Antioxidant activity of water extracts from fruit body of Lentinus edodes enriched with selenium

AB  - Shiitake (Lentinus edodes) belongs to medically important and delicious fungi. It is recognizable for its healing properties, excellent taste and rich aroma. According to the traditional Japanese and Chinese medicine, shiitake mushroom significantly increases the strength and vitality of the body. Shiitake contains immunostimulants, compounds that lower cholesterol, prevents clogging of blood vessels, regulates the pressure, balances blood sugar levels, regulates digestion, and improves the performance of respiratory organs by its antirheumatic and antiallergic activities. Shiitake is recommended to use as food, prevention and cure, usually in a form of a spice (dried and ground) or tea. It can be consumed fresh, too. The objective of this study was to test the effect of enrichment in selenium on antioxidant, reducing and free radical scavenging activity of water extracts from fruit body of Lentinus edodes. The fungus was enhanced by adding organic selenium, zinc (II) complex with the ligand 2.6-bis diacetylpyridine (selenosemicarbazon) and inorganic compounds (Na2SeO3) of selenium in nutritional substrate where the fungus was grown. The total selenium content in fruit body was around 50 ppm for the sample enriched with selenium originating from organic sources, and 80 ppm for the sample enriched with selenium from inorganic sources. Samples were prepared by extraction of fruiting bodies in heated water. The results indicated that water extracts of whole fruit bodies, from both control and mushrooms supplemented with selenium, had quite good antioxidant activity. However, there was no significant difference between the samples supplemented with selenium content and those that were not.
AB  - Shiitake (Lentinus edodes) pripada grupi medicinski značajnih i delikatesnih gljiva. Prepoznatljiva je po svojoj lekovitosti, izvanrednom ukusu i bogatoj aromi. Prema tradicionalnoj japanskoj i kineskoj medicini, gljiva shiitake značajno povećava snagu i vitalnost organizma. Shiitake sadrže imunostimulanse, sastojke koji snižavaju holesterol, sprečavaju začepljenje krvnih sudova, regulišu pritisak, uravnotežuju nivo šećera u krvi, regulišu probavu, poboljšavaju rad disajnih organa, deluju antireumatski i antialergijski. Preporuka je da se shiitake koriste kao ukusna hrana, preventiva i lek, najčešće kao začin (sušene i mlevene) ili čaj. Mogu se konzumirati i kao sveže pripremljene. Cilj rada bio je da se razjasni da li selen dodat u supstrat za gajenje gljive Lentinus edodes utiče na redukciona svojstva ekstrakta, antioksidativnu aktivnost ekstrakta, kao i procena reaktivnosti ekstrakata prema radikalskim vrstama. Gljiva je obogaćena selenom dodavanjem organskih, Zn (II) kompleks sa ligandom 2,6-diacetilpiridin bis (selenosemicarbazon), i neorganskih jedinjenja (Na2SeO3) selena u hranljivi supstrat na kojem je gljiva uzgajana. Ukupan sadržaj selena u plodonosnom telu kretao se oko 50 ppm za uzorak obogaćen selenom iz organskog izvora i 80 ppm za uzorak obogaćen selenom iz neorganskog izvora. Uzorci su pripremljeni ekstrakcijom plodonosnih tela u zagrejanoj vodi. Dobijeni rezultati ukazuju na to da vodeni ekstrakti celih gljiva, kako kontrolnih tako i sa dodatkom selena, imaju dobru antioksidativnu aktivnost. Međutim, nije primećena značajna razlika između uzoraka sa i bez sadržaja selena.

AB  - Genoderma lucidum among all other cultivated mushrooms is unique for its medicinal, rather than nutritional value. A woody mushroom for centuries highly regarded in oriental folk medicine in recent years came in to focus of numerous pharmacological and medical researchers. The paper gives a brief overview of the obtained results and state of the art of knowledge about its bioactive components and pharmacological functioning indicating its possible use in brewing as a raw material for the production of beer with improved functional properties. It deals with the procedure for Ganoderma extract production, determination of main bioactive substance contents, procedure for beer enrichment and sensory evaluation of the final products. The consumer's acceptance test was carried out by 105 untrained young subjects. The results indicate that both male and female tasters evaluated the enriched beer similar or even better than initial commercially produced Pilsner beer. Male tasters especially showed great affinity for new sensory properties, particularly the body, liveliness, and taste. Female tasters evaluated both beers quite similarly. According to their opinion, beer with Ganoderma compared with initial beer differs statistically significantly better only by its bodiness.

AB  - The paper highlights the facts about the possibilities of improving the functionality of beer with extracts of thyme (Thymus vulgaris), lemon balm (Melissa officinalis) and mushroom Ganoderma lucidum. It briefly summarizes the most important data about possible positive action of moderate beer consumption and the benefits of beer as a base for developing a variety of products with enhanced functionality. It gives an overview about the mentioned herbs and the mushroom, their use in traditional medicine, chemical composition, pharmacodynamic properties and possible benefits from the brewing point of view. Procedures for extraction of biological material, experimental results of antimicrobial properties, antioxidant capacity and sensory evaluation of beer enriched with these extracts are given. Experimental results indicate that commercially produced and bottled pils beer enriched with tinctures of Thymus vulgaris and Melissa officinalis shows improved antimicrobial and antioxidative properties. Ganoderma is particularly important because of its unique functional properties and sensory compatibility with beer. Products obtained like this could fulfill several goals: developing novel beer types, developing products with health-promoting properties that meet market needs and eventually gain new beer consumers. Alcohol content of such products depends on the type and alcohol content of initial beer.

