Neopestalotiopsis clavispora: a causal agent of twig dieback on highbush blueberries in Serbia

AB  - Highbush blueberry (Vaccinium corymbosum L.) is becoming an important berry fruit in Serbian agriculture, and the areas under blueberries are rapidly expanding. Since 2011, blueberry bushes with the symptoms of twig dieback and shoot canker were noticed in four important producing regions in the country. To identify the causal agent, 40 symptomatic bushes from four districts were sampled and analyzed. Based on morphological characteristics, all analyzed isolates were preliminary identified as Neopestalotiopsis clavispora. Selected isolates were further characterized by sequence analysis of the rDNA internal transcribed spacer region, translation elongation factor 1-alpha, and β-tubulin (TUB). Sequence and phylogenetic analysis confirmed that all isolates belong to N. clavispora. The pathogenicity tests on blueberry ‘Elliot’ confirmed that tested isolates were pathogenic. The detection of N. clavispora, a causal agent of twig dieback on blueberries in Serbia, is the first report of this pathogen in the country.

Morphological and molecular identification of Eutypa lata on grapevine in Serbia

AB  - During a survey from 2004 to 2016, symptoms of grapevine dieback were observed in six vineyards in Serbia. Symptoms initially appeared as small, chlorotic, and necrotic spots along the rim of the leaves, deformation of leafs, and the appearance of shortened shoots, often with the so-called zigzag internodes. Over time, partial or complete dying of the vines developed. Symptomatic samples were collected and submitted to laboratory analysis. Based on the morphological characterization, the isolated fungus was initially identified as belonging to Eutypa species. Pathogenicity tests showed that 47 selected isolates caused tissue necrosis around the site of inoculation, chlorosis, and deformation of the leaves, along with the appearance of tiny, necrotic spots on the periphery of the leaf that fall off over time. Inoculated cuttings were stunted, dwarf-like with zigzag internodes. Molecular identification was done with sequence and phylogenetic analysis of ITS, TUB, and RPB2 genomic regions. Based on the phylogenetic analysis, all isolated fungi were determined as Eutypa lata.

Flight dynamics and species composition of aphids landing on plum and apricot leaves in the orchards in Western Serbia

AB  - More than 40 different aphid species (Hemiptera: Aphididae) landing on plum and apricot trees were captured with Sticky shoot method and identified in the experimental orchards in Western Serbia during 2009-2011. The most abundant species found in 2009 and 2011 belongs to the genus Aphis. In 2010, Myzocallis spp. were most abundant. Specimens of the genus Muscaphis were found for the first time in Serbia. The aphid population dynamics varied greatly during the growing season. Spring peaks of aphid populations were evidenced in all 3 years in the period of May-June. Autumn peaks were recorded during the period of August-September in 2009 and 2011, but not in 2010. Twelve out of more than twenty species reported in the literature as a potential Plum pox virus (PPV) vectors were found in the orchards. Rhopalosiphum padi (Linnaeus), Aphis fabae Scopoli, Aphis craccivora Koch and invasive aphid species Aphis spiraecola Patch were the most common potential PPV vectors found. Other known PPV vectors were present in much lower specimen numbers in the following decreasing order: Hyalopterus pruni (Geoffroy), Phorodon humuli (Schrank), Brachycaudus helichrysi (Kaltenbach), Myzus persicae (Sulzer), Brachycaudus cardui (Linnaeus), Aphis gossypi Glover, Macrosiphum rosae (Linnaeus) and Metopolophium dirhodum (Walker). During all 3 years, statistically significantly greater number of known PPV vectors were captured on the plum trees during the spring peaks compared to the autumn peaks. The number of known PPV vectors captured during the spring peaks on the plum trees was also significantly larger than the number of known vectors captured on the apricot trees.

Current research in the field of fruit protection

AB  - The study results of new economically important causal agents of diseases and fruit pests were presented. Over 20 types of Erwinia amylovora have been isolated and identified on pome fruits. The most frequent peach, apricot, and plum diseases are the following: brown fruit rot, drying of small branches and canker. The importance of virus free planting material and resistant cultivars was emphasized. Using the nested PCR and PCR-RFLP methods showed the presence of Phytophthora fragarie var. rubi on three raspberry cultivars. Cecidial raspberry drying is one of the most economically harmful disease. Bacterial raspberry blight was described for the first time in Serbia. Acarology studies are directed towards the following: mechanisms of plant resistance, resistance of mites towards acaricides, efficacy of biopesticides and development of molecular methods for the faster identification of species. The increase in the population of Lymatria dispar, Cacopsylla pruni and Carcopidae was assessed in orchards.

