AB  - Two square-planar complexes, [PtLCl] (1) and [PdLCl] (2), were synthesized with quinoline-2-carboxaldehyde thiosemicarbazone ligand (HL), and characterized by IR and NMR spectroscopy and single crystal X-ray diffraction analysis. In both complexes, L- is coordinated tridentately via the same donor atom set, while the fourth coordination site is occupied by a chloride ion. However, the complexes are not isostructural due to different types of non-covalent intermolecular interactions. These interactions were analyzed using Hirshfeld surfaces and two-dimensional fingerprint plots.

AB  - Cobalt complexes with semi-and thiosemicarbazones of 8-quinolinecarboxaldehyde have been synthesized and characterized by X-ray diffraction analysis. These novel complexes and a previously synthesized cobalt complex with a selenium-based selenosemicarbazone ligand showed myeloid differentiation activity on all trans retinoic acid resistant HL-60 acute myeloid leukaemia cells. They also showed varying levels of cytotoxicity on five human tumor cell lines: cervix carcinoma cells (HeLa), lung adenocarcinoma cells (A549), colorectal adenocarcinoma cells (LS-174), breast carcinoma cells (MDA-MB-361), and chronic myeloid leukaemia (K562) as well as one normal human cell line: fetal lung fibroblast cells (MRC-5). Leukaemia differentiation was most strongly induced by a metal-free oxygen ligand and the selenium ligand, whilst the latter and the cobalt(II) complex with an oxygen ligand showed the strongest dose-dependent cytotoxic activity. In four out of five investigated tumor cell lines, it was of the same order of magnitude as cisplatin. These best compounds, however, had lower toxicity on non-transformed MRC-5 cells than cisplatin.

AB  - Copper(II) complexes with the condensation derivative of methyl hydrazinoacetate and 2-acetylpyridine were synthesized. The X-ray crystal structures for both complexes revealed that they are polymerized isomers. A common feature of both complexes is the bidentate coordination of the ligand by one hydrazone and one pyridine nitrogen atom. In the monomeric complex, the copper(II) center is tetracoordinate, whereas dimerization through chlorido bridges results in a pentacoordinate arrangement about the metal ions in the dimer. The electronic and magnetic properties of both complexes are discussed on the basis of their X-ray structures, electron paramagnetic resonance (EPR) spectroscopy studies, and superconducting quantum interference device (SQUID) magnetization measurements combined with DFT calculations. Magnetostructural comparisons with structurally similar copper(II) complexes are also provided, and a possible correlation has been established. The antitumor activities of the Cu-II complexes were investigated against six different cancer cell lines, and the results suggest that the antiglioma action of the dimeric species is based on oxidative-stress-mediated phosphatidylserine externalization and caspase activation, which indicate apoptosis.

AB  - New square-planar Pd(II) and Pt(II) complexes with 8-quinolinecarboxaldehyde selenosemicarbazone have been synthesized and characterized by use of elemental analysis, molar conductivity measurements, and IR and NMR spectroscopy. The cytotoxic activity of the ligand, new Pt(II) and Pd(II) compounds, and previously synthesized Pd(II), Pt(II), Cd(II), and Ni(II) complexes with the analogous ligand, 2-quinolinecarboxaldehyde selenosemicarbazone, was tested against two human cancer cell lines: lung carcinoma (H460) and glioma (U251). The potential of these compounds to induce perturbations of the H460 cell cycle was also evaluated. These substances had an excellent radical-scavenging effect against ABTS radical cations. The best antimicrobial activity, among two yeasts and eight bacterial strains tested, was against Bacillus cereus.

