Primena Rep-PCR i nekih klasičnih metoda u detekciji Xanthomonas arboricola pv. pruni

Abstract
Xanthomonas arboricola pv. pruni represents economically very important pathogen of stone fruits worldwide. This bacterium has not been detected in Serbia, and therefore is considered quarantine pathogen in this country. Its status in European countries and in Serbia requires inspection of imported planting material at the border check points, in order to protect introduction of the pathogen in our country. Therefore, methods for detection and identification of this pathogen were proposed and established by European Plant Protection Organization (EPPO). Absence of the bacterium in Serbia consequently resulted in lack of practical experience in symptomatology of the disease as well as detection methods. Thus, in collaboration with the colleagues from University of Modena and Reggio Emilia, Italy, we acquired common laboratory techniques recommended by EPPO for preliminary diagnosis and detection of the pathogen and enabled their routine use in Laboratory for phytobacteriology, Faculty o...f Agriculture, Belgrade. In this paper we presented our practical experience in applying some of these methods.

Xanthomonas arboricola pv. pruni spada u ekonomski veoma značajne fitopatogene bakterije u svetu. Njeno prisustvo u našoj zemlji nije do sada utvrđeno pa se s toga nalazi na nacionalnoj A1 listi karantinskih patogena. Karantinski status nameće potrebu kontrole zdravstvenog stanja biljnog materijala kojim bi se ovaj patogen mogao proširiti iz zaraženih područja u nove predele pa i u našu zemlju. Stoga su razrađene i od strane Evropske organizacije za zaštitu bilja (European Plant Protection Organization - EPPO) preporučene laboratorijske metode kojima se na standardizovan način može izvršiti detekcija i indentifikacija patogena (EPPO Standards PM 7/64 (1). Odsustvo patogena u Srbiji imalo je za posledicu nedostatak praktičnog iskustva u prepoznavanju karakterističnih simptoma bolesti i detekciji navedenog patogena. Stoga smo u saradnji sa bakteriolozima na Univerzitetu Modena i Reggio Emilia, Italija, savladali najčešće korišćene laboratorijske tehnike neophodne za preliminarnu dijagnoz...u i detekciju patogena procedurama propisanim od strane EPPO i tako omogućili njihovu rutinsku primenu u Laboratoriji za fitobakteriologiju Poljoprivrednog fakulteta u Beogradu. Ovim radom prenosimo domaćoj naučnoj javnosti praktična iskustva u primeni pojedinih procedura.
