cyclic voltammograms of the compounds showed the peaks of the reduction of metal ions. A molecular docking study showed that the Ni(II) complex exhibited promising activity towards Janus kinase (JAK), as a potential therapy for inflammatory diseases, cancers, and immunologic disorders.

AB - Medicinal plants represent a rich source of phytochemicals, many of which possess diverse biological activities, including antioxidants. The objectives of this study were to assess antioxidant, antibacterial and anticancer activities of savory (Satureja montana L.) ethanolic extracts and to monitor the changes of oxidative status during long-term storage conditions of the cold pressed sunflower oil (CPSU) with the addition of extracts at various concentrations. Extracts were obtained by Soxhlet extraction and ultrasound-assisted maceration, using 70% and 96% ethanol. Antioxidant activity was evaluated by DPPH and ꞵ-carotene bleaching assays. In both assays, extracts showed remarkable antioxidative properties which were equal or stronger than those of BHT and BHA. Also, regardless of the extraction method and assay applied, the 70% ethanol extracts proved to be more potent than 96% ethanol ones. For antibacterial activity assay, minimal inhibitory concentrations (MIC) were obtained by the microdilution method. Tested Gram-negative bacteria (E. coli and S. typhimurium) were more resistant than Gram-positive (S. aureus and L. monocytogenes) with MIC=1,25–10 μg/mL and 0,0781–0,3125 μg/mL, respectively. As results obtained by MTT test, the extracts didn’t show anticancer activity (IC50>100 μg/L) against tested tumor cell lines: PC-3 (prostate), HT-29 (colon) and HeLa (cervix) in the applied concentration range. For investigation of oxidative stability of CPSU, only 70% ethanol extracts were used. Samples of CPSU (with addition of 250, 500 and 1000 ppm of extracts), as well as control samples (with addition of 200 ppm of BHT and CPSU without any additives), were stored at room temperature for six months. In order to determine the level of oxidative changes, samples were analyzed for peroxide, anisidine value and conjugated dienes and trienes. Based on obtained results, it can be concluded that savory extracts are powerful natural antioxidants that can delay the oxidative deterioration of CPSU and whose effectiveness is comparable to the synthetic antioxidant BHT.

AB - The synthesis of novel triphenyltin(IV) compounds, Ph3SnLn (n = 1–3), with oxaprozin (3-(4,5-diphenyloxazol-2-yl)propanoic acid), HL1, and the new propanoic acid derivatives 3-(4,5bis(4-methoxylphenyl)oxazol-2-yl)propanoic acid, HL2, and 3-(2,5-dioxo-4,4-diphenylimidazolidin1-yl)propanoic acid, HL3, has been performed. The ligands represent commercial drugs or their derivatives and the tin complexes have been characterized by standard analytical methods. The in vitro antiproliferative activity of both ligands and organotin(IV) compounds has been evaluated on the following tumour cell lines: human prostate cancer (PC-3), human colorectal adenocarcinoma (HT29), breast cancer (MCF-7), and hepatocellular cancer (HepG2), as well as on normal mouse embryonic fibroblast cells (NIH3T3) with the aid of MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-12 diphenyltetrazolium bromide) and CV (crystal violet) assays. Contrary to the inactive ligand precursors, all organotin(IV) carboxylates showed very good activity with IC50 values ranging from 0.100 to 0.758 µM. According to the CV assay (IC50 = 0.218 ± 0.025 µM), complex Ph3SnL1 demonstrated the highest cytotoxicity against the caspase 3 deficient MCF-7 cell line. Inductively coupled plasma mass spectrometry (ICP-MS) analysis indicated a two-fold lower concentration of tin in MCF-7 cells in comparison to platinum. To investigate the mechanism of action of the compound Ph3SnL1 on MCF-7 cells, morphological, autophagy and cell cycle analysis, as well as the activation of caspase and ROS/RNS and NO production, has been performed. Results suggest that Ph3SnL1 induces caspase-independent apoptosis in MCF-7 cells.

AB - Pollen (floral or bee-collected) can be used as a good source of different nutrients including several important biogenic elements (minerals). The aim of this study was to determine content of selected biogenic elements (phosphorous, potassium, sulphur, calcium, magnesium, iron and zinc) in monofloral bee-collected pollen originated from sunflower (Helianthus annuus L.) plants grown in Serbia and to estimate its nutritional value regarding to elements content. In that purpose ICP-OES analytical method was applied. According to obtained results the examined pollen sample contains above mentioned elements in following quantities: P – 3523.98 mg/kg; K – 2869.13 mg/kg; S – 1951.74 mg/kg; Ca – 1919.20 mg/kg; Mg – 654.36 mg/kg; Fe –83.55 mg/kg; Zn – 45.92 mg/kg of dry weight. Based on recommended daily intakes for iron, phosphorus and zinc application of pollen, as food or food ingredient, will cover meaningful part of human needs for these three important elements.