AB  - In search for novel biologically active metal based compounds, an evaluation of in vitro cytotoxic, antioxidant, and antimicrobial activity of new Pt(II) complex and its Zn(II), Cu(II), and Co(III) analogues, with NNO tridentately coordinated N-heteroaromatic Schiff base ligand (E)-2-[N-(1-pyridin-2-yl-ethylidene)hydrazino]acetate, was performed. Investigation of antioxidative properties showed that all of the compounds have strong radical scavenging potencies. The Zn(II) complex showed potent inhibition of DNA cleavage by hydroxyl radical. A cytotoxic action of investigated compounds was evaluated on cultures of human promyelocitic leukaemia (HL-60), human glioma (U251), rat glioma (C6), and mouse melanoma (B16) cell lines. It was shown that binuclear pentacoordinated Zn(II) complex possesses a strong dose-dependent cytotoxic activity, of the same order of magnitude as cisplatin on B16, C6, and U251 cells. Furthermore, Zn(II) complex causes oxidative stress-induced apoptotic death of HL-60 leukemic cells, associated with caspase activation, phosphatidylserine externalization, and DNA fragmentation.

AB  - Novel Pd(II) complex with N-heteroaromatic Schiff base ligand, derived from 8-quinolinecarboxaldehyde (q8a) and ethyl hydrazinoacetate (haOEt), was synthesized and characterized by analytical and spectroscopy methods. The structure of novel complex, as well as structures of its quinoline and pyridine analogues, was optimized by density functional theory calculations, and theoretical data show good agreement with experimental results. A cytotoxic action of the complexes was evaluated on cultures of human promyelocytic leukemia (HL-60), human glioma (U251), rat glioma (C6), and mouse fibrosarcoma (L929) cell lines. Among investigated compounds, only complexes with quinoline-based ligands reduce the cell numbers in a dose-dependent manner in investigated cell lines. The observed cytotoxic effect of two isomeric quinoline-based complexes is predominantly mediated through the induction of apoptotic cell death in HL-60 cell line. The cytotoxicity of most efficient novel Pd(II) complex is comparable to the activity of cisplatin, in all cell lines investigated.

AB  - Thermal stability of a series of palladium(II) complexes with N-heteroaromatic bidentate hydrazone ligands was investigated using a combination of experimental measurements and DFT calculations. All complexes exhibit a reversible second-order transition around 333 K, which can be attributed to structural reorganization of the ligand molecules. Thermal degradation begins in 570-610 K temperature region, with an endothermic peak, followed by exothermic peaks in DSC. TG measurements show a well-defined mass loss corresponding to the initial degradation, while subsequent processes are poorly separated. DFT calculations suggest that the initial degradation step occurs with release of Cl, which then reacts with remaining part of the complex molecule in an exothermic process. This leads to decomposition of the ligand molecule into four fragments corresponding to ethyl chloride, carbon dioxide, methyl amine, and the fragment with the aromatic group. Mass spectrum suggests that creation of these fragments most likely corresponds to the initial degradation, after which some of these coordinate to Pd center, whose coordination sphere is left incomplete by release of Cl. TG measurement to 1123 K indicates that the final degradation product at this temperature is palladium.

AB  - Novel metal-based compounds with therapeutic potential have become the subject of intense investigations in inorganic chemistry and biomedical science. Recently, strong dose-dependent cytotoxic activities of selenosemicarbazone metal complexes against several human cancer cell lines were demonstrated. The aim of the present study was to investigate in vitro antioxidative potential of Ni(II), Cd(II) and Zn(II) selenosemicarbazone complexes. All three investigated complexes exhibited high 2,2'-azinobis(3-ethyl-benzothiazoline-6-sulphonic acid) radical cation (ABTS●+) scavenging capacity, comparable with ascorbic acid. In an acute toxicity study, administration of the compounds was performed orally to mice at single doses. The mice were observed for clinical signs, body weight effects and mortality for 14 days, after which they were sacrificed for gross organ necropsy. The body weight did not vary after administration, and the autoptic analysis failed to show appreciable macroscopic alterations of internal organs. Generally, the compounds exhibited low toxic effects as required for further in vivo therapeutic studies.
AB  - Nova jedinjenja na bazi metala sa terapeutskim dejstvom postala su predmet istraživanja u neorganskoj hemiji i biomedicinskim naukama. Pokazana je jaka dozno-zavisna citotoksična aktivnost kompleksa selenosemikarbazona na većem broju ćelijskih linija humanih ćelija kancera. Cilj rada je ispitivanje in vitro antioksidativnog dejstva selenosemikarbazonskih kompleksa nikla, cinka i kadmijuma. Sva tri ispitivana kompleksa pokazala su jaku antioksidativnu aktivnost prema ABTS radikalu, uporedivu sa aktivnošću askorbinske kiseline. Tokom ispitivanja akutne toksičnosti, jedinjenja su oralno davana miševima u pojedinačnim dozama i praćeni su klinički znaci, težina tela i mortalitet nakon 14 dana, a potom su životinje žrtvovane radi autopsije organa. Težina tela nije varirala nakon aplikacije. Generalno, jedinjenja su pokazala mali toksični efekat što i zahteva buduće in vivo terapeutsko ispitivanje.