Medicinal mushroom Ganoderma lucidum in the production of special beer types

AB  - Mushrooms like Ganoderma lucidum have been used for thousands of years as a traditional medicine in the Far East. Ganoderma received wide popularity as an eating mushroom with high nutritive value, but even more as medical fungi. It has been used for the treatment of various diseases: hepatitis, hypertension, insomnia, and even cancer. Due to its extraordinary action, it is often called 'Elixir of life', 'Food of gods' and 'Mushroom of universe'. The intracellular and extracellular polysaccharides (b-glucane) inhibit the growth of several types of cancer. Mushroom produces triterpenes of which especially ganoderic acid showed cytotoxicity on primary tumor liver cells, inhibition of histamine release, hepatoprotective effect, stimulation of the immune system functions, inhibition of the aggregation of blood plates, etc. On the other hand, beer as a purely natural beverage obtained in the process of fermentation, contains a number of ingredients which are important for human organism, and in moderate usage has favorable reaction on the general health condition of the body. As such, beer is a very good basis for the development of a number of new products with defined pharmacodynamics influence. In this work, we have investigated the possibilities of using extracts of mushroom Ganoderma lucidum in the production of special beer types. The composition of mushroom, properties of the most important active ingredients, extraction procedures, and sensory characteristics of the beers on the basis of such extracts were determined. The most important parameters of quality and possibility of adjustments using extracts of different medicinal herbs were investigated.
AB  - Gljive poput Ganoderma lucidum korišćene su hiljadama godina u tradicionalnoj narodnoj medicini Dalekog istoka. Svoju veliku popularnost Ganoderma je stekla kao jestiva gljiva sa visokom nutritivnom vrednošću, ali još više kao medicinska gljiva. Korišćena je za lečenje raznih bolesti, između ostalih i hepatitisa, hipertenzije, insomnije, pa i karcinoma. Upravo zbog svog izvanrednog delovanja nazivana je 'Eliksir života', 'Hrana bogova', 'Gljiva univerzuma'. Njeni intracelularni i ekstracelularni polisaharidi (b-glukani) dokazano inhibiraju rast nekoliko vrsta karcinoma, a triterpeni koje produkuje, posebno ganoderinska kiselina, pokazuju citotoksičnost na primarne tumorne ćelije jetre, inhibiciju oslobađanja histamina, hepatoprotektivni efekat, stimulaciju funkcije imunog sistema, inhibiciju agregacije krvnih pločica itd. Sa druge strane, pivo kao potpuno prirodno piće dobijeno fermentacijom, sadrži mnogobrojne sastojke od značaja za organizam i u umerenoj količini pokazuje povoljno delovanje na opšte zdravstveno stanje organizma. Kao takvo, pivo je veoma dobra podloga za razvoj niza novih proizvoda definisanog farmakodinamičkog delovanja. U radu je ispitana mogućnost korišćenja ekstrakata gljive Ganoderma lucidum u proizvodnji specijalnih piva. Dati su sastav gljive, svojstva najvažnijih aktivnih sastojaka, postupci ekstrakcije i senzorne karakteristike piva dobijenih na bazi datih ekstrakata. Ispitani su najvažniji parametri kvaliteta i mogućnost korigovanja upotrebom ekstrakata različitog lekovitog bilja.

Njeni intracelularni i ekstracelularni polisaharidi (b-glukani) dokazano inhibiraju rast nekoliko vrsta karcinoma, a triterpeni koje produkuje, posebno ganoderinska kiselina, pokazuju citotoksičnost na primarne tumorne ćelije jetre, inhibiciju oslobađanja histamina, hepatoprotektivni efekat, stimulaciju funkcije imunog sistema, inhibiciju agregacije krvnih pločica itd. Sa druge strane, pivo kao potpuno prirodno piće dobijeno fermentacijom, sadrži mnogobrojne sastojke od značaja za organizam i u umerenoj količini pokazuje povoljno delovanje na opšte zdravstveno stanje organizma. Kao takvo, pivo je veoma dobra podloga za razvoj niza novih proizvoda definisanog farmakodinamičkog delovanja. U radu je ispitana mogućnost korišćenja ekstrakata gljive Ganoderma lucidum u proizvodnji specijalnih piva. Dati su sastav gljive, svojstva najvažnijih aktivnih sastojaka, postupci ekstrakcije i senzorne karakteristike piva dobijenih na bazi datih ekstrakata. Ispitani su najvažniji parametri kvaliteta i mogućnost korigovanja upotrebom ekstrakata različitog lekovitog bilja.",