AB - Two novel triphenyltin(IV) compounds, [Ph(3)SnL1] (L1 = 2-(5-(4- fluorobenzylidene)-2,4-dioxotetrahydrothiazole-3-yl)propanoate (1)) and [Ph(3)SnL2] (L2 = 2-(5-(5-methyl-2-furfurylidene)-2,4-dioxotetrahydrothiazole-3-yl)propanoate (2)) were synthesized and characterized by FT-IR, (H-1 and C-13) NMR spectroscopy, mass spectrometry, and elemental microanalysis. The in vitro anticancer activity of the synthesized organotin(IV) compounds was determined against four tumor cell lines: PC-3 (prostate), HT-29 (colon), MCF-7 (breast), and HepG2 (hepatic) using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-12 diphenyltetrazolium bromide) and CV (crystal violet) assays. The IC50 values are found to be in the range from 0.11 to 0.50 mu M. Compound 1 exhibits the highest activity toward PC-3 cells (IC50 = 0.115 +/- 0.009 mu M; CV assay). The tin and platinum uptake in PC-3 cells showed a threefold lower uptake of tin in comparison to platinum (as cisplatin). Together with its higher activity this indicates a much higher cell inhibition potential of the tin compounds (calculated to ca. 50 to 100 times). Morphological analysis suggested that the compounds induce apoptosis in PC-3 cells, and flow cytometry analysis revealed that 1 and 2 induce autophagy as well as NO (nitric oxide) production.

AB - A novel triphenyltin(IV) compound with 1-(4-carboxyphenyl)-3-ethyl-3-methylpyrrolidine-2,5-dione was synthesized and characterized by IR, NMR spectroscopy, mass spectrometry, and elemental analysis. In vitro anticancer activity of ligand precursor and synthesized organotin(IV) compound was determined against tumor cell lines: human adenocarcinoma (HeLa), human myelogenous leukemia (K562), and human breast cancer (MDA-MB-453), using microculture tetrazolium test (MTT) assay. The results indicate that complex exhibited very high antiproliferative activity against all tested cell lines with IC50 values in the range of 0.22 to 0.53 mu M. The highest activity organotin(IV) compound expressed against the HeLa cells (IC50=0.22 +/- 0.04 mu M). The ligand precursor did not show anticancer activity (IC50>200 mu M). Furthermore, fluorescence microscopy analysis of HeLa cells reveal that organotin(IV) complex induced apoptosis as a mode of cell death, which is consistent with the increase of cells in the sub-G1 phase.

AB - The goal of this study is to evaluate the quality of seven natural mineral water sources in the Vrnjacka Banja area used in the treatment of various diseases. The 24 macro- and microelements were determined by inductively coupled plasma optical emission spectrometry (ICP-OES). The results show that element content was within allowed limits of concentration, except for B, As, and Se. The evaluated content of these elements can be associated with geological and anthropogenic factors. The sample S6 contains more than twice the concentration of As, meaning that the use of water from this source should be limited due to the negative impact of this element on human health. The calculated values of weekly (EWI) and oral (OI) intakes as well as the acute hazard quotient (HQ(A)) of selected toxic and potentially toxic elements indicate no health risks to the consumers. The highest hazard quotient (HQ(L)) observed for As in sample S6 (2.0 center dot 10(-1)) demonstrated a significant risk of carcinogenic diseases in long-term consumption from this source. The estimated non-carcinogenic (DIA) and carcinogenic (DIL) dermal intakes with acute (HQ(A)) and long-term (HQ(L)) hazard quotient values for sample S3 show that there is no health risk to those who use the water from this thermal source for bathing. The presence of aerobic mesophilic bacteria at 22 degrees C and 37 degrees C in samples S3, S6, and S7 as well as sulphite-reducing Clostridia species in sample S5 were detected, potentially indicating some non-faecal or faecal contamination.

AB - The biosynthetic pathway of biogenic amines is very active during the growth of many tumour cells. Non-Hodgkin lymphoma represents a very heterogeneous group of malignancies. The concentration level of some biogenic amines (putrescine, histamine, spermidine, epinephrine, and spermine) was investigated to elucidate whether they could be observed as markers for the patients with non-Hodgkin's lymphoma. In this study, the method of acidic extraction of five biogenic amines from human serum, dansyl chloride pre-column derivatization, and LC/DAD analysis were used to determine the content of biogenic amines in biological fluids. The results indicate that statistically significant differences exist in putrescine, spermidine and histamine contents in non-Hodgkin's patients versus healthy controls. The concentrations of putrescine, spermidine and epinephrine were elevated, and histamine was lower compared to controls. Based on their content, serum biogenic amines could be considered as potential tumour markers.

abstract = "The biosynthetic pathway of biogenic amines is very active during the growth of many tumour cells. Based on their content, serum biogenic amines could be considered as potential tumour markers.",

AB - Water is one of the most precious natural resources. The downtrend in the quality of lake water and pollution with heavy metals cause serious threats to the environment in last decades. Particularly vulnerable are the lakes that are located near towns and villages. The aim of this study was to investigate the physicochemical parameters and levels of macro– and microelements in order to determine the water quality of Lake Gruža in the summer period. The results showed that the studied parameters were within the permissible levels, except of concentrations of iron, cobalt and vanadium.

