AB  - The aim of this study was to compile a detailed phytochemical profile and assess the antioxidant properties of bee-collected pollen (PBP) obtained from corn poppy (Papaver rhoeas L.) plants. To achieve this, a lipid fraction was prepared for quantifying fatty acids using GC-FID. Extractable and alkaline-hydrolysable PBP fractions (obtained from a defatted sample) were used to determine the qualitative and quantitative profiles of phenolic compounds, phenylamides and alkaloids using UHPLC/Q-ToF-MS. Additionally, various spectrophotometric assays (TAC, FRP, CUPRAC, DPPH⦁) were conducted to evaluate the antioxidant properties. Phenolic compounds were more present in the extractable fraction than in the alkaline-hydrolysable fraction. Luteolin was the predominant compound in the extractable fraction, followed by tricetin and various derivatives of kaempferol. This study presents one of the first reports on the quantification of tricetin aglycone outside the Myrtaceae plant family. The alkaline-hydrolysable fraction exhibited a different phenolic profile, with a significantly lower amount of phenolics. Kaempferol/derivatives, specific compounds like ferulic and 5-carboxyvanillic acids, and (epi)catechin 3-O-gallate were the predominant compounds in this fraction. Regarding phenylamides, the extractable fraction demonstrated a diverse range of these bioactive compounds, with a notable abundance of different spermine derivatives. In contrast, the hydrolysable fraction contained six spermine derivatives and one spermidine derivative. The examined fractions also revealed the presence of seventeen different alkaloids, belonging to the benzylisoquinoline, berberine and isoquinoline classes. The fatty-acid profile confirmed the prevalence of unsaturated fatty acids. Furthermore, both fractions exhibited significant antioxidant activity, with the extractable fraction showing particularly high activity. Among the assays conducted, the CUPRAC assay highlighted the exceptional ability of PBP’s bioactive compounds to reduce cupric ions.

The Morpho-Anatomy of Nectaries and Chemical Composition of Nectar in Pear Cultivars with Different Susceptibility to Erwinia amlylovora

AB  - The topography and morpho-anatomical characteristics of floral nectaries and the chemical analysis of nectar have been studied in seven pear cultivars with different susceptibility to Erwinia amylovora. The susceptible cultivar Williams, the moderately resistant cultivars Bella di guigno, Poire de Cure and the low susceptible cultivar Alexander Lucas originated from Pyrus communis, while highly resistant cultivars Chojuro and Nijisseiki from P. pyraster and Kieffer as interspecies hybrid P. communis × P. pyraster were included in this experiment and studied for the first time. Large differences in size and structure of the nectaries were observed in these seven pear cultivars. The epidermal cells were with cuticle being more or less striated in Alexander Lucas, Kieffer and Williams. Resistant cultivars had a narrow, elongated cell shape of epidermal cells while those susceptible had an isodiametric. Stomata were mesomorphic in all cultivars except in Poire de Cure and Williams, being slightly xeromorphic since they were situated in deep hollows. Guard cells of the modified stomata were much larger in resistant cultivars. Hypanthium cells were larger in resistant compared to susceptible cultivars. The most abundant sugars were glucose, fructose, sorbitol and sucrose in nectar of all studied pear cultivars. The resistant cultivars (Chojuro, Kieffer and Nijisseiki) had a ~2-fold higher level of sorbitol and galactose, ~2.2-fold higher isomaltose, ~2.7-fold turanose, ~3.35-fold maltose, ~4.4-fold melibiose and ~12.7-fold higher melesitose compared to susceptible cultivars. The sum of quantified phenolic acids varied from 0.049 (Williams) up to 4.074 µg CAE/mL (Kieffer), while flavonoid glycosides levels ranged from 1.224 (Williams) up to 11.686 µg RE/mL (Nijisseiki). In the nectar of the resistant cultivars, rutin, apigetrin, together with patuletin and luteolin glycosides were detected but not in susceptible cultivars, which could be considered as the markers of resistance. © 2023 by the authors.

