AB  - Mushrooms are experiencing a kind of renaissance as a part of the contemporary human diet. These valuable organisms are more than food, they fit in perfectly as a novel market group known as nutra-mycoceuticals. Immune-balancing mushroom dietary fibers and secondary metabolites such as polyphenols are the main focus of the healthcare industry. Wellness and cosmetic companies are increasingly using mushroom extracts rich in these ingredients. This review considers the basic molecular immunomodulatory mechanisms of action of the most commonly used mushroom dietary fibers, β-glucans. The literature data on their bioavailability, metabolic transformations, preclinical and human clinical research, and safety are discussed. Immunomodulatory mechanisms of polyphenol ingredients are also considered. These molecules present great potential in the design of the new immunity balancer formulations according to their widespread structural diversity. Finally, we draw attention to the perspectives of modern trends in mushroom nutraceutical and cosmeceutical formulations to strengthen and balance immunity. © 2022

Mushroom polyphenols as immune system balancers: What's the mechanism behind it and possible interactions with dietary fibers?

AB  - Mushroom polyphenols have been proposed as a new therapy that can improve the immune system.
Different immune cells express multiple types of polyphenol receptors that recognize and allow cellular
uptake of polyphenols, which subsequently activate signaling pathways to initiate immune responses.
Polyphenols can be useful for immunocompromised patients with malignancies, viral or bacterial
infections and chronic disorders of organ systems, as well as for those whose immunity is naturally
weakened. Cosmetic formulations rich in mushroom polyphenols have the potential to regulate inflammatory
skin disorders, as well as eczema or atopic dermatitis and photocarcinogenesis.
As primary antioxidants, polyphenols have an important role in preserving immune cells in a reduced
environment and in protecting them against oxidative damages and immunosuppression as well as
maintaining their suitable function. As signaling pathway modulators, mushroom polyphenols can
affect immune cell regulation, cytokine synthesis, and gene expression, in both innate and adaptive
immune responses. There are different pathways in the integrated immunomodulatory polyphenol response,
such as: 1) the nuclear NF-κB signaling pathway leading to suppression of various inflammatory
cytokines expression, and enzymes such as COX-2; 2) MAPK signaling pathways that play a key role in
many fundamental cellular processes such as cell growth, proliferation, death and differentiation, and
also regulate gene transcription and the activity of transcription factors involved in inflammation; 3)
arachidonic acid signaling pathway leading to a decrease in the release of inflammatory mediators;
and 4) the Nrf2 / ARE signaling pathway involved in the activation of genes encoding cytoprotective and
antioxidant enzymes.
Given that polyphenols represent great potential in the design of immune-boosting formulations in line
with their widespread structural diversity, it should be noted that some functionality issues require further
clarification. For instance different observations and conclusions were reported by the scientists in
absence of enzymes involved in their biosynthesis steps.
Concerning the dietary application of mushroom polyphenols, it is necessary to mention that after
oral consumption they are recognized by the human body as xenobiotics and often a small amount is
absorbed in the intestine. Likewise, there are significant differences between the activities of the metabolic
form of phenols and their form in the mushroom nutraceutical matrix. For an example, dihydoferulic
acid, a metabolite of the ferulic acid, expresses anti-inflammatory activity, opposite to metabolites derived
after sulfation and glucuronidation. The scientific focus is on improving and increasing polyphenols bioavailability
by designing colloidal systems and using nanosystems. Moreover many studies have found
that polyphenols can interact with macromolecules like dietary fibers, i.e. chitin and β-glucans. It has
been shown that these interactions can affect the bioaccessibility of polyphenols in a food matrix as
well as in nutraceutical formulations. Therefore, chitin and β-glucans have an impact on polyphenols’
immunomodulating activities if they were applied together in commercial formulations. Another disadvantage
in the commercialization of polyphenol-based formulations is quantitative and qualitative variations
in their content among different mushroom species. As secondary metabolites with a protective
role e.g. phenols, their content in mushrooms depends on the locality and environmental conditions of
growth. Using extracts or preparations that are based on mycelia grown under controlled conditions
may be the solution for the reduction of the natural variability in polyphenol composition.
Integrated information on all aspects of functionality will confirm the use of polyphenol-rich mushroom
formulations as effective enhancers of immunity.

