Dietary intake and health risk assessment of essential and toxic elements in pepper (Capsicum annuum)

AB  - The content of twenty-one essential and toxic elements in fresh, dried, and processed peppers (Capsicum annuum) obtained from the Serbian market was analyzed. The concentration of the elements varied significantly between the analyzed samples. The most abundant essential element was K with an average concentration of 8.4 g/kg fresh weight. All processed products also had increased Na content, reaching a maximum value of 15.8 g/kg fresh weight in peppers from the pepper-meat products. Toxic elements Pb, Cd, and As were found in all analyzed samples, while Hg was detected in a few samples. Based on guideline values or recommended dietary intake, pepper and pepper products are not a significant source of essential elements. The health risk assessment was conducted by calculating target hazard quotient (THQ) and carcinogenic risk (CR), as well as comparing estimated weekly (Al, Hg, Cd) and daily intake (As, Pb) with appropriate PTWI and BMDL values. The risk assessment results indicate that peppers and pepper products are mostly safe for consumption both for adults and children. However, consumption of some pepper samples may pose a health risk due to an incremental lifetime cancer risk (ILCR) value higher than 10−4

Rosehip Extract-Loaded Liposomes for Potential Skin Application: Physicochemical Properties of Non- and UV-Irradiated Liposomes

AB  - In the present study, rosehip (Rosa canina L.) extract was successfully encapsulated in phospholipid liposomes using a single-step procedure named the proliposome method. Part of the obtained liposomes was subjected to UV irradiation and non-treated (native) and UV-irradiated liposomes were further characterized in terms of encapsulation efficiency, chemical composition (HPLC analysis), antioxidant capacity, particle size, PDI, zeta potential, conductivity, mobility, and antioxidant capacity. Raman spectroscopy as well as DSC analysis were applied to evaluate the influence of UV irradiation on the physicochemical properties of liposomes. The encapsulation efficiency of extract-loaded liposomes was higher than 90%; the average size was 251.5 nm; the zeta potential was -22.4 mV; and the conductivity was found to be 0.007 mS/cm. UV irradiation did not cause a change in the mentioned parameters. In addition, irradiation did not affect the antioxidant potential of the liposome-extract system. Raman spectroscopy indicated that the extract was completely covered by the lipid membrane during liposome entrapment, and the peroxidation process was minimized by the presence of rosehip extract in liposomes. These results may guide the potential application of rosehip extract-loaded liposomes in the food, pharmaceutical, or cosmetic industries, particularly when liposomal sterilization is needed.

AB  - This study investigates the effects of ultrasound, in combination with chemical pretreatments, on the quality attributes (total phenolic and carotenoid content, antioxidant activity (2,2-Diphenyl-1-picrylhydrazyl assay (DPPH)), ferric-reducing ability (FRAP), CIE L* a* b* color, non-enzymatic browning, rehydration ratio, textural and morphological properties) of red pepper subjected to drying (hot air drying or freeze drying). The fractional factorial design was used to assess the impact of factors. The global Derringer desirability function was used to determine the optimal conditions for the best quality attributes of dried pepper. The drying method influenced total phenolic content, a* (redness), and initial rehydration ratio; pretreatment time significantly affected FRAP antiradical activity, a*, chroma and non-browning index, while pH-value had a significant effect on the texture of dried pepper. Non-enzymatic browning was reduced to 72.6%, while the DPPH antioxidant capacity of freeze-dried peppers was enhanced from 4.2% to 71.9%. Ultrasonic pretreatment led to changes in the pepper morphology, while potassium metabisulfite (KMS) was a more effective additive than citric acid.