AB  - Varroa destructor is a parasitic mite responsible for the loss of honey bee (Apis mellifera) colonies. This study aimed to find a promising marker in honey for the bee colony infestation level using fluorescence spectroscopy and biochemical analyses. We examined whether the parameters of the honey samples’ fluorescence spectra and biochemical parameters, both related to proteins and phenolics, may be connected with the level of honey bee colonies’ infestation. The infestation level was highly positively correlated with the catalase activity in honey (r = 0.936). Additionally, the infestation level was positively correlated with the phenolic spectral component (r = 0.656), which was tentatively related to the phenolics in honey. No correlation was found between the diastase activity in honey and the colonies’ infestation level. The results indicate that the catalase activity in honey and the PFC1 spectral component may be reliable markers for the V. destructor infestation level of the colonies. The obtained data may be related to the honey yield obtained from the apiaries. © 2023 by the authors.

Micro/trace/toxic elements and insecticide residues level in monofloral bee-collected sunflower pollen- health risk assessment

None of the twenty four examined insecticides were detected above the limit of detection (LOD) which makes studied sunflower bee-collected pollen eco-friendly both to bees and humans. Based on presence of several toxic as well as potentially toxic elements calculations for estimated weekly intakes (EWI), and oral intakes (OI) were made and used for health risk assessment based on the computation of two different health risk quotients (HQ)- acute (HQA) and long-term (HQL). The obtained results proved that all HQ values for adults were negligible or low except in case of HQL value for arsenic (0.32) which can be characterized as medium. However, in case of children much more precaution is needed due to significant HQL risk for arsenic (1.511). The attained data can help to make additional linkage between bee-collected pollen as food ingredients and potential benefits/risks for human health.

Sophora flower corresponds to the pharmacopoeia requirements for the quality of an herbal drug. The concentrations of rutin and quercetin in acacia honey were 0.08 and 0.14 mg/kg, respectively. AB  - The current study intended to determine, for the first time, phenolic and fatty acid profile, antioxidant and certain nutritional properties of monofloral bee-collected artichoke (Cynara scolymus) pollen. Based on UHPLC-DAD MS-MS analysis the main phenolics in extractable fraction were different flavonol glycosides (in particular Isorhamnetin-3-O-glucoside, 49.2 mg/kg of dry weight) while ferulic acid was the predominant phenolic compound (39.4 mg/kg of dry weight) in the alkaline hydrolyzable fraction. Among fatty acids (FAs), results of GC-FID analysis revealed prevalence of unsaturated FAs with cis-5,8,11,14,17-eicosapentaenoic acid (EPA) and oleic acid as the main ones-28.4% and 24.9%, respectively. Based on the FA composition, nutritional analysis proved that artichoke bee-collected pollen had balanced ω-6 and ω-3 FAs content. To determine the antioxidant properties of pollen, five different assays were applied. It was proved that bioactive compounds in artichoke pollen possessed significant ability to quench DPPH radical as well as ABTS radical cation. In addition, in vitro phosphomolybdenum assay confirmed that artichoke pollen is an excellent source of different antioxidants. Pollen extracts exhibited moderate ferric reducing power as well as low ferrous chelating ability. Some further antioxidant studies (preferably in vivo) should be performed to confirm the observed results.

AB  - The aim of this study was to provide information on the phenolic and sugar profiles of different parts of the buckwheat plant, which can define that buckwheat is a functional food, with a high nutritional value and very useful for human health. Therefore, the extracts of buckwheat leaf, stem, and flower, as well as buckwheat grain were analysed for the content of polyphenol and antioxidant tests. The identification of a notable number of phenolic compounds and quantification of sugars in different parts of buckwheat indicates that buckwheat is a highly valuable plant. A total of 60 phenolic compounds were identified (18 cinnamic acid derivatives, 14 flavonols, 13 flavan-3-ols (including proanthocyanidins), 10 hydroxybenzoic acid derivatives, and 5 flavones) using ultra-high-performance liquid chromatography (UHPLC), coupled with a hybrid mass spectrometer which combines the Linear Trap Quadrupole (LTQ) and OrbiTrap mass analyzer. The highest number of phenolic compounds was found in the analysed buckwheat flower sample, and then in the leaf, followed by the grain and the stem. In addition, the sugar profile of buckwheat leaf, stem, flower and grain, as well as the buckwheat pollen and the nectar was analysed. Hence, 16 sugars and 5 sugar alcohols were detected by the high-performance anion exchange chromatography (HPAEC) with a pulsed amperometric detector (PAD). Sucrose was found in a significant amount with the highest content in buckwheat leaf. Trisaccharides had similar accumulation in the sample extracts, while disaccharides dominated in buckwheat leaf, followed by nectar and pollen. The sugar alcohols showed the highest content in buckwheat grain, where erythritol was predominant. The obtained results show that buckwheat is very rich in phenolic compounds and sugars. In addition to grain, the other parts of the buckwheat plant can be used as a very good source of different classes of phenolic compounds. This study provides useful information on the distribution of phytochemicals in different parts of the buckwheat plant, which contribute to the maintaining of the status of buckwheat as a functional food.