The Antimicrobial Activities of Silver Nanoparticles Synthesized from Medicinal Mushrooms

AB  - Crude polysaccharide extracts derived from Agaricus bisporus (A), A. brasiliensis (B), and Phellinus linteus (P) were used for the synthesis of silver nanoparticles (AgNPs, labeled AAgNP, BAgNP, PAgNP, respectively) UV and FT-IR spectra, size distribution, and zeta potential of the respective particles were recorded, and TEM and SEM-EDS characterization was done. UV-Vis spectroscopy showed maximum absorbance at 415 mu for A, at 400 nm for P, and at 420 nm for B. SEM-EDS analysis showed BAgNP to consist of almost 90% of silver, while silver content in AAgNP and PAgNP was much lower, 46.5 and 52.3%, respectively. Individual extracts as well as the Ag-NPs were tested against a panel of clinically isolated and ATCC derived pathogens. In almost all cases minimum inhibitory concentration (MIC) was significantly lower than those of antibiotics, suggesting up to 100 times more effectivity. BAgNP, PAgNP, and AAgNP appeared very active against Pseudomonas aeruginosa ATCC 27853 (MIC: 0.19, 0.19, 0.97 mu g/mL, respectively). The same nanoparticles were also very effective toward Candida albicans ATCC 10231 (MIC: 0.39, 0.78, 0.97 mu g/mL, respectively). Our study indicates that the bactericidal effect of PAgNP, AAgNP, BAgNP on E. coli 25922 ATCC (MBC: 1.56, 0.97, 6.25 mu g/mL, respectively) is significantly more pronounced than that of amoxicillin. A pure solution of colloidal silver nanoparticles showed significantly weaker microbiostatic/microbiocidal potential than all tested mushroom extracts' AgNPs, as well as amoxicillin. Silver nanoparticles made with extracts of A, B, and P seem welcome as an addition to the inventory of antimicrobial compounds used in clinical medicine against bacterial and yeast infection.

Characterisation and antimicrobial activity of silver nanoparticles derived from Vascellum pratense polysaccharide extract and sodium citrate

AB  - Silver nanoparticles (AgNPs) were synthesized by “green”, cheap hydrothermal method in an autoclave using sodium citrate and Vascellum pratense polysaccharide extract as reducing and stabilizing agents. Presence of spherical AgNPs was confirmed by UV-VIS spectrophotometry and scanning electron microscopy; particle size was determined as ~ 40 nm. Even though colloidal solution had relatively low absolutevalue of zeta potential (-15 mV), short term stability studies suggested a stable system, with AgNPs being stabilized by both citrate and fungal polysaccharides, as FTIR spectra confirmed. The colloidal solution showed good antimicrobial activity against both G+/G- bacteria and Candida albicans, including methicilin resistant Staphylococcus aureus (MRSA). Products containing AgNPs and fungal polysaccharides, which possess various biological activities - most important being immunostimulation - may find use in treatment of skin conditions caused by pathogens.

AB  - Kaolinite and the modified natural zeolite minazel plus (M+) were used as supplements in substrate used for the production of the medicinal mushroom Grifola frondosa. Growth stimulation, expressed as yield and biological efficiency, was observed when M+ (1%) was added. The production cycle was shortened by half as a result of the zeolites' ion-exchange ability, stimulation of enzyme activity, and water retain capacity. Inductively coupled plasma optical emission spectrometry of fruiting bodies showed the absence of heavy metals (arsenic, cadmium, and lead), whereas the concentration of calcium increased greatly and the concentrations of iron, magnesium, and zinc increased moderately under the influence of M+. Solid-state nuclear magnetic resonance showed a positive impact on the beta-glucanratio, which could have been caused by the epimerization reaction stimulated by zeolites. The functionality of the mushroom was evaluated through several antioxidant activity assays, and in all cases a positive effect was established: M+ was statistically more effective in comparison with kaolinite. A strong correlation was established between the antioxidative activity of cultivated fruiting bodies and the tested compounds (total phenolic compounds, carbohydrates, and minerals).