AB  - Copper(II) and zinc(II) complexes with the condensation derivative of 2-acetylpyridine and hydrolyzed ethyl hydrazinoacetate were synthesized in a one pot reaction. Crystal structure of the zinc(II) complex was determined by single-crystal X-ray diffraction. In this binuclear complex, the ligand is coordinated via pyridine and imine nitrogen, while carboxylate oxygen is a bridge between metal centers. The fifth coordination site is occupied by a chloride, giving a distorted square pyramidal geometry around each zinc(II). The molecular structure of the copper(II) complex was determined from elemental analysis, infrared and electron paramagnetic resonance spectroscopy, molar conductivity, mass spectrometry, and measurements of the magnetic moment. Thermal behavior of both complexes was studied by thermogravimetric analysis coupled with mass spectrometer.

AB  - Two novel cadmium(II) complexes with condensation derivatives of 2-acetylpyridine or 2,6-diacetylpyridine with ethyl hydrazinoacetate hydrochloride were synthesized. X-ray crystal structures of the complexes revealed a bidentate coordination of the 2-acetylpyridine derivative, while the symmetric 2,6-diacetylpyridine derivative was coordinated tridentately. It was found that the oxygen atoms were not coordinated to the cadmium ions, despite the fact that they are the position to form six membered chelate rings. NMR spectroscopy investigations showed that the coordination properties of the ligands do not change in solution. Both complexes possess cytotoxic potential, as determined by the toxicity test to Artemia

AB  - Organometallic Cd(II) compounds have recently attracted attention for their anticancer activity. The interaction of the dinuclear complex of Cd(II) with the condensation product of 2-acetylpyridine and malonic acid dihydrazide, N',N'(2)-bis[(1E)-1-(2pyridyl) ethylidene] propanedihydrazide (Cd(II)H(2)L), with calf thymus DNA (CT-DNA) was monitored by blue shift in UV-vis spectra of the complex. The binding constant of Cd(II) H2L complex with CT-DNA was determined (K(B) = 1.8 x 10(4) M(-1)) and was indicative of minor groove binding. Agarose gel electrophoretic changes in mobility of supercoiled and circular forms of pBR322 and pUC18 plasmids in the presence of the complex suggest that conformational changes in the plasmids occur upon binding of the Cd(II) H2L complex. The Cd(II) H2L complex induced perturbation of the cell cycle phase distribution and an increase in the percentage of cells in the sub-G1 phase of human cervical cancer HeLa cell line and murine melanoma B16 cell line. Immunoblotting analysis showed the overexpression of Bcl-2 protein with the Cd(II)H(2)L complex.