AB  - The aim of this study was to examine nutritional (total lipid, carbohydrate, protein and ash content, fatty acids and protein profile) and techno-functional properties of monofloral bee-collected sunflower pollen (Helianthus annuus L.). The content of water, total protein, carbohydrate, lipid and ash was 24.99, 14.36, 82.01, 1.62 and 2.01 g/100g DW, respectively, with the total energy value of 400.06 kcal/100 g DW. The fatty acid profile revealed the presence of five fatty acids with the stearic acid as dominant one (31.4%) followed by α-linoleic (20.7%), pentadecanoic (18.2%), heneicosanoic (17.1%) and palmitic (12.5%) acids. FTIR analysis of bee pollen grains confirmed the presence of the main pollen chemical constituents such as proteins, water, carbohydrates and lipids, but also the presence of sporopollenin and polyphenols. Raman spectroscopy analysis indicated that the surface of pollen grains was rich in carotenoids. Low protein (3.64 g/100g DW) but high carbohydrate (77.09 g/100 g DW) solubility of bee pollen was observed. The good emulsifying properties (ESI, 19.98 min; EAI, 80.54 m2/g) and excellent oil (2.43 g/g DW) but poor water (0.87 g/g DW) absorption capacities were also registered. On the other hand, the sunflower bee pollen did not show foaming properties. These findings indicate to its applicability as useful nutritional, lipophilic and anti-foaming food ingredients.

AB  - Pollen (floral or bee-collected) can be used as a good source of different nutrients including several important biogenic elements (minerals). The aim of this study was to determine content of selected biogenic elements (phosphorous, potassium, sulphur, calcium, magnesium, iron and zinc) in monofloral bee-collected pollen originated from sunflower (Helianthus annuus L.) plants grown in Serbia and to estimate its nutritional value regarding to elements content. In that purpose ICP-OES analytical method was applied. According to obtained results the examined pollen sample contains above mentioned elements in following quantities: P – 3523.98 mg/kg; K – 2869.13 mg/kg; S – 1951.74 mg/kg; Ca – 1919.20 mg/kg; Mg – 654.36 mg/kg; Fe –83.55 mg/kg; Zn – 45.92 mg/kg of dry weight. Based on recommended daily intakes for iron, phosphorus and zinc application of pollen, as food or food ingredient, will cover meaningful part of human needs for these three important elements.

AB  - Most Varroa induced colony losses occur during the autumn or winter season as a consequence of an elevated Varroa infestation level and an insufficient health status of the adult bees. Even with an initial low Varroa infestation in early spring, critical mite and virus infection levels can be reached before winter if colonies continuously rear brood throughout the whole season. To overcome this challenge, beekeepers can artificially interrupt brood production by suitable management procedures, depending on their type of beekeeping operation. To assess their efficacy, associated workload, and impact on colony development, different methods for brood interruption (queen caging with the combination of oxalic acid treatment, total brood removal, trapping comb technique) were tested during two seasons in 11 locations on 370 colonies in 10 European countries. A protocol was developed to standardize the methods' application across different environmental conditions. The efficacy of queen caging depended on the mode of oxalic acid application and ranged from 48.16% to 89.57% mite removal. The highest efficacies were achieved with trickling a 4.2% solution (89.57%) and with the sublimation of 2 g of oxalic acid (average of 88.25%) in the broodless period. The efficacy of the purely biotechnical, chemical-free trapping comb and brood removal methods did not differ significantly from the queen caging groups. We conclude that a proper application of one of the described brood interruption methods can significantly contribute to an efficient Varroa control and to the production of honey bee products meeting the highest quality and food-safety standards.