AB  - The birch polypore Piptoporus betulinus was among two mushrooms that were found in the Iceman's bag. Recent studies indicated that P. betulinus was probably used as a religious and medicinal item. In order to examine the medicinal potential of P. betulinus, hot water (HW), partially purified (PP), and alkali extract (HA) were prepared and tested for antioxidant, antimicrobial, cytotoxic, and angiotensin I-converting enzyme (ACE) inhibitory activity. All tested samples exhibited moderate cytotoxic activity, and HW appeared as the most effective (IC50 = 0.8 +/- 0.1 mg/ml for HeLa cells). HA proved to be a good 1,1-diphenyl-2-picrylhydrazyl radical scavenger and exhibited the strongest ferric-reducing power (EC50 = 0.07 +/- 0.3 mg/ml). The same extract (HA) also expressed the strongest ferric-reducing power (EC50 = 0.99 +/- 0.1 mg/ml). Hot alkali extraction contributed significantly to ACE inhibitory activity (EC50 = 0.06 +/- 0.00 mg/ml) and to antimicrobial activity, especially against highly resistant Enterococcus faecalis (minimum inhibitory concentration: 0.156 +/- 0.000 mg/ml; and minimum bactericidal concentration: 1.25 +/- 0.00 mg/ml).

AB  - Oxidative stress caused by an imbalanced metabolism and an excess of reactive oxygen species (ROS) lead to a range of health disorders in humans. Our endogenous antioxidant defense mechanisms and our dietary intake of antioxidants potentially regulate our oxidative homeostasis. Numerous synthetic antioxidants can effectively improve defense mechanisms, but because of their adverse toxic effects under certain conditions, preference is given to natural compounds. Consequently, the requirements for natural, alternative sources of antioxidant foods identified in edible mushrooms, as well as the mechanistic action involved in their antioxidant properties, have increased rapidly. Chemical composition and antioxidant potential of mushrooms have been intensively studied. Edible mushrooms might be used directly in enhancement of antioxidant defenses through dietary supplementation to reduce the level of oxidative stress. Wild or cultivated, they have been related to significant antioxidant properties due to their bioactive compounds, such as polyphenols, polysaccharides, vitamins, carotenoids and minerals. Antioxidant and health benefits, observed in edible mushrooms, seem an additional reason for their traditional use as a popular delicacy food. This review discusses the consumption of edible mushrooms as a powerful instrument in maintaining health, longevity and life quality.

The structural properties of FP and FNa samples were investigated by FT-IR and high resolution 1H- and 13C-NMR spectroscopy. From a group of various G- and G+ bacteria the antibacterial effects were highest against the G + B. cereus. FNa was the better antioxidant shown by the lower EC50 values of DPPH scavenging ability, ferric-reducing antioxidant power and ferrous ion-chelating ability. Ferric-reducing antioxidant power and ferrous ion-chelating ability were mostly linked to total polysaccharides, total- and beta-glucan content, as well as total protein content. Both extracts displayed a moderate dose dependent antiproliferative action towards malignant human breast cancer MDA-MB-453, cervical adenocarcinoma HeLa and myelogenous leukemia K562 cells not observed in the non cancer derived MRC-5 fibroblasts. The highest effect was found in HeLa cells for FP extract. The mean diameter of Ca-alginate bead loading FP was 960.7 mu m while the mean diameter of beads encapsulating FNa extract was 1051.7 mu m.

Phenols were the major antioxidant components found in the extract (49.8 mg g(-1)), followed by flavonoids, whose content was approximately 86% of the total phenol content. Antioxidant activity, measured by four different methods, was high for inhibition of lipid peroxidation (EC50 = 1.21 mg mL(-1)) and chelating ability (EC50 = 0.64 mg mL(-1)). The antioxidant activity of the C. cibarius methanol extract was achieved through chelating iron compared to hydrogen atom and/or electron transfer. The extract showed good selectivity in cytotoxicity on human cervix adenocarcinoma HeLa, breast carcinoma MDA-MB-453 and human myelogenous leukemia K562, compared to normal control human fetal lung fibroblasts MRC-5 and human lung bronchial epithelial cells BEAS-2B. The extract had inhibitory activity against angiotensin converting I enzyme (ACE) (IC50 = 0.063 mg mL(-1)). The extract revealed selective antimicrobial activity against Gram-positive bacteria with the highest potential against E. faecalis. The medicinal and health benefits, observed in wild C. cibarius mushroom, seem an additional reason for its traditional use as a popular delicacy food.