AB  - Selenium (Se) is a trace mineral that is essential to good health but only in small quantities. Soils, grains and forage have very low amounts of selenium. In most countries, selenium content in soil ranges between 0,1 and 0,2 ppm. There are many ways for selenium supplementation but it is considered that the best way is to increase selenium content in the plants. Mushrooms contain relatively high protein levels and they can accumulate large amounts of selenium. It is reasonable to expect that Se could be incorporated into the proteins as selenomethionine and selenocysteine. Lentinus edodes (Shiitake) is an edible mushroom native to East Asia, which is cultivated and consumed in many Asian countries for over 2,000 years. Shiitake Mushroom is revered for its health-promoting activity; it boosts the immune system, lowers cholesterol, works as an anticoagulant and is helpful in cancer treatment. Bioaccumulation of selenium in industrial mushroom was investigated. Vegetative growth of mycelium were measured as colony diam in pure cultures supplemented with organic selenium from new synthetic compound Zn(II) complex with ligand 2,6-diacetylpyridines bis(selenosemicarbazone), (Zn(dapsesc)). Accumulation of selenium in the fruit body was determined by the use of ICP-OES (Inductively coupled plasma optical emission spectrometry). Selenium as a complex, was effectively taken up from substrates and accumulated in fruit bodies. Highest selenium content was in the cap (60 ppm). In mushrooms cultivated without selenium supplement, selenium content was approximately 1 ppm. .
AB  - Selen je esencijalni mikroelement, neophodan za normalno funkcionisanje organizma ljudi i životinja. Rezultati prvih istraživanja o obezbeđenosti zemljišta, žitarica i stočne hrane ukazali su na vrlo nizak nivo selena. U većini zemljišta sveta sadržaj ukupnog selena je u granicama od 0,1 do 2 ppm. Postoji više mogućnosti suplementacije selena (selenizovana kuhinjska so, pelete, inekcije), ali se smatra da je najbolji način povećanjem sadržaja selena u biljkama. S obzirom na to da gljive sadrže visok procenat proteina, očekuje se da će apsorbovati i inkorporirati selen u proteinima, u obliku kao što su selenometionin i selenocistein. Gljiva shiitake (Lentinus edodes) vodi poreklo iz Istočne Azije - Kina, Japan, a prvi pisani podaci govore da je poznata više od 2000 godina. Pregled literature ukazuje da ova gljiva smanjuje nivo holesterola, jača imunitet, ima antikoangulantnu ulogu i da se koristi u prevenciji nastanka tumora. Praćen je uticaj novosintetisanog organskog kompleksa Zn (II) sa ligandom 2,6-diacetilpiridin bis-(selenosemicarbazon) (H2dapsesc) - (Zn (dapsesc)) sa Hemijskog fakulteta Univerziteta u Beogradu na porast micelijuma gljiva i mogućnost akumulacije selena u plodonosnim telima. Ukupan sadržaj selena je određivan optičkim emisionim spektrometrom sa indukovano spregnutom plazmom, ICP-OES. Dobijeni rezultati govore da je selen iz organskog izvora uspešno usvojen. Sadržaj selena je bio očekivano najveći u šeširu, 60 µg/g suve mase. U kontrolnim pečurkama, ova vrednost se kretala u proseku oko 0.4 µg/g suve mase. .

AB  - Mushrooms are a popular and valuable functional food, low in calories and high in minerals, essential amino acids, vitamins and fibers. More than 270 mushroom species are high valuable nutritive food, but they also have different pharmacologicaly effects, what classify them in the group of the medicinal mushrooms. These mushrooms show immunomodulating and anticancerous properties, they act like antioxidants and antihypertensive substances, have ability to lower cholesterol and protect the liver, possess antiinflammatory, antidiabetic, antiviral and antimicrobial properties. Medicinal mushrooms are very reach sources of new biomedicinal molecules which can be used in treating different disorders in human organism. Among these biomolecules the most important are protein bounded polysaccharides which are made by basidiomycetes; thanks to their extraordinary properties these biomolecules are classified like antitumorous substances by American National Cancer Institute. .
AB  - Pečurke su popularna i vredna funkcionalna hrana, zahvaljujući niskom sadržaju kalorija i sa druge strane visokom sadržaju minerala, esencijalnih amino kiselina, vitamina i vlakana. Pored nutritivnog aspekta, dokazano je da više od 270 vrsta gljiva ima i različita farmakološka dejstva, što ih svrstava u grupu lekovitih gljiva. Ove gljive pokazuju imunomodulatorska i antikancerogena svojstva, deluju kao antioksidanti, antihipertenzivne supstance, snižavaju holesterol, štite jetru, pokazuju antiinflamatorna, antidijabetska, antivirusna i antimikrobna svojstva. Lekovite gljive su bogat izvor niza novih biomedicinskih molekula koji mogu da se koriste u tretiranju različitih poremećaja u ljudskom organizmu. Među ovim biomolekulima najznačajniji su polisaharidi vezani za proteine, koje stvaraju bazidiomicete, a zbog izuzetnih svojstava koje imaju Američki nacionalni institut za kancer klasifikovao ih je kao antitumorne supstance. .