AB - Aims: Platinum(II) and platinum(IV) complexes [PtCln{(S,S)-(i-Am)(2)eddip}] (n = 2, 4: 1, 2, respectively; (S,S)-(i-Am)(2)eddip = O,O'-diisoamyl-(S,S)-ethylenediamine-N,N'-di-2-propanoate) were synthesized and characterized by elemental analysis, IR, H-1 and C-13 NMR spectroscopy and mass spectrometry. Method: Quantum chemical calculations were used to predict formed isomers of 1 and 2. Furthermore, reduction of 2 with ascorbic acid was followed by time-dependant C-13 NMR spectroscopy in order to enable assignation of the formed isomers for complex 1. In vitro cytotoxic activity was determined for 1 and 2 on a panel of five human tumor cell lines derived from cervix adenocarcinoma (HeLa), alveolar basal adenocarcinoma (A549), breast adenocarcinoma (MDA-453), colorectal cancer (LS 174), erythromyeloblastoid leukemia (K562), as well as one non-malignant human lung fibroblast cell line (MRC-5), using MTT assay. Result: Both complexes exhibited high (2 against K562: IC50 = 5.4 mu M), more active than cisplatin, to moderate activity (1). Both complexes caused considerable decrease of cell number in K562 cells in G1, S and G2 phases, concordantly increasing subpopulation in sub-G1 fraction. Morphological analysis of K562 cell death induced by platinum(II/IV) complexes indicate apoptosis.

AB - Six gold(III) complexes with esters of cyclohexyl-functionalized ethylenediamine-N,N'-diacetate, general formula [AuCl2{(S,S)-R2eddch}]PF6, [(S,S)-eddch = (S,S)-ethylenediamine- N,N'-di-2-(3-cyclohexyl)propanoate, R = Me, Et, n-Pr, n-Bu, i-Bu, i-Am, 1-6, respectively], were tested against cancer cell lines such as human melanoma Fem-x, human colon carcinoma LS174T and non-small cell lung carcinoma A549 as well as a non-cancerous human embryonic lung fi broblasts MRC-5 using 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide (MTT) assay with the aim of assessing in vitro antitumoral activity and selectivity. All investigated complexes showed lower cytotoxicity and better or similar selectivity in comparison to cisplatin, used as reference compound. Complex [AuCl2{(S,S)-(i-Am)2eddch}]PF6 (6) demonstrated the highest activity against Fem-x (IC50 = 14.98 ± 0.34 μM). Additionally, the same complex expressed 4.5 times higher selectivity than cisplatin.

AB - Four novel gold(III) complexes of general formulae [AuCl2{(S,S)-R(2)eddl}]PF6 (R(2)eddl = O,O'-dialkyl-(S,S)-ethylenediamine-N,N'-di-2-(4-methyl)pentanoate, R= n-Pr, n-Bu, n-Pe, i-Bu; 1-4, respectively), were synthesized and characterized by elemental analysis, UV/Vis, IR, and NMR spectroscopy, as well as high resolution mass spectrometry. Density functional theory calculations pointed out that (R,R)-N,N'-configuration diastereoisomers were energetically the most favorable. Duo to high cytotoxic activity complex 3 was chosen for stability study in DMSO, no decomposition occurs within 24 h, and for the reaction with ascorbic acid in which was reduced immediately. Additionally, 3 interacts with bovine serum albumin (BSA) as proven by UV/Vis spectroscopy. In vitro antitumor activity was determined against human cervix adenocarcinoma (HeLa), human myelogenous leukemia (K562), and human melanoma (Fem-x) cancer cell lines, as well as against non-cancerous human embryonic lung fibroblast cells MRC-5. The highest activity was observed against K562 cells (IC50: 5.04-6.51 mu M). Selectivity indices showed that these complexes are less toxic than cisplatin. 3 had a similar viability kinetics on HeLa cells as cisplatin. Drug accumulation studies in HeLa cells showed that the total gold uptake increased much faster than that of cisplatin pointing out that 3 more efficiently enters the cells than cisplatin. Furthermore, morphological and cell cycle analysis reveal that gold(III) complexes induced apoptosis in time- and dose-dependent manner.
