AB  - The aim of this study was to compare the aromatic profile of plum spirits, obtained from the Požegača and Stanley varieties, as affected by different combinations of pre-distillation steps used in traditional and six modified production methods that are common in Serbian distilleries. Traditional plum spirits (produced from spontaneously fermented mashes of crushed plums with stones distilled after two months of storage) had the highest contents of ethyl acetate, benzaldehyde, and total acids, which resulted in the occurrence of an unpleasant solvent-like and stone-like odour and acidic taste. These sensory defects were overcome by the distillation of fermented mashes immediately after the completion of alcoholic fermentation. Depending on the combination of the pre-distillation steps, plum spirits from mashes distilled immediately after alcoholic fermentation were characterized by different aromatic profiles: closer to traditional (from spontaneously fermented crushed plums with/without stones) or with a more pronounced fruity character (from pulped plums without stones regardless of the way of fermentation). These differences in aroma profiles have arisen mostly because of the significantly different contents and OAVs of ethyl esters and volatile fatty acids. The appropriate combination of the pre-distillation steps, which is adapted to the variety, can significantly improve the quality of the plum spirit compared to the traditionally produced spirit. © 2023 by the authors.

AB  - Pollination and temperature are predominant factors affecting the progamic phase of sexual plant reproduction. In light of climate change and the emergence of increasing temperatures, new knowledge is necessary to encourage fruit breeding programmes to develop cultivars adapted to these changes, as well as to find the best incompatibility relationships with the aim of achieving high fruit quality and yield. This research was conducted to reveal the effect of pollination treatments and temperature on the progamic phase and fruit set in three European plum (Prunus domestica L.) cultivars (‘Jojo’, ‘Hanita’ and ‘Katinka’) resistant/tolerant to Sharka virus using fluorescent microscopy. Reproductive parameters (flowering overlapping, pollen germination, number of pollen tubes in the upper part of the style and locule of the ovary, dynamics of pollen tubes growth and initial and final fruit set) were investigated in vivo under different pollination treatments (cross, open and self-pollination). The influence of different temperature regimes (20 °C, 23 °C and 26 °C) on the progamic phase was also observed under cross and self-pollination. The pollination treatments significantly influenced the number of pollen tubes in the style and ovary, as well as dynamics of pollen tubes in different regions of pistil. Pollen tubes showed better growth in different crossing combinations compared to open and self-pollination. Higher temperatures accelerated pollen tube growth and reduced the time they need to reach ovules. Generally, ‘Jojo’ appeared to be the best pollenizer among the examined cultivars, while ‘Katinka’ exhibited the best response of pollen tube growth to various temperature regimes. The differences observed between growth of the pollen tubes in each genotype-temperature combination revealed that certain crossing combinations were better adapted to higher temperatures so this can be taken into account as one of the adjustments to climate change. The ability of cultivars to respond to high temperatures in the progamic phase can also be helpful for breeding programs in developing new cultivars better adapted to different temperature conditions.

Neopestalotiopsis clavispora: a causal agent of twig dieback on highbush blueberries in Serbia

AB  - Highbush blueberry (Vaccinium corymbosum L.) is becoming an important berry fruit in Serbian agriculture, and the areas under blueberries are rapidly expanding. Since 2011, blueberry bushes with the symptoms of twig dieback and shoot canker were noticed in four important producing regions in the country. To identify the causal agent, 40 symptomatic bushes from four districts were sampled and analyzed. Based on morphological characteristics, all analyzed isolates were preliminary identified as Neopestalotiopsis clavispora. Selected isolates were further characterized by sequence analysis of the rDNA internal transcribed spacer region, translation elongation factor 1-alpha, and β-tubulin (TUB). Sequence and phylogenetic analysis confirmed that all isolates belong to N. clavispora. The pathogenicity tests on blueberry ‘Elliot’ confirmed that tested isolates were pathogenic. The detection of N. clavispora, a causal agent of twig dieback on blueberries in Serbia, is the first report of this pathogen in the country.

