Sorbus sp - New host of Erwinia amylovora in Serbia

AB  - In 1989 Erwinia amylovora was confirmed to be a pathogen of pear and quince in Serbia. Subsequently, apple, firethorn, hawthorn, Mespilus spp., Cotoneaster horisontalis and Cheanotneles spp. were confirmed as hosts of the pathogen. During the summer 2005, fire blight symptoms were observed on Sorbus spp. for the first time in the south-eastern Serbia, near Nis. Disease symptoms included leaf and shoot blight and cankers with purple-brown colouration of necrotic tissue on mature branches. The diseased tissue was macerated and the suspension was streaked on nutrient sucrose agar (NAS) and King's medium B. Typical light grey, levan-positive colonies developed on NAS medium after two day incubation at 25 degrees C. Colonies on King's medium B were white and nonfluorescent. The results of physiological and biochemical tests of the bacterium were as follows: gram negative, oxidative and fermentative metabolism of glucose; oxidase negative, gelatin hydrolysis positive, aesculin hydrolysis negative and acid produced from most carbon sources. In pathogenicity tests, all isolates induced HR in tobacco leaves and necrosis on artificially inoculated pear fruits followed by appearence of bacterial ooze. After inoculation of petioles and shoots of the host plant (Sorbus spp.) initial symptoms were dark green, watersoaked, eliptical spots on inoculated tissues. Leaf and shoot blight resembling the natural infection appeared five to six days after inoculation. The investigated strains reacted positively with antisera specific to E. amylovora in DASI ELISA test. Identity of isolated strains also was confirmed by polymerase chain reaction (PCR). According to the biochemical and physiological characteristics, pathogenicity tests, results of PCR analyses and ELISA test, strains isolated from Sorbus spp. plants were identified as E. amylovora. This is the first report of E. amylovora on Sorbus spp. plants in Serbia.

Efficacy of strobilurin-related and multi-site fungicide mixtures against apple scab

AB  - The efficacy of several fungicide mixtures in controlling Venturia inaequalis in apple was evaluated in field trials. The efficacies of Flint Plus (trifloxystrobin + captan) and Tercel (pyraclostrobin + dithianon) in comparison with standard fungicides Zato 50-WG (trifloxystrobin) and Stroby + Delan (kresoxim-methyl + dithianon) were tested in the localities Mihajlovac, Radmilovac and Landol in 2004 and 2005. Both tested fungicides exhibited high efficacy in controlling apple scab. There were significant differences in the efficacies of Flint Plus (91.3-98.5%) and Zato 50-WG (68.2% and 78.4%); and Tercel (88.7-93.5%) and Stroby + Delan (77.9% and 82.1%). Our experiments showed that the investigated fungicide mixtures are highly effective against V. inaqeulais, even under high disease pressure.
AB  - Ispitivana je efikasnost nekoliko kombinacija fungicida u suzbijanju Venturia inaequalis na jabuci u poljskim uslovima. U 2005-oj i 2006-oj godini ispitivana je efikasnost Flint Plus (trifloksistrobin + kaptan) i Tercel (piraklostrobin + ditianon) u odnosu na standardne fungicide Zato 50-WG (trifloksistrobin) i Stroby + Delan (kresoksim-methil + ditianon) na lokalitetima Mihajlovac, Radmilovac i Landol. Oba ispitivana fungicida ispoljila su visoku efikasnost u suzbijanju čađave pagavosti jabuke. Između efikasnosti Flint Plus (91.3-98.5%) i Zato 50-WG (68.2% i 78.4%), i Tercel (88.7-93.5%) i kombinacije fungicida Stroby + Delan (77.9% i 82.1%) zabeležena je statistički značajna razlika. Naši ogledi su pokazali da su ispitivane kombinacije fungicida visoko efikasne protiv V. inaequalis čak i u uslovima jake zaraze.