The production of certified nursery stock material of fruit crops and grapevines

AB  - Although an initial step, the production of reproductive material and nursery stock of fruit crops and grapevines ranks first in terms of successful and profitable production. Nursery stock which is not true-to-type and infected with viruses, viroids and phytoplasms may endanger the following steps within production, minimazing huge financial investments in the planting establishment and its maintenance. Over a long-term in most developed countries, the programmes and schemes related to production of the certified reproductive material and nursery stock have been under way. The production of quality virus-free or viruses tested reproductive material and nursery stock has been in accord with strictly defined stages and work procedures for each classified category. The paper deals with the categorization of reproductive material and nursery stock, as well as with the procedures for obtaining of the specific categories and scheme for fruit crops and grapevines certification. The particular sress was laid on the major viruses and other infective pathogenes on fruit crops and grapevines, their detection methods, coupled with the necessity for the haromonization of the existing regulation in our country with the law regulation of the European Union with the aim to obtain quality nursery stock and create the possibility to take a share within the unique market of the region and Europe.
AB  - Proizvodnja reprodukcionog i sadnog materijala voćaka i vinove loze, mada početni, predstavlja jedan od najvažnijih koraka u uspešnoj i rentabilnoj proizvodnji. Sortno neispravan i virusima, viroidima ili fitoplazmama zaražen sadni materijal maze ugroziti naredne korake proizvodnje i učiniti bezvrednim velika materijalna ulaganja u podizanje i održavanje zasada. U većini razvijenih zemalja već duži niz godina funkcionišu programi i seme proizvodnje certificiranog reprodukcionog i sadnog materijala. Proizvodnja kvalitetnog bezvirusnog ill na viruse testiranog reprodukcionog i sadnog materijala se obavlja po strogo definisanim fazama i procedurama rada za svaku propisanu kategoriju. U ovom radu data je kategorizacija reprodukcionog i sadnog materijala, kao i načini dobijanja pojedinih kategorija i šema certificiranja voćaka i vinove loze. Istaknuti su najvažniji virusi na voćkama i vinovoj lozi, načini detekcije ovih virusa, kao i neophodnost harmonizacije postojeće zakonske regulative u našoj zemlji sa zakonskom regulativom Evropske Unije, u cilju obezbeđivanja neophodnog kvaliteta sadnog materijala i otvaranja mogućnosti za uključenje u jedinstveno tržište regiona i Evrope.

abstract = "Although an initial step, the production of reproductive material and nursery stock of fruit crops and grapevines ranks first in terms of successful and profitable production. Nursery stock which is not true-to-type and infected with viruses, viroids and phytoplasms may endanger the following steps within production, minimazing huge financial investments in the planting establishment and its maintenance. Over a long-term in most developed countries, the programmes and schemes related to production of the certified reproductive material and nursery stock have been under way. The production of quality virus-free or viruses tested reproductive material and nursery stock has been in accord with strictly defined stages and work procedures for each classified category. The paper deals with the categorization of reproductive material and nursery stock, as well as with the procedures for obtaining of the specific categories and scheme for fruit crops and grapevines certification. The particular sress was laid on the major viruses and other infective pathogenes on fruit crops and grapevines, their detection methods, coupled with the necessity for the haromonization of the existing regulation in our country with the law regulation of the European Union with the aim to obtain quality nursery stock and create the possibility to take a share within the unique market of the region and Europe., Proizvodnja reprodukcionog i sadnog materijala voćaka i vinove loze, mada početni, predstavlja jedan od najvažnijih koraka u uspešnoj i rentabilnoj proizvodnji. Sortno neispravan i virusima, viroidima ili fitoplazmama zaražen sadni materijal maze ugroziti naredne korake proizvodnje i učiniti bezvrednim velika materijalna ulaganja u podizanje i održavanje zasada. U većini razvijenih zemalja već duži niz godina funkcionišu programi i seme proizvodnje certificiranog reprodukcionog i sadnog materijala. Proizvodnja kvalitetnog bezvirusnog ill na viruse testiranog reprodukcionog i sadnog materijala se obavlja po strogo definisanim fazama i procedurama rada za svaku propisanu kategoriju. U ovom radu data je kategorizacija reprodukcionog i sadnog materijala, kao i načini dobijanja pojedinih kategorija i šema certificiranja voćaka i vinove loze. Istaknuti su najvažniji virusi na voćkama i vinovoj lozi, načini detekcije ovih virusa, kao i neophodnost harmonizacije postojeće zakonske regulative u našoj zemlji sa zakonskom regulativom Evropske Unije, u cilju obezbeđivanja neophodnog kvaliteta sadnog materijala i otvaranja mogućnosti za uključenje u jedinstveno tržište regiona i Evrope.",
