IN VITRO SENSITIVITY OF COLLETOTRICHUM ACUTATUM ISOLATES FROM STRAWBERRY TO TEBUCONAZOLE, PROCHLORAZ, FLUDIOXONIL AND THIOPHANATE-METHYL

AB  - The objective of this study was to determine the Colletotrichum acutatum isolates sensitivity to several chemical compounds in vitro and to find their possibilities as a potential control agents for anthracnose disease on strawberry. C. acutatum J.H. Simmonds, a causing agent of anthracnose, is a very important pathogen of strawberry, which leads to devastating losses in its production. The pathogen is effectively controlled by the fungicides. Thus their application is necessary for achieving high yield and fruit quality. The sensitivity of 14 isolates of C. acutatum, collected from commercial strawberry plantations in Serbia, to four fungicides, was examined by an in vitro sensitivity assay. Based on the results of morphological, pathogenic and molecular characterization (up to the complex level), all 14 isolates were determined as C. acutatum. The commercial formulation of tebuconazole, fludioxonil, prochloraz and thiophanate-methyl were used for the sensitivity test. The mycelial growth assay method was used to investigate isolates sensitivity to fungicides. The tested isolates were very sensitive to prochloraz and fludioxonil, with mean EC50 values of 0.067±0.062 mg L-1 and 0.093±0.043 mg L-1, respectively. Significantly higher mean EC50 values were observed for tebuconazole (1.473±0.878 mg L-1) and thiophanate-methyl (1.718±1.592 mg L-1). The toxicity of tested fungicides in the mycelial growth assay of C. acutatum isolates indicates the potential implementation of these fungicides in the protection programs against strawberry anthracnose disease. © Journal of Agricultural Sciences (Belgrade).All rights reserved.

Efikasnost fungicida u suzbijanju prouzrokovača plamenjače paradajza (Phytophthora infestans (Mont.) de Bary) u uslovima jake pojave oboljenja

AB  - U toku 2020. godine, vršeno je ispitivanje biološke efikasnosti fungicida propamokarb – hidrohlorid + fluopikolid, cimoksanil, mandipropamid + difenokonazol
i dimetomorf + mankozeb u suzbijanju prouzrokovača plamenjače (Phytophthora
infestans) u usevu paradajza.
 Ogledi su izvedeni na lokalitetima Sovljak (Ub), Belosavci (Topola) i Garaši
(Aranđelovac), po tipu potpuno slučajnog blok sistema u četiri ponavljanja (EPPO,
1996). Na svakom lokalitetu izvršeno je po pet tretiranja, a ocene efekata vršene su
u dva navrata. Povoljni agrometeorološki uslovi rezultirali su značajnom pojavom
oboljenja u kontroli, koja je iznosila 17,5-41,3% u prvoj, odnosno, 28,8-95,0% u
vreme druge ocene efekata.
Primenom kombinacije fungicida propamokarb-hidrohlorid+fluopikolid ostvarena je efikasnost u iznosu od 94,5-99,4% u prvoj, odnosno, 92,3-97,9% u drugoj
oceni. Cimoksanil je ispoljio efikasnost koja je iznosila 83,0-92,9% u prvoj, i 80,0-
89,5% u drugoj oceni. Kombinacijom fungicida mandipropamid+difenokonazol
postignuta je efikasnost od 89,1 do 99,7% u prvoj, odnosno 88,2-98,5% u vreme
druge ocene efekata. Sa kombinacijom fungicida dimetomorf+mankozeb ostvarena
efkasnost iznosila je 94,1-97,8% u prvoj, odnosno 90,7-97,7% u drugoj oceni.
AB  - During the season 2020, the biological efficacy of fungicides propamocarb
hydrochloride + fluopicolide, cymoxanil, mandipropamide + difenoconazole and
dimothomorph + mancozeb in the control of late blight (Phytophthora infestans)
in tomato, was investigated. The experiments were conducted in the localities of
Sovljak (Ub), Belosavci (Topola) and Garaši (Aranđelovac). The experiments were
carried out as a randomized block system in four replication (EPPO, 1996). A total
of five treatments and two assessments were performed at each location. Favorable
conditions resulted in a significant disease intensity on untreated plots, which amounted from 17,5-41,3% in the first, and 28,8-95,0% during the second assessment.
Fungicide mixture propamocarb-hydrochloride+fluopicolide, showed the efficacy from 94,5-99,4% at the first, and 92,3-97,9% at the second assessment. Fungicide cymoxanil, showed the efficacy of 83,0-92,9% at the first, and 80,0-89,5%