Investigation of Anticholinesterase Activity of Chemically Characterised Hieracium s. str. Methanol Extracts and Their Selected Metabolites

AB  - The composition and anticholinesterase activity of the dried MeOH extracts of Hieracium scheppigianum and H. naegelianum underground parts (rhizomes and roots), as well as the anticholinesterase activity of the dried, previously chemically characterised MeOH extracts of the flowering aerial parts of these two and 26 other Hieracium species in the strict sense (s. str.), were investigated. Furthermore, the anticholinesterase activity of 12 selected secondary metabolites of these extracts was evaluated. Using semi-preparative LC-MS, five caffeoylquinic acids and the sesquiterpene lactone crepiside E were isolated from H. scheppigianum underground parts extract. All these compounds were also identified in the underground parts extract of H. naegelianum. Quantitative LC-MS analysis showed that the analysed underground parts extracts were rich in both caffeoylquinic acids (139.77 and 156.62 mg/g of extract, respectively) and crepiside E (126.88 and 116.58 mg/g). In the Ellman method, the tested extracts showed an interesting anti-AChE and/or anti-BChE activity (IC50=0.56–1.58 mg/mL), which can be explained, at least partially, by the presence of some of their constituents. Among the metabolites tested, the best activity was revealed for the flavonoids apigenin, luteolin and diosmetin, and the sesquiterpene lactone 8-epiixerisamine A (IC50=68.09–299.37 μM). © 2023 Wiley-VHCA AG, Zurich, Switzerland.

AB  - The production of functional beverages has gained more attention because of growing consumers' interest in personal health. The process of carrier production and characterization of a novel type of protein-carbohydrate carriers for functional beverages with a focus on cell protection during fermentation, gastrointestinal, and storage conditions are shown in this study. Probiotic starter culture was microencapsulated using spray and freeze-drying techniques and then implemented in whey-based beverages. Encapsulation materials used are whey protein concentrate, whey, and sodium alginate. The encapsulation efficiency results were over 76.8% and 81.4%, after spray and freeze-drying respectively, depending on the carrier formulation. The culture in freeze-dried carriers reached a higher survival rate (>7.5 log10(CFUg−1)) than one's encapsulated by spray-drying technique during storage. Microencapsulation of the culture significantly improved cell viability in gastrointestinal and storage conditions compared with free cells. Usage of both encapsulation techniques showed potential regarding cell preservation in production and storage.

The effect of process variables was investigated by using Box–Behnken design. Each essential element had a different optimal condition for minimal leaching, while the toxic elements were predominantly extracted with acetic acid solution as extractant. The highest correlation between extraction conditions and concentration of extracted elements was found for aqueous extracts of mature green pepper. The concentration of acetic acid highly influenced the leaching of elements and a strong correlation between the concentration of Al, B, Fe, and Mn, and the UAE experimental conditions were observed. Practical applications: This study investigates the leaching of minerals and heavy metals present in vegetables to the most commonly used media (water and acetic acid) during processing and canning. The data from this study may be useful for improving the quality of processed pepper products due to new information on how different parameters influence these losses (ripening stage of peppers, medium/solvent used for preparation, or storage, applied temperatures, time, mass to volume ratio). Essential elements are better preserved during the processing of intermediate breaker and mature red pepper compared to green mature fruits. Better retention of essential elements in the breaker and mature red pepper (that are mostly used in industrial processing) can be achieved by reducing the contact time with water, as well as mass to volume ratio. AB  - This study collected agricultural surface soil samples from 200 sites in the district of Braničevo, located in the Carpathian Mountains – Pannonian Basin south of the Danube River (Serbia). The main objective was to determine the soil contamination by ten potentially toxic elements (As, B, Cd, Cr, Cu, Mn, Mo, Ni, Pb, Zn) and evaluate the associated ecological risk via different indices. The physicochemical parameters, pH, organic carbon, water content, and soil texture were also analyzed. The mean values of most metal concentrations remained below their corresponding national target values, except for Mo and Ni. The main soil texture types were silt loam (40.5 %) and silty clay loam (34.5 %). According to the mean values, pollution load index (PLI) and potential ecological risk (RI) demonstrated that the soil in the study area was exposed to moderate pollution and moderate ecological risk, while enrichment factor (EF), geoaccumulation index (Igeo), and contamination factor (CF) revealed very high enrichment and contamination with Mo, implying the impact of anthropogenic activities. There was a lack of strong correlations among elements and soil properties, except for Cd and Corg, while moderate to strong positive inter-metal relationships suggested their common sources. The chemometric analysis illustrated the classification of sampling sites into two distinct clusters of spatial similarities according to higher and lower metal concentrations. AB  - A study was conducted to investigate the effect of different storage periods and temperatures on pollen viability in vitro and in vivo in plum genotypes ‘Valerija’, ‘Čačanska Lepotica’ and ‘Valjevka’. In vitro pollen viability was tested at day 0 (fresh dry pollen) and after 3, 6, 9 and 12 months of storage at four different temperatures (4, −20, −80 and −196◦C), and in vivo after 12 months of storage at distinct temperatures. In vitro germination and fluorescein diacetate (FDA) staining methods were used to test pollen viability, while aniline blue staining was used for observing in vivo pollen tube growth. Fresh pollen germination and viability ranged from 42.35 to 63.79% (‘Valjevka’ and ‘Čačanska Lepotica’, respectively) and 54.58 to 62.15%, (‘Valjevka’ and ‘Valerija’, respectively). With storage at 4◦C, pollen viability and germination decreased over the period, with the lowest value after 12 months of storage. Pollen germination and viability for the other storage temperatures (−20, −80 and −196◦C) were higher than 30% by the end of the 12 months. Pollination using pollen stored at 4◦C showed that pollen tube growth mostly ended in the lower part of the style. With the other storage temperatures, pollen tube growth was similar, ranging between 50 and 100% of the pistils with pollen tubes penetrated into the nucellus of the ovule in the genotype ‘Čačanska Lepotica’. Freeze vs. Spray Drying for Dry Wild Thyme (Thymus serpyllum L.) Extract Formulations: The Impact of Gelatin as a Coating Material