AB  - The aim of this work was to determine phenolic profile and antioxidant properties of methanolic (MEP) and ethanolic (EEP) extracts of monofloral bee-collected sunflower pollen from Serbia. In total, 37 different polyphenols (19 flavonol glycosides and 18 other polyphenols) were identified by UHPLC/MS-MS Orbitrap analysis. The different phenolic profile of MEP and EEP was observed with flavonols as most abundant, 94.1% (MEP) and 93.4% (EEP), out of which flavonoid glycosides represented 73.2% (MEP) and 66.1% (EEP). The higher amount of total phenolics was recorded in EEP (200.58 mg/kg DW) than in MEP (244.44 mg/kg DW). The quercetin 3-O-galactoside was the major compound in both extracts, 112.86 mg/kg in MEP and 128.64 mg/kg in EEP. The presence of coumaroylated and acetylated flavonoid glycosides were registered, among which the quercetin 3-O-(6 ''-acetyl)hexoside was recored in the significant amount in MEP (111.10 mg Q3G/kg DW), whereas in EEP the content of coumaroylated kaempferol glycosides (137.95 mg Q3G/kg DW) was significantly higher. The ferric reducing power and ABTS radical scavenging activity assays revealed that pollen extracts possessed significant antioxidant activity. The monofloral bee-collected pollen of Helianthus annuus L. can be used as good food supplement with high flavonoids content and antioxidant activity.

Clostridium botulinum spores in European honey bees from Serbia

The microbiological methods and molecular methods (multiplex PCR/mPCR and PCR method) were utilized to examine multiple subunits of each honey bees samples. The C. botulinum spores in PCR-positive samples were estimated by the most probable number method (MPN). The presence of C. botulinum spores, by applying mPCR and PCR methods, was detected in 1 of the 61 honey bees (1.64%). Using MPN method, the number of spores in positive sample was 110/kg. Detection of C. botulinum spores directly from untreated honey bees, without prior enrichment, is impossible by applying PCR. Using conventional microbiological methods, detection of C. botulinum spores in dead honey bees is not possible without preenrichment. Therefore, conventional, microbiological methods are not suitable for the detection of C. botulinum spores in honey bees. In order to detect C. botulinum spores in honey bees using PCR methods, due to the small and/or unequal distribution of spores in the samples, it is desirable to use multiple subunits/replicates for each sample examined.

Economic justification of honey production in Serbia

AB  - Although beekeeping in Serbia has a long tradition, the interest of farmers for this branch of agricultural production depends on the expected economic benefits. Assessing the economic justification of beekeeping is based on the analysis of the revenues from honey production, as the main and often the only beekeeping product, and the expenses generated in production. The aim of the paper is to compare revenues and expenses in the production of honey and to determine the threshold of profitability. The calculation of the revenues and the expenses was made on the basis of data obtained from the survey of beekeepers and from the available secondary sources. The analysis showed that the profitability threshold was achieved with 68 colonies, or with production volume of 1,450 kg of honey. The values of the indicators of economic efficiency, productivity and profitability indicate that beekeeping was economically justified on farms with 100 and 200 colonies.
AB  - Iako pčelarstvo u Srbiji ima dugu tradiciju, zainteresovanost poljoprivrednika za ovu granu poljoprivredne proizvodnje zavisi od ekonomskih koristi koje mogu da očekuju. Sagledavanje prihoda od proizvodnje meda, kao glavnog a često i jedinog proizvoda pčelarstva i troškova koji nastaju u proizvodnji predstavlja osnovu za ocenu ekonomske opravdanosti ove delatnosti. Cilj rada je da se izvrši poređenje prihoda i troškova u proizvodnji meda, kao i da se utvrdi prag rentabilnosti proizvodnje. Kalkulacija prihoda i troškova je sačinjena na osnovu podataka koji su prikupljeni anketiranjem pčelara i iz dostupnih sekundarnih izvora. Sprovedena analiza ukazuje da se prag rentabilnosti ostvaruje sa 68 pčelinjih društava, odnosno pri obimu proizvodnje od 1.450 kg meda. Vrednosti pokazatelja ekonomičnosti, produktivnosti i rentabilnosti ukazuju da je pčelarenje ekonomski opravdano na gazdinstvima sa 100 i 200 pčelinjih društava.