AB  - Polysaccharides of the European strain of A. brasiliensis were obtained by hot water extraction and ethanol precipitation (HWPE I) of fruiting bodies, and further purified by dialysis (HWPE II) and pronase incubation (PPE). These polysaccharides consisted mainly of (1 -> 6)-beta-D-glucans. PPE was free of proteins and polyphenols as demonstrated by quantitative assays and NMR profiling. They showed a clear IFN-gamma inducing activity in human PBMCs, which suggests these polysaccharides to have proinflammatory effects. Treatment by beta-glucosidase caused the polysaccharides to be degraded into smaller fragments and at the same time increased their IFN-gamma inducing activity in PBMCs fourfold. In vitro, PPE showed a dose-dependent inhibition of the proliferation of the human leukemia Jurkat cell. At 100 mu g/mL the cells' viability was decreased by appr. 51% compared to the control. EPR spin trapping demonstrated a high antioxidative activity against (OH)-O-center dot and (center dot)O2(-) radicals of HWPE land PPE. Further, the results of the antioxidant assays indicated that antioxidant activity against (OH)-O-center dot radicals in the Fenton system was achieved through scavenging or through chelating iron mechanisms. The good immunomodulating and antioxidative properties of A. brasiliensis polysaccharide extract obtained by hot water extraction and ethanol precipitation make it suitable for everyday use as an inexpensive dietary supplement.

AB  - The fungal polysaccharides attract a lot of attention due to their multiple challenging biological properties, such as: anti-tumor, anti-viral, anticomplementary, anticoagulant, hypolipidemic, immunomodulatory and immune-stimulatory activities, which all together make them suitable for application in many quite distinctive areas, such as food industry, biomedicine, cosmetology, agriculture, environmental protection and waste water management. This article presents results with respect to biological properties, structure and procedures related to the isolation and activation of polysaccharides of higher fungi. It is considered and presented along with a review of the critical antioxidative activity and possible influence of the structural composition of polysaccharide extracts (isolated from these higher fungi) upon their antioxidative properties.
AB  - Polisaharidi gljiva privlače veliku pažnju zbog svojih interesantnih bioloskih svojstava kao sto su: antitumorno, antivirusno, antikomplementarno, antikoagulaciono, hipolipidemijsko dejstvo, kao i imunomodulatorska i imunostimulativna aktivnost, sto ih sve čini pogodnim za primenu u mnogim oblastima, među kojima su i prehrambena industrija, kozmetika, biomedicina, poljoprivreda, zastita životne sredine i otpadnih voda. Oksidativna ostecenja prirodnih i industrijski pripremljenih namirnica predstavljaju veliki ekonomski problem na globalnom nivou, s obzirom da direktno utiču na promenu organoleptičkih osobina proizvoda, pri čemu dolazi i do stvaranja potencijalno toksičnih jedinjenja. Zbog toga se danas sve veća pažnja posvećuje istraživanjima novih prirodnih izvora antioksidanasa, među kojima su veoma značajni i polisaharidni ekstrakti različitih bazidiomiceta. U ovom radu su prikazana bioloska svojstva, struktura, kao i postupci izolovanja i aktivacije polisaharida visih gljiva. Dat je i pregled antioksidativne aktivnosti polisaharidnih ekstrakata visih gljiva i razmatran je uticaj strukturnih karakteristika ovih ekstrakata na antioksidativna svojstva.

AB  - Hot water extract (LN), partially purified polysaccharides (LP) and hot alkali extracted polysaccharides (LNa) obtained from fruiting bodies of the wild basidiomycete Laetiporus sulphureus were examined for their antioxidant activities. LNa was the most active antioxidant, as shown by the median effective concentrations (EC50 values) of 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity (0.5 +/- 0.2 mg/ml), reducing power (4.0 +/- 0.3 mg/ml) and ferrous ion-chelating ability (1.5 +/- 0.1 mg/ml). LNa contained the highest level of alpha-glucan (17.3 +/- 1.2 g/100 g dw), whereas LP contained mostly beta-glucans (66.8 +/- 1.3 g/100 g dw). The prevalent monosaccharide in all extracts was glucose. The EC50 values of all three antioxidant activity assays were well-correlated with the alpha-glucan content. Strong and significant correlation was found between total phenolic compounds and DPPH scavenging ability and also reducing power. The three investigated extracts (at concentrations of 0.1-10 mg/ml) were not toxic to HTR-8/SVneo trophoblast cell line.