AB  - Freeze drying was compared with spray drying regarding feasibility to process wild thyme drugs in order to obtain dry formulations at laboratory scale starting from liquid extracts produced by different extraction methods: maceration and heat-, ultrasound-, and microwave-assisted extractions. Higher total powder yield (based on the dry weight prior to extraction) was achieved by freeze than spray drying and lower loss of total polyphenol content (TPC) and total flavonoid content (TFC) due to the drying process. Gelatin as a coating agent (5% w/w) provided better TPC recovery by 70% in case of lyophilization and higher total powder yield in case of spray drying by diminishing material deposition on the wall of the drying chamber. The resulting gelatin-free and gelatin-containing powders carried polyphenols in amount ~190 and 53-75 mg gallic acid equivalents GAE/g of powder, respectively. Microwave-assisted extract formulation was distinguished from the others by a higher content of polyphenols, proteins and sugars, higher bulk density and lower solubility. The type of the drying process mainly affected the position of the gelatin-derived -OH and amide bands in FTIR spectra. Spray-dried formulations compared to freeze-dried expressed higher thermal stability as confirmed by differential scanning calorimetry analysis and a higher diffusion coefficient; the last feature can be associated with the lower specific surface area of irregularly shaped freeze-dried particles (151-223 µm) compared to small microspheres (~8 µm) in spray-dried powder.

AB  - The alkali treated subglebal tissue of the mosaic puffball (Handkea utriformis) (Sa) and Sa modified with hydroxyapatite (Sa-HAp), obtained by successive ionic layer adsorption and reaction (SILAR) method, were used for the removal of Pb2+, Cd2+ and Ni2+ from aqueous solution. The materials were characterized by FT-IR, Raman, SEM and EDS analysis and by determination of pHPZC. The adsorption performances of Sa and Sa-HAp were assessed in batch experiments at different pH, contact times, temperatures and mass of the adsorbent. Different models of adsorption isotherms were used, and the best fit was obtained with the Langmuir model. Maximum adsorption capacities of Sa towards Pb2+, Cd2+ and Ni2+ were 44.82, 15.54 and 17.21 mg g−1, while for Sa-HAp were 79.55, 52.59 and 45.01 mg g−1, respectively. Kinetic data were well fitted by a pseudo second-order model, while thermodynamic studies disclose spontaneous and endothermic adsorption process. The Sa-Hap was successfully regenerated with 1 M NaCl and after the fifth desorption cycle and 10 h achieved 82.9, 69.7 and 60.4 %, while for 0.5 M NaCl + 0.5 M NaOH and 1 h was 78.3, 64.1, 57.5 % of desorbed Pb2+, Cd2+ and Ni2+, respectively. The competitive study and results from a column system confirmed good applicability of Sa-HAp adsorbent.