abstract = "Although beekeeping in Serbia has a long tradition, the interest of farmers for this branch of agricultural production depends on the expected economic benefits. Assessing the economic justification of beekeeping is based on the analysis of the revenues from honey production, as the main and often the only beekeeping product, and the expenses generated in production. The aim of the paper is to compare revenues and expenses in the production of honey and to determine the threshold of profitability. The calculation of the revenues and the expenses was made on the basis of data obtained from the survey of beekeepers and from the available secondary sources. The analysis showed that the profitability threshold was achieved with 68 colonies, or with production volume of 1,450 kg of honey. The values of the indicators of economic efficiency, productivity and profitability indicate that beekeeping was economically justified on farms with 100 and 200 colonies., Iako pčelarstvo u Srbiji ima dugu tradiciju, zainteresovanost poljoprivrednika za ovu granu poljoprivredne proizvodnje zavisi od ekonomskih koristi koje mogu da očekuju. Sagledavanje prihoda od proizvodnje meda, kao glavnog a često i jedinog proizvoda pčelarstva i troškova koji nastaju u proizvodnji predstavlja osnovu za ocenu ekonomske opravdanosti ove delatnosti. Cilj rada je da se izvrši poređenje prihoda i troškova u proizvodnji meda, kao i da se utvrdi prag rentabilnosti proizvodnje. Kalkulacija prihoda i troškova je sačinjena na osnovu podataka koji su prikupljeni anketiranjem pčelara i iz dostupnih sekundarnih izvora. Sprovedena analiza ukazuje da se prag rentabilnosti ostvaruje sa 68 pčelinjih društava, odnosno pri obimu proizvodnje od 1.450 kg meda. Vrednosti pokazatelja ekonomičnosti, produktivnosti i rentabilnosti ukazuju da je pčelarenje ekonomski opravdano na gazdinstvima sa 100 i 200 pčelinjih društava.",

AB  - Honey is a natural substance produced by honey bees (the genus Apis) enjoyed by people due to its unique nutritional and medicinal properties. The aim of this study was to determine the physicochemical parameters (moisture, ash, water-insoluble content, reducing sugars, sucrose, five acidity, diastase activity, hydroxymethylfurfural, and electrical conductivity) and microbiological status (total number of aerobic mesophilic bacteria, total number of sulfite-reducing clostridia, the presence of Salmonella spp., total numbers of fungi and yeasts and the presence of Clostridium botulinum) in honey (honeydew, blossom, sunflower, acacia, and linden) produced in an urban environment in Serbia. We analyzed 19 apiary samples of honey, collected during the 2011 harvesting season, by using recommendation methods. Physicochemical parameters of the examined honey produced in the urban environment indicated the honeys were of acceptable quality. Bacillus spp. were detected in four honeys, yeasts were detected in three honeys, and Clostridium botulinum type E was detected in one honey using PCR. The current study also showed the presence of diverse honey varieties in Serbia.

AB  - To contribute to the development of urban beekeeping, we designed this study to obtain more information about the contamination of urban bee products with toxic metals, polycyclic aromatic hydrocarbons (PAHs), and pesticides. The samples of honey (N=23), pollen (N=13), and floral nectar (N=6) were collected from the experimental stationary apiary of the Belgrade University Faculty of Agriculture located in centre of Zemun (a municipality of the Belgrade metropolitan area) in 2015 and 2016. Metals (Pb, Cd, As, Cu, Zn, Fe, Mn, Ni, Cr, and Hg) were determined with inductively coupled plasma quadrupole mass spectrometry (ICP-QMS). Polycyclic aromatic hydrocarbons (PAHs) were analysed with high-performance liquid chromatography with fluorescence detection (HPLC-FLD). Pesticides were analysed with gas chromatography - mass spectrometry (GC-MS). The honey samples were generally within the European and Serbian regulatory limits. The levels of all the 123 analysed pesticides were below the limit of quantification (LOQ). Regarding PAH levels in honey, the highest content was found for naphthalene. The elevated levels of Hg and Cr and of PAHs in the pollen samples indicated air pollution. Pesticide residues in pollen, however, were below the LOQ. In nectar, metal levels were relatively similar to those in honey. Our results suggest that the investigated urban honey meets the regulatory requirements for metals, PAHs, and pesticides and is therefore safe for consumption.

