Alginate/soy protein system for essential oil encapsulation with intestinal delivery

AB  - Preparation of alginate-soy protein isolate (AL/SPI) complex beads containing essential oil of thyme was carried out by emulsification of thyme oil in aqueous sodium alginate solution blended with SPI solution, followed by atomization via electrostatic extrusion and gelification with calcium ions. The process parameters were optimized by variation of the alginate (1-2.5 wt.%) and SPI (0-1.5 wt.%) concentrations. Dry alginate-SPI particles exhibited wrinkle surface while shape distortion of hydrogel beads occurred with >= 1.5 wt.% alginate concentration, whereas SPI induced reduction of the particle size. Encapsulation efficiency of 72-80 % based on total polyphenols was achieved. In SGF the samples exhibited oil release of 42-55 % (due to matrix shrinkage and proteins degradation by pepsin activity), while the rest was delivered in SIF within 2.5 h simultaneously with swelling and degradation of the matrix.

AB  - Mechanical, morphological and thermal properties of optical fiber adhesives, based on poly(ethylene-co-vinyl acetate) (EVA) with 18 and 40% of vinyl acetate, polymer blend of EVA/poly(methyl methacrylate) (PMMA), and graft polymer, raw EVA-g-PMMA, prepared via in situ free radical polymerization, were studied. Extraction of PMMA homopolymer from raw EVA-g-PMMA produced purified EVA-g-PMMA, which enable quantitative determination of graft initiating centers by NMR spectroscopy. FTIR and image analysis allowed determination of the graft content/level in purified EVA-g-PMMA. Grafting effect to compatibilization efficiency in EVA-g-PMMA was monitored by image analysis of gelation processes and determination of wetting angle. Single lap joint of adhesives/optical fibers were subjected to adhesive testing before and after aging on 60 degrees C for 60 h and 120 h. Thermogravimetry coupled with mass spectrometry showed different thermal stability and degradation pathways of adhesives, and reduced the degree of deacetylation of thermally treated EVA-g-PMMA adhesive. DSC analysis showed higher thermal stability of EVA-g-PMMA adhesive. Better mechanical properties of EVA-g-PMMA, i.e. 62.4% higher Charpy impact strength value in relation to EVA, was obtained.

Erythrocyte membranes from slaughterhouse blood as potential drug vehicles: Isolation by gradual hypotonic hemolysis and biochemical and morphological characterization

AB  - The present study was aimed at investigating the effect of isolation process-gradual hypotonic hemolysis on chosen parameters of the erythrocyte membranes (ghosts) originating from bovine and porcine slaughterhouse blood. The estimation of the gradual hypotonic hemolysis as a drug loading procedure for the erythrocyte ghosts was performed as well. Based on the results derived from analysis of the osmotic properties of the erythrocytes, the gradual hemolysis was performed with high volume of erythrocytes and 35 mM hypotonic sodium-phosphate/NaCl, enabling >90% of hemolysis for both types of erythrocytes. Detailed insight into ghosts' morphology by field emission-scanning electron microscopy revealed a distortion from erythrocyte shape and an altered surface texture with increased bilayer curvature for both samples. Compared to erythrocytes, an average diameter of ghosts from both type of erythrocytes decreased for only about 10%. The reported unidispersity of the isolated ghosts is of great importance for their potential application as vehicles of active compounds. Gradual hemolysis did not lead to substantial loss of cholesterol and membrane/cytoskeleton proteins. This result indicated the ghosts' possibility to mimic the chemical and structural anisotropic environment of in vivo cell membranes, which is of significance for drug diffusion and partition coefficients. Induced shift of phosphatidylserine to external surface of the ghosts demonstrated their potential application as vehicles for targeted drug delivery to cells of reticuloendothelial system. Ultra high-performance liquid chromatography and Fourier transform infrared spectroscopy revealed the presence of a drug model - dexamethasone-sodium phosphate, and its interaction with structural components in both types of erythrocyte ghosts.