In vitro assessment of pesticide residues bioaccessibility in conventionally grown blueberries as affected by complex food matrix

AB  - The aim of this study was to investigate the bioaccessibility of pesticide residues in blueberries (com-mercial and sample from controlled field trial) from Serbia, involving the presence of a complex food matrix and to assess the potential risk to human health. The presence of nine active substances (azox-ystrobin, boscalid, fludioxonil, cyprodinil, pyrimethanil, pyridaben, pyriproxyfen, acetamiprid and thia-metoxam) in initial blueberry samples was determined in concentration range from 5.15 mu g/kg for thiametoxam to 187 mu g/kg for azoxystrobin. Clothianidin, metabolite of thiametoxam, was not detected in any blueberry sample. However, after in vitro digestion, the content of initially detected pesticides residues was significantly decreased or it was below limit of quantification resulting in the total bio-accessibility of about 15%. Azoxystrobin, pyrimethanil and fludioxonil was quantified in digestive juice at concentrations which were about 81%, 37% and 10% less than the inital concentration, respectively. The presence of food matrix during digestion of blueberries even more severely reduced concentration of pesticide residues (total bioaccessibility was about 7%) compared to digestion without the food matrix. Only azoxystrobin was quantified after digestion with food matrix in concentration of 27 mu g/kg in sample from controlled field trial and detected in two commercial samples but below the limit of quantification. Furthermore, chronic risk assessment indicated that risk is acceptable for the health of different human subpopulation groups. The current study on pesticides residues, most commonly applied on blueberries, provides for the first time an insight into their bioaccessibility under conditions that mimic physiological environment of human digestive tract.

Effects of cyflufenamid in the control of apple powdery mildew (Podosphaera leucotricha)

AB  - During 2019, the examination of cyflufenamid efficacy in apple protection from powdery mildew, caused by fungus Podosphaera leucotricha, was performed. The experiments were conducted in the orchards at the following locations: OŠD Radmilovac, Vinča (Topola), Kukujevci (Šid) and Donja Kravarica (Lučani), as a completely randomized block system in four replications (EPPO, 1999). The application of fungicides was performed using motorized mist blower sprayer type Solo 423, with water consumption of 1000 l ha-1. Starting from the growth stage "half-inch green" (BBCH 56-57) to the growth stage "beginning of fruit development" (BBCH 70-73), a total of five treatments were done. Cyflamid has shown high efficacy in the control of P. leucotricha. Effective protection of apple was achieved in the orchards at all four localities. At the locality Radmilovac, the efficacy was 98.8%, at the locality Vinča 96.2%. At the localites Kukujevci and Donja Kravarica, the efficacy was 96.7 and 98.8%, respectively
AB  - Tokom 2019. godine vršeno je ispitivanje efikasnosti preparata Cyflamid (a. s. ciflufenamid, 50 g/l) u zaštiti jabuke od prouzrokovača pepelnice (Podosphaera leucotricha). Ogledi su izvedeni na lokalitetima OŠD Radmilovac, Vinča (Topola), Kukujevci (Šid) i Donja Kravarica (Lučani), po potpuno slučajnom blok sistemu u četiri ponavljanja (EPPO, 1999). Primena fungicida vršena je leđnim atomizerom tipa Solo 423 uz utrošak vode od 1000 l/ha. Izvršeno je ukupno pet tretiranja počevši od fenofaze "zeleni buketić" (BBCH 56-57) do početka plodonošenja (BBCH 70-73). Ciflufenamid je ispoljio izuzetno visoku efikasnost u suzbijanju prouzrokovača pepelnice jabuke, na sva četiri lokaliteta. Na lokalitetu Radmilovac efikasnost je iznosila 98,8%. U Vinči efikasnost je bila 96,2%, dok je na lokalitetima Kukujevci i na lokalitetu Donja Kravarica utvrđena efikasnost 96,7 odnosno 98,8%.