AB  - Assessment of microbiological quality of bee collected pollen is very important, because of its use as a supplement in the human diet. In this study, 26 samples collected from different location in Serbia were tested for the presence of mold through mycologial analysis. The presence of aflatoxin B1, one of the most dangerous and the most widespread mycotoxin was also determined. It was established that 10 of the investigated samples were contaminated with some genera or species of mold, but all of the investigated samples were contaminated with aflatoxin B1. Considering that there is no unique and official procedure for mycological analysis of bee collected pollen, these findings suggest the need for their establishment. Mycological analysis should be followed by mycotoxicological analysis since the absence of mold does not confirm the absence of aflatoxin B1 in bee pollen.

AB  - Oblacinska' sour cherry, an autochthonous cultivar, is the most planted cultivar in Serbian orchards. Since fruit trees in temperate zone reward insects by producing nectar which 'quality' affects the efficiency of insect pollination, the aim of this study was analyzing of sugars and polyphenolics in floral nectar of 16 'Oblacinska' sour cherry clones with different yielding potential. The contents of sugars and sugar alcohols were analyzed by ion chromatography, while polyphenolic profile was established using liquid chromatography/mass spectrometry technique. Fourteen sugars and six sugar alcohols were detected in nectar samples and the most abundant were fructose, glucose, and sucrose. Eleven polyphenols were quantified using available standards, while another 17 were identified according to their exact masses and characteristic fragmentations. Among quantified polyphenols, rutin, naringenin, and chrysin were the most abundant in nectar. Principal component analysis showed that some polyphenol components (naringin, naringenin, and rutin) together with sugars had high impact of spatial distribution of nectar samples on score plot.

Impact of Year and Type of Feed on Winter Honeybees' Feed Consumption

AB  - Winter honeybees intake minimal amounts of food, only for maintenance needs. This study aims to determine the effect of year of research and the different types of food on the amount of consumed food. The survey found a statistically significant difference between mean values of food consumption by years of research, as well as mean values of consumption of different types of food. According to our results, it can be concluded that winter honeybees need easily acceptable energy for maintenance needs, so supplemental feeding during preparation for winter should be consisted only of invert syrup, without additon of protein or other nutrients.
AB  - Zimske pčele konzumiraju minimalne količine hrane, održavajući svoj metabolizam. Cilj istraživanja je da se utvrdi uticaj dva faktora - godina istraživanja i različite vrste hrane na količinu konzumirane hrane. Istraživanjem je utvrđeno da postoji statistički visoko značajna razlika između srednjih vrijednosti potrošnje hrane po godinama istraživanja, kao i srednjih vrijednosti potrošnje različitih vrsta hrane. Dobijeni rezultati istraživanja nas navode na zaključak da je u periodu pripreme pčela za zimu potrebno pčele prihranjivati invertovanom hranom, bez dodataka proteina i drugih hranjivih materija, jer je za kvalitetno prezimljavanje pčelama potrebna lako pristupačna energija.

The survey found a statistically significant difference between mean values of food consumption by years of research, as well as mean values of consumption of different types of food. According to our results, it can be concluded that winter honeybees need easily acceptable energy for maintenance needs, so supplemental feeding during preparation for winter should be consisted only of invert syrup, without additon of protein or other nutrients., Zimske pčele konzumiraju minimalne količine hrane, održavajući svoj metabolizam. Cilj istraživanja je da se utvrdi uticaj dva faktora - godina istraživanja i različite vrste hrane na količinu konzumirane hrane. Istraživanjem je utvrđeno da postoji statistički visoko značajna razlika između srednjih vrijednosti potrošnje hrane po godinama istraživanja, kao i srednjih vrijednosti potrošnje različitih vrsta hrane. Dobijeni rezultati istraživanja nas navode na zaključak da je u periodu pripreme pčela za zimu potrebno pčele prihranjivati invertovanom hranom, bez dodataka proteina i drugih hranjivih materija, jer je za kvalitetno prezimljavanje pčelama potrebna lako pristupačna energija.",

AB  - The presence of Clostridium botulinum spores in 59 honey samples originating from different regions of the Republic of Serbia was studied. In addition to microbiological methods, after enrichment, centrifugation and membrane filtration, molecular methods (PCR methods) were utilized. The number of spores in PCR positive samples was estimated by the most probable number (MPN) method. PCR confirmed C. botulinum spores in 5 (8.47%) honey samples. MPN of spores varied from 20/kg to 204/kg honey. PCR was more sensitive than cultural methods. Natural honey contamination with C. botulinum spores is low-level and not homogeneous, and therefore, PCR methods require multiple sub-sampling.