AB  - Condensation derivatives of ethyl hydrazinoacetate with 2-formylpyridine and quinoline-2-carboxaldehyde were synthesized. Pd(II), Pt(II) and Cd(II) complexes with the 2-formylpyridine derivative and a Cd(II) complex with the quinoline-2-carboxaldehyde derivative were synthesized and characterized by spectroscopic techniques. In the complexes, both ligands are coordinated in neutral NN bidentate modes, while the remaining two coordination sites are occupied by chloride. All compounds showed biological activity when tested against Escherichia coli, Bacillus subtilis and Staphylococcus aureus.

X-ray crystal structures of all three complexes were determined. Crystallographic data indicate a common basis of the structures with some major differences. Both pyridine derivatives form complexes of the type [CuCl2L] with distorted square-planar geometry around copper(II), but show a decisive difference in the side chain orientation. The quinoline derivative forms a dimeric complex of type [Cu2Cl4L2] with distorted trigonal bipyramidal geometry around copper(II), but a rather short Cu center dot center dot center dot Cu contact, which is reflected in its magnetic properties. All three complexes showed antibacterial activity.

The assumed geometry of Pt(II) and Pd(II) complexes was square planar where the ligand was tridentately coordinated via the quinoline and imine nitrogen atoms and the selenium atom. The cytotoxic activity of the new Pt(II) and Pd(II) compounds, as well as of some previously synthesized Cd(II), Zn(II) and Ni(II) complexes with the same or analogous ligand, was tested against a panel of three human cancer cell lines: human cervix carcinoma cells (HeLa), human melanoma cells (FemX) and breast cancer cells (MDA-361). All investigated compounds, except Pt(II) complex, possess a strong dose-dependent cytotoxic activity of the same order of magnitude as cisplatin (CDDP). The investigation of potential of these compounds to induce HeLa cell cycle perturbations was also evaluated.

In complex (1) each Ni(II) ion is coordinated with two NNO donor atom sets from two H(2)L2 ligands forming an octahedral geometry. Similarly, in complex (2) the octahedral geometry of each Ni(II) ion is attained by coordination of two NNO donor atom sets, one from the neutral and the other from the monoanionic form of the ligand. The antimicrobial activity of the ligand and complexes is also presented.

AB  - A new dinuclear complex of Cd(II) with N',N'(2)-bis[(1E)-1-(2-pyridyl)ethylidene]propanedihydrazide (H2L) was synthesized and its crystal structure determined. The basic structural unit of the complex is the binuclear [Cd-2(H2O)(2)(H2L)(2)](4+) cation, where each cadmium atom is heptacoordinated by two tridentate H2L ligands, by means of the NNO chelating system, and one water molecule. A 12-membered macrocycle is defined at the core of the binuclear cation. The complex showed a pronounced cytotoxic activity to murine melanoma B16 cells and human cervical cancer HeLa cells.

AB  - New complexes of Pt(II) with condensation derivatives of ethyl hydrazino acetate and either 2-acetylpyridine or 2-quinolinecarboxaldehyde, and of Pd(II) with the condensation derivative of ethyl hydrazinoacetate and 2-quinolinecarboxaldehyde were synthesized and characterized by elemental analysis, IR, H-1- and C-13-NMR spectroscopy, and molar conductivity measurements. The complexes have a square planar geometry, ligands binding as bidentates in the neutral form, and the remaining two coordination sites being occupied by chloride ions. Biological activity of new complexes, and of previously synthesized Pd(II), Cd(II), Co(III) and Zn(II) complexes with this ligand type was evaluated by the brine shrimp test. All the complexes showed a moderate activity.

In the major Cd(II) complex one qasesc(-) ligand is coordinated as a tridentate, the fourth coordination site being occupied by acetate, while in the major Zn(II) complex two qasesc- ligands are coordinated as bidentates. In both minor complexes two qasesc- ligands are coordinated as tridentates forming the octahedral geometry around the central metal ion. The only paramagnetic complex in the series is Ni(II) complex for which X-ray structure analysis was performed. The complex has the angularly distorted octahedral geometry with two qasesc- ligands coordinated as tridentates, in a similar way as in the minor complexes of Cd(11) and Zn(11).