The experiments were conducted in the orchards at the following locations: OŠD Radmilovac, Vinča (Topola), Kukujevci (Šid) and Donja Kravarica (Lučani), as a completely randomized block system in four replications (EPPO, 1999). The application of fungicides was performed using motorized mist blower sprayer type Solo 423, with water consumption of 1000 l ha-1. Starting from the growth stage "half-inch green" (BBCH 56-57) to the growth stage "beginning of fruit development" (BBCH 70-73), a total of five treatments were done. Cyflamid has shown high efficacy in the control of P. leucotricha. Effective protection of apple was achieved in the orchards at all four localities. At the locality Radmilovac, the efficacy was 98.8%, at the locality Vinča 96.2%. At the localites Kukujevci and Donja Kravarica, the efficacy was 96.7 and 98.8%, respectively, Tokom 2019. godine vršeno je ispitivanje efikasnosti preparata Cyflamid (a. s. ciflufenamid, 50 g/l) u zaštiti jabuke od prouzrokovača pepelnice (Podosphaera leucotricha). Ogledi su izvedeni na lokalitetima OŠD Radmilovac, Vinča (Topola), Kukujevci (Šid) i Donja Kravarica (Lučani), po potpuno slučajnom blok sistemu u četiri ponavljanja (EPPO, 1999). Primena fungicida vršena je leđnim atomizerom tipa Solo 423 uz utrošak vode od 1000 l/ha. Izvršeno je ukupno pet tretiranja počevši od fenofaze "zeleni buketić" (BBCH 56-57) do početka plodonošenja (BBCH 70-73). Ciflufenamid je ispoljio izuzetno visoku efikasnost u suzbijanju prouzrokovača pepelnice jabuke, na sva četiri lokaliteta. Na lokalitetu Radmilovac efikasnost je iznosila 98,8%. U Vinči efikasnost je bila 96,2%, dok je na lokalitetima Kukujevci i na lokalitetu Donja Kravarica utvrđena efikasnost 96,7 odnosno 98,8%.",

AB  - Didymella applanata, the causal agent of raspberry spur blight, is a destructive plant pathogen that can cause serious reductions in total yield. The use of fungicides plays a crucial role in successful control of the pathogen. The sensitivity of 94 isolates (66 collected during 2013 and 28 in 2017) of D. applanata to the succinate dehydrogenase inhibitor, fluxapyroxad, was evaluated in this study. In addition, the efficacy of fluxapyroxad in raspberry spur blight control in the field was determined. The isolates tested in this study showed different sensitivity to fluxapyroxad in vitro. The EC50 values of total number of isolates tested were ranged from 0.82 to 5.92 mu gml(-1). The ranges of EC50 values for the isolates varied between the localities where the isolates originated from and also the year of isolation. The mean EC50 values in the group of isolates obtained in 2013 were 1.95 mu gml(-1)+/- 0.81 (0.82 to 4.05 mu gml(-1)), while the EC50 values for those isolated in 2017 were ranged from 1.16 to 5.78 mu gml(-1) (mean=3.24 mu gml(-1)+/- 1.38). The efficacy trials were conducted during 2017 and 2018 in a commercial raspberry field at two locations in western part of Serbia. Applied in both concentration rates 0.02 and 0.03% of the formulated product (0.006 and 0.009% of a.i.), fluxapyroxad showed a very high efficacy in spur blight control in practical condition.

In vitro toxicity of fungicides with different modes of action to alfalfa anthracnose fungus, Colletotrichum destructivum