Characterization of untreated/treated HNT was performed by Fourier-transformation infrared (FTIR) spectroscopy, and X-ray diffraction (XRD). The quantity of grafted molecules and thermal stability of newly synthesized materials were determined by thermogravimetric (TG) and derivative thermogravimetric (DTG) analysis. Tensile properties of newly synthesized materials were compared, and scanning electron microscopy (SEM) and transmission electron microscopy (TEM) analysis of the fracture surfaces were performed. Incorporation of APTES modified HNT (HNTAPT) and two-step modification APTES followed by DGEBA (HNTAPTDG) has increased the tensile strength of the nanocomposite materials up to 72% and 61%, and strain at break up to 1082% and 1216%, respectively, compared to neat epoxy. It was concluded that the modification of HNT contributed to the enhancement of the dispersion and the cross-linking in the epoxy resin matrix.

AB  - In this paper, analyzed are the effects of the plasma spray distance on the microstructure and mechanical properties of the NiCrAlCoY2O3 coating layers deposited at atmospheric pressure. The microstructure and mechanical properties of the coating layers are under the influence of the interaction of plasma particles (ions and electrons) with powder particles, providing the transfer of velocity and temperature of the plasma particles onto the powder particles. The effect of the interaction is directly dependent on the time the powder particles were present in the plasma which is defined by distance of the plasma gun from the substrate, depending on the granulation of the powder, the melting point and specific gravity. In order to obtain homogeneous and denser coating layers with high adhesion, in the experiment three distances from the substrate were used: 95 mm, 105 mm and 115 mm. The layers were deposited on thin sheets of AlMg1 aluminum thickness of 0.6 mm. Evaluation of mechanical properties of the layers was carried out by examining microhardness using the HV0.1 method and the bond strength by tensile testing. The morphology of the powder particles was examined on the SEM, while the microstructure of the layers was evaluated under a light microscope in accordance with the Pratt Whitney standard. The results of the experiment showed that the distance from the substrate substantially influenced the structure and mechanical properties of the coating layers. The best deposited layers were examined in the system with the ZrO224%MgO ceramic coating, which have proved to be reliable protectionfrom high temperature and abrasive rocket jet fuel.

AB  - The aim of this study was to deposit multi-functional ZrO28%Y2O3 coating layers using the plasma spray technology and then to characterise such layers. In combination with other biomedical ceramics, this coating is intended for the application in implant surface modification. The examination was focused on the mechanical properties and microstructure layers. Using the atmospheric plasma spraying, duplex ZrO28%Y2O3/Ni22Cr10Al1Y coating system was deposited on the X15Cr13 stainless steel, with two different thicknesses of the bond and ceramic coatings. The microstructure was analysed using an optical microscope, including the assessment of the content of micropores. The morphology of powder particles and ceramic coating surfaces were examined on a scanning electron microscope (SEM). The quality of the ZrO28%Y2O3 layers makes them suitable for the application and combination with other materials to create a system of biomedical or multifunctional coatings.

AB  - In this paper, studied was the melting of W powder particles in plasma, their behavior at oxidation as well as the mechanism of hardening on the surface of the substrate. Tungsten coating layers were deposited with vacuum plasma spray technology (VPS) on the test specimens of steel Č.4171 (X15Cr13 EN10027). VPS technology has advantages over the APS technology due to decreased oxidation of melted powder particles, by producing a coating with a controlled proportion of micro pores and greater uniformity of the deposited layers. Evaluation of mechanical characteristics of the layers was done by examining the microhardness using the HV0.3 method and tensile bond strength by tensile testing. The microstructures of the deposited layers were analyzed by means of optical microscopy (OM) and scanning electron microscopy (SEM). The results obtained showed that the tungsten coating consisted of well melted particles that interconnected and were overlapping, which shows a typical lamellar microstructure. Present in the microstructure are micro pores in very small proportion at the inter-lamellar boundaries. Tests have shown that layers of W coating deposited by plasma spray in inert gas shielding at low pressure have good mechanical properties and microstructure, which in the fully enables its use in exploitation.