AB  - IntroductionPropolis is a resinous natural substance collected by honeybees from different plant sources. Due to the presence of various phytochemicals, this bee-product exhibits numerous biological activities, including anti-bacterial, anti-viral, anti-inflammatory, anti-oxidant, immunostimulating and anti-tumour effects. As the chemical composition and biological activity of propolis depend on its botanical and geographical origin, searching for new bioactive substances in various types of propolis from unexplored regions is of great importance. ObjectiveThe aim of this study is the evaluation of the phenolic profile of poplar propolis samples in order to characterise Serbian propolis, to identify possible new constituents and to specify the phenolic components relevant for differentiation of poplar propolis samples into two subgroups through simultaneous analysis of poplar bud extracts. MethodsEthanolic extracts of propolis and poplar buds were comprehensively analysed using ultrahigh-performance liquid chromatography coupled with hybrid mass spectrometry, which combines the linear trap quadrupole and Orbitrap MS/MS mass analyser together with chemometric methods. ResultsExtensive fingerprint analysis of Serbian propolis was achieved for the first time. Seventy-five phenolic compounds were detected. Eight of them were identified in propolis for the first time. Pattern-recognition methods applied to the content of ten quantified phenolics verified the existence of two subgroups of propolis, with galangin, chrysin and pinocembrin as the most influential distinguishing factors. ConclusionThe phenolic composition of the analysed propolis samples confirm their affiliation to the European poplar type propolis and the existence of two subgroups according to botanical origin.

AB  - Cold stress adversely affects growth and productivity, and triggers a series of morphological, physiological, biochemical and molecular changes in plants. Since sugars are present in all floral nectars in greater amounts than any other constituent, the aim of this study was to examine how frost exposure changes sugar metabolism and how it affects on the content of sugar components in the nectar of quince. Three quince cultivars ('Vranjska', 'Triumph' and 'Leskovac. ka') were investigated in this study. The contents of sugars (glucose, fructose, sucrose, trehalose, maltose, isomaltose, rhamnose, arabinose, ribose, melezitose, raffinose, and panose) and sugar alcohols (sorbitol, erythritol, mannitol and galactitol) were analyzed by high performance anion exchange chromatography (HPAEC) with amperometric detection. The results showed that after late spring frosts and irreversible damage of flower parts, the nectar of the three quince cultivars contained elevated levels of fructose, trehalose, arabinose, ribose, rhamnose, raffinose, galactitol and mannitol, indicating an impairment of central carbohydrate metabolism. The ratios between individual sugars, such as the glucose/fructose ratio, were changed in the nectar of damaged flowers in all three quince cultivars. The examined cultivars showed similar sugar response to cold stress. The only exception was 'Leskovac. ka' for the glucose and melezitose pathway, which means that composition of those two sugars changed significantly according to the genotype. The larger are the carbohydrates reserves in different parts of a fruit tree, the higher is the tolerance to any form of frost damage, the results of this study could help in the understanding of how different quince cultivars react to this kind of stress and how they modulate their sugar metabolism.

Winter bees are unable to reach the natural water sources, even their need for water are minimal. Different types of food differentially affects the bees' physiological needs for water in the winter. Minimum consumption of water was found in bees fed with sugar syrup (0.017 g/bee/day), and the maximum in bees fed with enzymatic invert syrup with the addition of yeast (0.025 gr/bee/day). The water content and the level of simple sugars in food for bees determines the amount of water consumed in the winter life of bee., Cilj rada je da se utvrdi brzina konzumiranja vode zimskih pčela, koje su hranjene različitim vrstama hrane. U zimskom periodu pčela je u nemogućnosti da dođe do prirodnih izvora vode, iako su u tom periodu potrebe pčela za vodom minimalne. Različita vrsta hrane različito djeluje na fiziološke potrebe pčela za vodom u zimskom periodu. Minimalna konzumacija vode je utvrđena kod pčela hranjenih sa šećernim sirupom 0,017 gr./pčeli/dan, a maksimalna kod pčela hranjenih sa enzimskim sirupom uz dodatak pivskog kvasca 0,025 gr./pčeli/dan. Sadržaj vode i nivo prostih šećera u hrani za pčele određuje količinu konzumirane vode u zimskom periodu života medonosne pčele.",