In vitro effect of plant essential oils on growth of some soil-borne pathogens

AB  - A study of antifungal and antibacterial activity of 17 commercially available essential oils was carried out in vitro using cultures of Pythium sp., Fusarium oxysporum f, sp. lycopersici, F. oxysporum f. sp. pisi, Verticillium albo-atrum, Rhizoctonia sp. and Clavibacter michiganensis subsp. michiganensis as test organisms. The cultures, grown on artificial media in petri dishes, were exposed to volatile phase of the oils for seven days. The growth rate of the pathogens was partially or completely inhibited by all the essential oils at concentrations up to 0.65 mu L/mL of air. Cinnamon, thyme, basil, and fenchel essential oils showed fungicidal and bactericidal effect on all the tested pathogens at the concentration of 0.16 mu L/mL of air. However, turpentine, orange, and bergamot orange oils, at a concentration 0.65 mu L/mL of air or lower, were not lethal for any of the pathogens. V. albo-atrum was the most sensitive pathogen in this study, while both formae speciales of F. oxysporum showed moderate level of tolerance to the majority of the investigated oils. The results indicated that some of the tested essential oils could be used for the control of investigated soil-borne pathogens.

Current research in the field of fruit protection

AB  - The study results of new economically important causal agents of diseases and fruit pests were presented. Over 20 types of Erwinia amylovora have been isolated and identified on pome fruits. The most frequent peach, apricot, and plum diseases are the following: brown fruit rot, drying of small branches and canker. The importance of virus free planting material and resistant cultivars was emphasized. Using the nested PCR and PCR-RFLP methods showed the presence of Phytophthora fragarie var. rubi on three raspberry cultivars. Cecidial raspberry drying is one of the most economically harmful disease. Bacterial raspberry blight was described for the first time in Serbia. Acarology studies are directed towards the following: mechanisms of plant resistance, resistance of mites towards acaricides, efficacy of biopesticides and development of molecular methods for the faster identification of species. The increase in the population of Lymatria dispar, Cacopsylla pruni and Carcopidae was assessed in orchards.
AB  - Prikazani su rezultati istraživanja novih bolesti i štetočina voćaka sa posebnim osvrtom na: molekularnu detekciju patogena, Pseudomonas syringae na koštičavom voću, sojeve Erwinia amylovora na jabučastim voćkama, prouzrokovače mrke truleži (Monilinia spp.) i rak rana (Leucostoma spp.) na koštičavim vrstama voćaka, molekularne metode za utvrđivanje prisustva Phytophthora fragariae var. rubi na malini, cecidoizno sušenje maline, prouzrokovača bakteriozne plamenjače maline (Pseudomonas syringae pv. syringae), akarološka istraživanja, kao i na porast brojnosti Lymantria dispar, Cacopsylla pruni i cikada penuša (Cercopidae), zaštitu voćaka u organskoj proizvodnji.