AB  - Antioxidant activities of polysaccharide extracts of four of the most widely known mushrooms often used in medicinal applications as well as in tea and food, namely Ganoderma applanatum, Ganoderma lucidum, Lentinus edodes and Trametes versicolor, were studied. G. applanatum and L edodes extracts were the strongest DPPH scavengers, i.e. EC50  lt  0.1 mg/mL. G. applanatum extract had the highest chelating ability of ferrous ion (EC50 = 3.58 mg/mL), inhibition of lipid peroxidation (EC50 = 2.07 mg/mL) and had the highest reducing power, EC50 = 0.18. All four polysaccharide extracts showed a strong correlation between the reducing power and the total amount of phenols and alpha-glucans: no such correlation was found between the reducing power and the amount of total polysaccharides and proteins. The extracts contained a mixture/complex of polysaccharides, proteins and polyphenols, still present after hot water treatment, ethanol precipitation and dialysis. Glucose was the prevalent monosaccharide of all polysaccharides and samples contained both alpha- and beta-glucans. Total glucan contents varied considerably, i.e. from 35% for G. applanatum to 83% for G. lucidum. Uronic acid was found in the G. applanatum polysaccharides. Fourier transform infrared (FT-IR) analysis showed strong aromatic and ester peaks next to the different sugar and polysaccharide signals.

AB  - Antioxidant properties properties of hot water extract (HWE), hot water extracted polysaccharides (HWP) and hot alkali extracted polysaccharides (HWAE) were obtained from fruiting bodies of the wild basidiomycete Schizophyllum commune. All extracts contained both alpha- and beta-glucans as determined by Megazyme beta-glucan assay kit. Glucose was found by TLC and NMR to be the prevalent monosaccharide in all extracts. Total phenol contents were in descending order of HWP approximate to HWE > HWAE. Median effective concentrations (EC50 values) of antioxidant activities were 8.3 +/- 0.1, 6.9 +/- 0.0 and 8.9 +/- 0.1 mg/mL, and of the DPPH scavenging activity 0.8 +/- 0.0, 0.6 +/- 0.0 and 1.8 +/- 0.0 mg/mL, for HWE, HWP and HWAE, respectively. EC50 values of reducing power were 7.6 +/- 0.1, 7.9 +/- 0.0 and 12.5 +/- 0.1 mg/mL, whereas those of the chelating abilities on ferrous ions were 3.1 +/- 0.0, 4.6 +/- 0.1 and 4.9 +/- 0.1 mg/mL The EC50 values of the antioxidant activity, of the DPPH scavenging, and of the reducing power were correlated with total polysaccharide as well as with total phenol content. The antioxidant activities of all the extracts may be caused by both polysaccharides and polyphenols or by a complex of both.

Antioxidative and immunomodulating activities of polysaccharide extracts of the medicinal mushrooms Agaricus bisporus, Agaricus brasiliensis, Ganoderma lucidum and Phellinus linteus

AB  - Partially purified polysaccharides were obtained from four medicinal mushroom species, Agaricus bisporus, Agaricus brasiliensis, Phellinus linteus and Ganoderma lucidum by hot water extraction, followed by ethanol precipitation. The four samples contained varying amounts of both alpha- and beta-glucans as determined by FT-IR and by quantitative estimation after prior partial hydrolysis (Megazyme beta-glucan assay kit). EC50 values of the DPPH scavenging activity of the polysaccharides from G. lucidum spores and P. linteus fruiting bodies were found to be particularly low, i.e. EC50  lt  0.1 mg/ml. For A. brasiliensis and A. bisporus, EC50 values were 0.27 and 2.0 mg/ml. EC50 values of the antioxidant activity were 7.07 mg/ml for G. lucidum, 13.25 mg/ml for A. brasiliensis and >20 mg/ml for A. bisporus polysaccharide, respectively. EC50 values of the chelating activity of ferrous ions ranged from 0.59 mg/ml for G. lucidum to 7.80 mg/ml for A. bisporus. The EC50 values of the extracts in the reducing power assay ranged from 0.47 to 14.83 mg/ml. A correlation was found between EC50 values of the chelating and reducing power abilities and the amount of total glucans content in the extracts. In vitro measurements of immunomodulatory capacity of polysaccharide extracts showed that A. bisporus, A. brasiliensis fruiting bodies and G. lucidum spores extracts express an immunostimulating effect on activated human PBMCs and induce synthesis of IFN-gamma. The polysaccharide extract of P. linteus fruiting bodies showed an immunosuppressive effect.