Natural mycobiota and aflatoxin B1 presence in bee pollen collected in Serbia

AB  - Total fungal count, incidence of fungi and aflatoxin B1 (AFB1) concentration were studied in 33 samples of bee pollen randomly collected from beekeepers in Serbia. The total number of fungi was determined by dilution method whereas AFB1 was detected using the Enzyme-Linked Immuno-Sorbent Assay (ELISA). The mycological estimation showed the presence of nine genera of fungi as followed: Acremonium, Alternaría, Aspergillus, Cladosporium, Epiccocum, Fusarium, Mucor, Pénicillium and Rhizopus, with total number ranging from 1 x 103 to 1 x 105 CFU g-1. The results have shown the predominance of the fungi from the genera Aspergillus and Alternaria. Among Aspergillus species it was observed that the most frequent species was A. flavus with incidence of 27.27 %. Mycotoxin AFB1 was detected as 100% positive in all samples (100%) with an average concentration of 8.61 μg kg-1. The obtained results indicated that honey bee pollen must be strictly controlled during its manipulation in the harvesting and manufacturing. Therefore, the implementation of good manufacturing (beekeeping) practice to define procedures for honeybee products could be crucial to reduce the risk of possible contamination and provide natural and safety product without risk on the human health.
AB  - Ukupan broj gljiva, učestalost (incidenca) gljiva i koncentracija aflatoksina B1 (AFB1) ispitivani su u 33 uzoraka polena sakupljenih od pčelara iz različitih regiona u Srbiji. Ukupan broj gljiva određen je primenom metode razređenja a AFB1 je određen primenom imunoadsorpcione enzimske metode (ELISA). Mikološkim ispitivanjima identifikovano je devet rodova gljiva: Acremonium, Alternaria, Aspergillus, Cladosporium, Epiccocum, Fusarium, Mucor, Penicillium i Rhizopus, sa ukupnim brojem od 1 x 103 to 1 x 105 CFU g-1. Najučestalije vrste gljiva su u rodovima Aspergillus i Alternaria. Među Aspergillus vrstama najučestalija je vrsta A. flavus sa incidencom od 27,27%. AFB1 je detektovan u svim uzorcima sa prosečnom koncentracijom od 8,61 μg kg-1. Dobijeni rezultati ukazuju da pčelarski polen mora biti strogo kontrolisan tokom prikupljanja i njegove dalje prerade. Zbog toga, sprovođenje dobre proizvođačke (pčelarske) prakse podrazumeva definisanje procedura za pčelarske proizvode što bi moglo biti presudno za smanjenje rizika od moguće kontaminacije i dobijanje prirodnih i bezbednih proizvoda bez rizika po zdravlje ljudi.

Insecticidal Activity of Sage (Salvia Officinalis) Essential Oil to Varroa Destructor (Acari: Varroidae) and Apis Mellifera (Hymenoptera: Apidae)

AB  - The need to find alternative systems of the fight against Varroa mite without application of chemicals and provide healthy bee products resulted in investigation of application of different plant essences to arthropod control. In order to perceive the sage essential oil (Salvia officinalis) bioactivity, contact residual toxicity of mites and bees was examined in the laboratory conditions. The chemical composition of essential oil was determined by standard GC and GC/MS methods. Different doses of the sage essential oil dissolved in acetone (0.1–10 μl/Petri dish) were applied in Petri dishes and left to dry for 20 minute at a room temperature. Following this period of time, ten honey bees and five adult female mites were added in each Petri dish and they were all maintained in controlled conditions (T = 30°C, Relative humidity = 60%). Survival of examined honey bees and Varroa mites was recorded two times, after 24 h and 48 h. The most prominent toxic effect on the examined Varroa mites was observed after 24 h and 48 h, with application of 10 μl of sage oil (the average values for dead mite individuals were 3.25 and 3.50, respectively). Recorded biological activities of the oil tested in different doses on both honey bee and Varroa mite revealed opportunity to proceed with further investigation by selecting the most appropriate variants and combinations of the most prominent individual components of the examined sage oil.