abstract = "The study results of new economically important causal agents of diseases and fruit pests were presented. Over 20 types of Erwinia amylovora have been isolated and identified on pome fruits. The most frequent peach, apricot, and plum diseases are the following: brown fruit rot, drying of small branches and canker. The importance of virus free planting material and resistant cultivars was emphasized. Using the nested PCR and PCR-RFLP methods showed the presence of Phytophthora fragarie var. rubi on three raspberry cultivars. Cecidial raspberry drying is one of the most economically harmful disease. Bacterial raspberry blight was described for the first time in Serbia. Acarology studies are directed towards the following: mechanisms of plant resistance, resistance of mites towards acaricides, efficacy of biopesticides and development of molecular methods for the faster identification of species. AB  - During the past few years, a frequent appearance of bacterial speck of tomatoes was observed in all tomato growing areas in Serbia. Phytobacteriological analyzes of the diseased plant samples, collected from different localities, showed that symptoms were caused by phytopathogenic bacteria. In order to identify isolated bacteria, their pathogenic morfological, cultural, biochemical and physiological characteristics where studied. Based on the results of these studies, it was determined that investigated strains belonged to P. s. pv. tomato. Coronatine producing strains of P. s. pv. tomato were detected by polymerase chain reaction. Reaction of tomato differential cultivar Ontario 7710 indicated that out of 37 strains 21 belonged to races 0 and 16 to race 1 of P.s. pv. tomato.
AB  - Iz uzoraka obolelih biljaka paradajza sa simptomima bakteriozne pegavosti lišća i krastavosti plodova, prikupljenih sa nekoliko lokaliteta u Srbiji tokom poslednjih godina, izolovani su sojevi bakterija. U cilju identifikacije, proučene su njihove patogene, morfološke, odgajivačke i biohemijsko-fiziološke karakteristike. Na osnovu rezultata istraživanja utvrđeno je da svi proučavani sojevi pripadaju bakteriji Pseudomonas syringae pv. tomato (Okabe) Yang, Dye et Wilkie 1978. Primenom lančane reakcije polimeraze (PCR) u genomu proučavanih sojeva potvrđeno je prisustvo gena koji determiniše stvaranje fitotoksina koronatina, što se smatra diferencijalnom karakteristikom P.s. pv. tomato. Razlike ispoljene u reakciji inokulisanih biljaka diferencijalne sorte paradajza Ontario 7710, ukazuju da naši sojevi pripadaju rasama 0 i 1 bakterije Pseudomonas syringae pv. tomato.

AB  - Field trials were carried out to evaluate the efficacy of a new fungicide mixture fenamidone + propamocarb-hydrochioride in the control of potato late blight and tomato early blight. The experiments were contucted in three localities: Majur (Šabac), Dobanovci (Beograd) i Nova Pazova, according to EPPO/OEPP standard methods. Two doses (2.00 and 1.67 L/ha) of trade formulation Consente (propamocarb-hydrochloride 375 g/L + fenamidone 75 g/L) were used. According to the results Consente (2.00 L/ha) showed highest efficacy (90.11 - 97.23%) in the control of potato late blight and tomato early blight. Referent formulation Tataoo (propamocarb-hydrochloride 248 g/L + mancozeb 306.1 g/L) exhibited statistically lower efficacy (83.68 - 91.60%) than Consente.
AB  - Ispitivana je efikasnost nove kombinacije fungicida fenamidon propamokarb-hidrohlorid, u cilju suzbijanja prouzrokovača plamenjače i crne pegavosti krompira i paradajza. Ispitivanja su izvedena u usevima krompira i paradajza na lokalitetima Majur (Šabac), Dobanovci (Beograd) i Nova Pazova, u skladu sa standardnim metodama OEPP/EPPO. Primenjen je preparat Consento (propamokarb-hidrohlorid 375 g/L + fenamidon 75 g/L) u dve količine:2.00i 1.67 L/ha. Preparat Consento je pokazao visoku efikasnost (90.1 1 -97.23%) u suzbijanju Phytophthora infestans i Alternaria solani pri količini primene od 2.0 L/ha, a nešto slabiju (84.85 - 92.44%) pri količini od 1.67 L/ha. Referentni preparat Tatoo (propamokarb-hidrohlorid 248 g/L + mankozeb 306.1 g/L) primenjen u količini od 4.00 L/ha ispoljio je, na većini lokaliteta statistički značajno slabiju efikasnost (83.68 - 91.60%) u odnosu na preparat Consento.

