Survey of Phytophthora infestans populations in Serbia: Mating type, metalaxyl resistance and virulence properties

AB  - A total of 37 single-lesion isolates of Phytophthora infestans were collected during 2000 from different potato growing regions in the central part of Serbia. They were paired in A1 and A2 matings to determine their mating types. Of these 37 isolates, 28 were found to belong to the A2 mating type, eight to the A1 type and one was found to be self-fertile (A1/A2). Using the in vitro mycelial radial growth method no metalaxyl resistant P. infestans isolate could be found. The rate of occurrence of intermediate isolates was not high, on the average 2.70%. All other isolates were sensitive to metalaxyl. There was no correlation between mating type and the intermediate metalaxyl resistance found.

Identification of phytoplasmas associated with grapevine yellows in serbia

AB  - The molecular identification and characterization of phytoplasmas from infected grapevines in four locations in Serbia are reported. Phytoplasmas were detected and identified by restriction fragment length polymorphism (RFLP) analysis of polymerase chain reaction (PCR) amplified 16S rDNA. Grapevine yellows were associated with three molecularly distinguishable phytoplasmas: Flavescence doree phytoplasmas (elm yellows group: 16SrV-C subgroup) were present only in the Zupa Aleksandrovac region; Bois noir phytoplasmas (stolbur group: 16SrXII-A subgroup) were detected in the other surveyed regions; a mixed infection of European stone fruit yellows (apple proliferation group: 16SrX-B subgroup) and Bois noir phytoplasmas was identified in one sample. A finer molecular characterization by RFLP analysis of rpS3 and SecY genes of Flavescence doree phytoplasmas from Zupa Aleksandrovac confirmed that the Serbian genotype is indistinguishable from a strain from the Veneto region, Italy. Characterization of the tuf gene of Bois noir phytoplasmas showed lack of amplification of samples from Erdevik. HpaII profiles of tuf gene PCR products of samples from Pali and Radmilovac were identical, and were indistinguishable from one of the two profiles produced by samples from Italian grapevines used as reference strains.

Population of bacteria infecting cauliflower in Yugoslavia

AB  - During the last several years, samples of diseased cauliflower plants were collected from different growing regions in Yugoslavia. Two types of symptoms were noticed: dark brown or black rot of cauliflower heads and V-shaped chlorosis along the leaf margin. Both types of symptoms were caused by phytopathogenic bacteria. Cultural, biochemical, physiological and serological characteristics of the strains isolated were studied. The results obtained indicated that several pathogenic bacteria, belonging to different genera, were responsible for the symptoms on cauliflower plants. According to the bacteriological characteristics, as causal agents of dark brown cauliflower head rot in Yugoslavia Pseudomonas viridiflava, P. cichorii and P. marginalis pv. marginalis were identified. However, from the soft rotted tissue the strains of Erwinia carotovora subsp. carotovora were isolated. Apart from bacteria belonging to genera Pseudomonas and Erwinia, Xanthomonas campestris pv. campestris was identified as a causal agent of cauliflower leaf chlorosis and black rot of vascular system.

Potato Virus Y (PVY) on pepper in Serbia

AB  - During 1997, samples of plants showing symptoms of a viral disease were collected from thirteen pepper growing areas. On some plants characteristic symptoms of Potato Virus Y (PVY) infection were observed. Pepper samples were tested by applying the direct immunoenzyme test (DAS ELISA). For the identification of the virus a PVY antiserum was used, i.e. the kit of Loewe Biochemica, GmbH Germany. Isolation of the virus was performed by the method of mechanical inoculation using 0.01 M phosphate buffer (pH-7) and 0.1% Na2SO3 from the infected pepper samples. Identification of the virus strains was performed on the basis of the reaction of differential pepper genotypes. The virus was identified according to pepper disease symptoms, DAS ELISA test, reaction of inoculated test plants, way of transmission and its biophysical constants. On the basis of the observed reaction of the differential pepper genotypes, two strains of this virus were identified.

AB  - In these experiments, the reaction of 24 inbred lines of maize to the new herbicides Equip (formasulfuron), Callisto (mesotrione) and a combination of Motivel (nicosulfuron) with Banvel 480 S (dicamba) was investigated. The herbicides were applied in the growth phase of 3-5 leaves. Equip decreased the grain yield of 15 inbred lines to 30.5%, Callisto of 12 to 62.7% and the combination Motivel + Banvel 480 S of 5 inbred lines of maize to 19%).