AB  - Sensitivity of 24 isolates of Colletotrichum destructivum O'Gara, collected from alfalfa plants in Serbia, to eight selected fungicides, was investigated in this study. Molecular identification and pathogenicity test of isolates tested were also performed. Fungicide sensitivity was evaluated in vitro, using mycelial growth assay method. All isolates exhibited significant pathogenicity, causing necrosis at the alfalfa seedling root tips two days after inoculation. Using the primer pair GSF1-SR1 and by comparing the amplified fragments of the tested isolates with the marker (M), the presence of the amplicon of the expected size of about 900 bp was determined for all isolates. The isolates tested in this study showed different sensitivity towards fungicides in vitro. Mycelial growth was highly inhibited by QoI (quinone outside inhibitors) fungicide pyraclostrobin (mean EC50=0.39 mu g mL(-1)) and by DMI (demethylation-inhibiting) fungicide tebuconazole (mean EC50=0.61 mu g mL(-1)), followed by azoxystrobin (mean EC50=2.83 mu g mL(-1)) and flutriafol (mean EC50=2.11 mu g mL(-1)). Multi-site fungicide chlorothalonil and MBC (methyl benzimidazole carbamate) fungicide thiophanate-methyl evinced moderate inhibition with mean EC50=35.31 and 62.83 mu g mL(-1), respectively. Thirteen isolates were sensitive to SDHI (succinate dehydrogenase inhibitors) fungicide boscalid and fluxapyroxad, (mean EC50=0.49 and 0.19 mu g mL(-1), respectively), while the rest of isolates were highly resistant.

Efficacy of fungicides in Botrytis cinerea control on blueberry

AB  - During the season 2017., biological efficacy of several different fungicides in blueberry protection from gray mold (Botrytis cinerea), was performed. The trials were conducted in conventional blueberry orchards at the locality of Šutci (Ljig) and Orašac (Aranđelovac) on varieties Duke and Draper. The experiments were performed as a randomized block system in four replications. The application of fungicides was carried out using the back sprayer, and a total of three treatments were performed, starting from the growth stage of flowering (more than 50% of open flowers BBCH 65), until seven days prior to harvest (BBCH 85). The assessment was performed by sampling 100 fruit from each variant tested, 10 and 20 days after last treatment (two assessments on both sites tested). Fruits were stored in the cold chamber and the severity of disease was evaluated after obvious differences between treated variants and control occurred. At the location of Šutci (Ljig) the severity of gray mold in untreated (control) was 27% during the first and 20.3% in the second assessment, while at the location of Orašac (Aranđelovac) severity of the disease was 23.3% in the first and 24.5% in the second assessment. The highest efficacy in both locaties and in both assessments was observed for the combination of active substances fluopyram+trifloxystrobin. High efficacy was registered for boscalid and the combination of pyraclostrobin+boscalid and cyprodinil+fludioxonil (80.6- 89.2%). Moderate efficacy was achieved using cyprodinil (60.5 - 65.6%) while fenhexamid exhibited significantly higher efficacy in the first (about 71%) than in second assessment (19.8 - 33.7%).
AB  - U toku 2017. godine vršena su ispitivanja biološke efikasnosti različitih fungicida u zaštiti borovnice od prouzrokovača sive truleži (Botrytis cinerea). Ogledi su sprovedeni u konvencionalnim zasadima borovnice na lokalitetima Šutci (Ljig) i Orašac (Aranđelovac), po tipu slučajnog blok sistema u četiri ponavljanja, na sortama Duke i Draper. Aplikacija fungicida je vršena pomoću leđne prskalice, a obavljeno je ukupno tri tretiranja, počevši od fenofaze cvetanja (više od 50% otvorenih cvetova BBCH 65), do sedam dana pred berbu (BBCH 85). Ocenjivanje efekata vršeno je uzorkovanjem po 100 zrelih plodova iz svake ispitivane varijante, 10 i 20 dana od poslednjeg tretiranja (po dve ocene na oba ispitivana lokaliteta). Plodovi su skladišteni u komori, a intenzitet oboljenja je ocenjen nakon jasnih razlika između tretiranih varijanti i kontrole.Na lokalitetu Šutci (Ljig) intenzitet pojave sive truleži u kontroli iznosio je 27% nakon prve, odnosno 20,3% nakon druge ocene, dok je na lokalitetu Orašac (Aranđelovac) pojava oboljenja u kontroli bila 23,3% nakon prve, odnosno 24,5% nakon druge ocene. Najveću efikasnost na oba ispitivana lokaliteta i u obe ocene ispoljio je fungicid na bazi kombinacije a. s. fluopiram + trifloksistrobin (91,7- 93,5%). Takođe, visoka efikasnost je utvrđena i kod fungicida na bazi a.s. boskalid, piraklostrobin + boskalid i ciprodinil + fludioksonil (80,6 -89,2%).