In vitro effects of essential oils from aromatic and medicinal plants on soil borne pathogens

AB  - Mint(Menta piperita L), basil (Ocimum basilicum L), rosemary (Rusmarinus officinalis L), thyme (Thymus vulgaris L) and tea tree (Melaleuca altemifolia (Maid, and Bet) Cheel) essential oils were tested for their effectivness against soil borne pathogens including; Pythium sp., Rhizoctonia sp., Fusarium oxysporum f. sp. lycopersici, F. oxysporum f. sp. pisi Verticillium sp. and Clavibactermichiganensis ssp. Michiganensis. The isolates were exposed to volatile phase of the oils for seven days. Afterwards, the plates were opened and ventilated in order to remove volatiles and determine if the effect of the oil was fungistatic/bacteriostatic or fungicidal/bactericidal. The concentrations of the oil were considered to be fungicidal/ bactericidal if the microbial growth had not been observed seven days after the ventilation. The growth rate of all the tested microorganisms was partially or completely inhibited by the oils applied at concentrations 0.04-0.65 μl/ml air. Thyme and basil oils were the most toxic to all the pathogens investigated, having fungicidal/bactericidal effect at 0.04-0.65 μl/ml of air, depending of the pathogen. Rosemary oil showed the lowest inhibitory effect - minimal lethal concentrations were 0.32 μl/ml air for C. m. ssp. michiganensis, 0.65 μl/ml air for Verticillium sp. Pythium sp. and Rhizoctonia sp., and more than 0.65 μl/ml air for Fusarium species, respectively. These results indicate that thyme and basil essential oils could be effectively used for the control of soil borne pathogens.
AB  - Prikazani su rezultati in vitro testiranja efekata etarskih ulja nane (Menta piperita L), bosiljka (Ocimum basilicum L), ruzmarina (Rusmarinus officinalis L), timijana (Thymus vulgaris L) i čajnog drveta (Melaleuca altemifolia (Maid, and Bet) Cheel) na biljne patogene koji se prenose zemljištem Pythium sp., Rhizoctonia sp., Fusarium oxysporum f. sp. lycopersici, F. oxysporum f. sp. pisi, Verticillium sp. i Clavibacter michiganensis ssp. michiganensis. Izolati su izlagani dejstvu para proučavanih ulja tokom sedam dana, a po završetku ekspozicije kutije su provetravane strujanjem vazduha u laminarnoj komori. Ocenjivan je efekat potpune inhibicije fungistatičnost/bakteriostatičnost (izolat posle uklanjanja etarskog ulja provetravanjem nastavlja da raste) i fungicidnost/baktericidnost (ulje deluje letalno tako da nema porasta izolata posle provetravanja). Sva proučavana etarska ulja u koncentracijama 0.04-0.65 μl/ml vazduha delimično ili potpuno inhibiraju porast proučavanih mikroorganizama. Ulja timijana i bosiljka ispoljila su najveću toksičnost za sve proučavane patogene sa vrednostima minimalne letalne koncentracije između 0.04 i 0.65 μl/ml vazduha, zavisno od patogena. Najslabiji efekat pokazalo je ulje ruzmarina-minimalna letalna koncentracija za C. m. ssp. michiganensis iznosila je 0.32 μl/ml vazduha, za Verticillium sp. Pythium sp. i Rhizoctonia sp. 0.65 μl/ml vazduha, a za vrste roda Fusarium vise od 0.65 μl/ml vazduha. Rezultati in vitro ogleda ukazuju na mogućnost korišćenja ulja timijana i bosiljka u suzbijanju patogena koji se prenose zemljištem.

Ulja timijana i bosiljka ispoljila su najveću toksičnost za sve proučavane patogene sa vrednostima minimalne letalne koncentracije između 0.04 i 0.65 μl/ml vazduha, zavisno od patogena. Najslabiji efekat pokazalo je ulje ruzmarina-minimalna letalna koncentracija za C. m. ssp. michiganensis iznosila je 0.32 μl/ml vazduha, za Verticillium sp. Pythium sp. i Rhizoctonia sp. 0.65 μl/ml vazduha, a za vrste roda Fusarium vise od 0.65 μl/ml vazduha. Rezultati in vitro ogleda ukazuju na mogućnost korišćenja ulja timijana i bosiljka u suzbijanju patogena koji se prenose zemljištem.",