AB  - A study was conducted to investigate the effect of different storage periods and temperatures on pollen viability in vitro and in vivo in plum genotypes ‘Valerija’, ‘Čačanska Lepotica’ and ‘Valjevka’. In vitro pollen viability was tested at day 0 (fresh dry pollen) and after 3, 6, 9 and 12 months of storage at four different temperatures (4, −20, −80 and −196◦C), and in vivo after 12 months of storage at distinct temperatures. In vitro germination and fluorescein diacetate (FDA) staining methods were used to test pollen viability, while aniline blue staining was used for observing in vivo pollen tube growth. Fresh pollen germination and viability ranged from 42.35 to 63.79% (‘Valjevka’ and ‘Čačanska Lepotica’, respectively) and 54.58 to 62.15%, (‘Valjevka’ and ‘Valerija’, respectively). With storage at 4◦C, pollen viability and germination decreased over the period, with the lowest value after 12 months of storage. Pollen germination and viability for the other storage temperatures (−20, −80 and −196◦C) were higher than 30% by the end of the 12 months. Pollination using pollen stored at 4◦C showed that pollen tube growth mostly ended in the lower part of the style. With the other storage temperatures, pollen tube growth was similar, ranging between 50 and 100% of the pistils with pollen tubes penetrated into the nucellus of the ovule in the genotype ‘Čačanska Lepotica’. Using scanning electron microscopy to characterise plum (Prunus domestica L.) genotypes

AB  - Pollen grain micromorphological characterisation of four plum (Prunus domestica L.) genotypes from two different breeding programmes (Fruit Research Institute, Serbia, and Hohenheim, Germany) was performed using scanning electron microscopy (SEM). The studied characteristics were pollen size and shape, colpus length, mesocolpium width as well as exine characteristics. All genotypes had tricolpate pollen grains and striate exine ornamentation, with more parallel longitudinal ridges. The most significant differences among the analysed genotypes concerned the length of the pollen grain and exine sculpturing. The genotypes 'Pozna Plava', 'Presenta' and 'Čačanska najbolja' had the largest pollen grains (> 57 µm), whereas 'Hanita' showed a smaller pollen size, below 54 µm. 'Pozna Plava' had perprolate pollen in the equatorial view (L/W ratio 2.06), while the other three genotypes had prolate shaped pollen. By analysing the pollen morphological characteristics of the plum varieties in this paper, primarily the shape, size, length of colpi and, most importantly, the sculpture of the exine, each variety showed a specificity which can be used together with other morphological characteristics for description and characterisation.

The most significant differences among the analysed genotypes concerned the length of the pollen grain and exine sculpturing. The genotypes 'Pozna Plava', 'Presenta' and 'Čačanska najbolja' had the largest pollen grains (> 57 µm), whereas 'Hanita' showed a smaller pollen size, below 54 µm. 'Pozna Plava' had perprolate pollen in the equatorial view (L/W ratio 2.06), while the other three genotypes had prolate shaped pollen. By analysing the pollen morphological characteristics of the plum varieties in this paper, primarily the shape, size, length of colpi and, most importantly, the sculpture of the exine, each variety showed a specificity which can be used together with other morphological characteristics for description and characterisation.",

Physicochemical and sensory characteristics of promising plum (Prunus domestica L.) genotypes bred at fruit research institute, Čačak

AB  - Five plum hybrids (38/62/70, IV/63/81, 32/21/87, 34/41/87 and 22/17/87) and newly released cultivar ‘Nada’, obtained by planned hybridisation and singled out within breeding programme at Fruit Research Institute, Čačak, were assessed for the main physical (fruit and stone weight and flesh percentage), chemical (soluble solids content, total and inverted sugars content, sucrose content, total acids content, pH value of fruit juice, ratio of soluble solids and total acids content and ratio of total sugars and total acids content) and sensorial (attractiveness, taste, aroma and consistency) traits compared with the standard cultivar ʻČačanska Lepoticaʼ. Results showed that the studied plum genotypes differed significantly in all of the assessed traits. Regarding the physical features, the best results were shown by hybrid 38/62/70, which had the highest fruit weight (56.92 g) and flesh percentage (96.91%), as well as by the new cultivar ‘Nada’, for which a larger fruit was observed in comparison to the standard (45.54 g and 42.24 g, respectively). Also, ‘Nada’ had better sensorial properties such as attractiveness, taste, aroma and consistency in relation to the other promising hybrids and the standard cultivar. From the aspect of all the studied sensorial characteristics, in addition to ‘Nada’, only hybrid 38/62/70 was aligned with the standard cultivar. Out of the studied genotypes, late ripening hybrid 22/17/87 had the best values of parameters of fruit chemical composition such as the soluble solids content (17.01%), total and inverted sugars contents (12.31% and 8.96%, respectively). The highest sucrose content (3.39%), pH value of fruit juice (3.51), as well as the highest ratio between soluble solids and total acids content (43.72) and ratio between total sugars and total acids content (32.58) were found in cultivar ‘Nada’. The highest total acids content (1.42%) was recorded in hybrid 32/21/87. Compared to ʻČačanska Lepoticaʼ, the same or better results in terms of the fruit chemical composition were determined in cultivar ‘Nada’ and hybrids 34/41/87 and 22/17/87. The study revealed existence of significant correlations between individual studied parameters of the fruit physical, chemical and sensorial properties.
