Isolation and characterization of xanthomonas euvesicatoria bacteriophages

AB  - Host range, plaque morphology, thermal inactivation point, genome size and restriction fragment patterns of ten bacteriophage isolates originating from soil, pepper seed and irrigation water collected from five localities in Serbia were studied. The bacteriophage isolates were selected based on their specificity to Xanthomonas euvesicatoria, causal agent of bacterial spot of pepper. The phages had similar plaque morphology, except for two isolates producing a plaque-surrounding halo in culture of X. euvesicatoria strain KFB 189. Four phage isolates were inactivated at 70 degrees C and six at 71 degrees C. All phages had genome size of approximately 22 kb and were differentiated into four types by their EcoRI and BamHI restriction fragment patterns. Examination of two phages by transmission electron microscopy classified them as A1 morphotype members of the Myoviridae family, order Caudovirales. Although specific to X. euvesicatoria, the phages were differentiated into three groups based on their ability to lyse 59 strains of this bacterium. Adsorption rates and one-step growth curves were determined for each group representative phage isolates.

Differentiation of Phytopathogenic agrobacterium spp.

AB  - Due to the difficulties in differentiation of phytopathogenic Agrobacterium spp. and lack of a standardized protocol, we carried out selection and evaluation of suitable methods based on the bacterial physiological, genetic and pathogenic properties. Strains of Agrobacterium tumefaciens, A. rhizogenes and A. vitis were differentiated using standard bacteriological and molecular methods. The biochemical and physiological tests confirmed authenticity of the strains. Two duplex PCR methods were conducted with four different primer pairs. In all strains, presence of plasmid virD2 and virC pathogenicity genes was detected. Chromosomal pehA gene was determined in A. vitis strain. Pathogenicity was confirmed on carrot slices and young plants of tomato and sunflower. Strains of A. tumefaciens and A. vitis were pathogenic on all test plants, while strain of A. rhizogenes induced characteristic symptoms only on carrot slices. The tests used in this study provided reliable discrimination between the three species and confirmed their identity as tumorigenic (Ti) Agrobacterium tumefaciens and A. vitis, and rhizogenic (Ri) A. rhizogenes.
AB  - Usled poteškoća u razlikovanju vrsta roda Agrobacterium i nedostatka standardizovanog protokola izvršena je procena i odabir pogodnih metoda u cilju njihove diferencijacije na osnovu fizioloških, genetskih i patogenih odlika. Diferencirani su sojevi Agrobacterium tumefaciens, A. rhizogenes i A. vitis primenom standardnih bakterioloških i molekularnih metoda. Primenom diferencijalnih testova sojevi su ispoljili očekivane biohemijsko-fiziološke karakteristike. Izvedene su dve 'duplex' PCR metode sa 4 različita tipa prajmera. Kod proučavanih sojeva detektovano je prisustvo virD2 i virC gena patogenosti, koji se nalaze na plazmidnoj DNK bakterije. Prisustvo hromozomskog gena pehA, odgovornog za sintezu enzima poligalakturonaze, utvrđeno je kod soja A. vitis. Patogenost je proverena na kriškama mrkve i mladim biljkama paradajza i suncokreta. Sojevi A. tumefaciens i A. vitis bili su tumorogeni na svim test biljkama, dok je soj A. rhizogenes ispoljio patogenost jedino na kriškama mrkve. Na osnovu dobijenih rezultata, proučavani sojevi diferencirani su kao tumorogeni (Ti) Agrobacterium tumefaciens i A. vitis, i kao rizogeni (Ri) A. rhizogenes.

Fatty acid analysis of Erwinia amylovora from Serbia and Montenegro

AB  - Automated method of fatty acid analysis was used to identify and study heterogeneity of 41 Erwinia amylovora strains, originating from 8 plant species grown in 13 locations in Serbia and one in Montenegro. All strains contained 14:0 3OH fatty acid, characteristic for the 'amylovora' group. According to fatty acid composition 39 strains were identified as E. amylovora as the first choice from the database. Due to their specific fatty acid composition, two strains were identified as E. amylovora, but as a second choice. Fatty acid analysis also showed that E. amylovora population from Serbia could be differentiated in three groups, designated in this study as α, β and γ. All strains originating from central or south Serbia, as well as four strains from north Serbia clustered into group α. Group β and γ contained only strains isolated in northern Serbia (Vojvodina). The results show that E. amylovora population in this area is heterogeneous and indicate pathogen introduction from different directions. Fatty acid analysis enabled identification at species level, as well as new insights of heterogeneity of E. amylovora population.
AB  - Automatizovana metoda analize masnih kiselina primenjena je za identifikaciju i proučavanje heterogenosti Erwinia amylovora. Kao materijal za analizu prikupljen je 41 soj E. amylovora izolovan iz 8 različitih vrsta domaćina gajenih u 13 lokaliteta u Srbiji i jednom lokalitetu u Crnoj Gori. Rezultati ukazuju da svi proučavani sojevi poseduju 14:0 3OH masnu kiselinu, koja je karakteristična za 'amylovora' grupu. Na osnovu sastava masnih kiselina 39 sojeva je identifikovano kao E. amylovora, kao prvi izbor iz baze podataka. Dva soja su identifikovana kao E. amylovora, ali tek kao drugi izbor iz baze podataka, što je najverovatnije posledica specifičnosti u sastavu njihovih masnih kiselina. Rezultati analize masnih kiselina takođe pokazuju da populacija E. amylovora poreklom iz Srbije nije homogena i da među sojevima postoje tri grupe ili profila, koji su u ovom radu obeleženi sa α, β i γ. Svi sojevi koji su izolovani na prostoru centralne ili južne Srbije pripadaju grupi α, kao i četiri soja izolovana na području Vojvodine. Grupama β i γ pripadaju samo sojevi izolovani na području Vojvodine. Dobijeni rezultati predstavljaju dokaz heterogenosti populacije E. amylovora na ovim prostorima i ukazuju na mogućnost prodora patogena u naše područje iz različitih pravaca. Analiza masnih kiselina omogućila je ne samo identifikaciju do nivoa vrste, već i nova saznanja o heterogenosti populacije E. amylovora na ovim prostorima.

Bacterial and phytoplasma diseases of stone fruits

AB  - Stone fruit production represents an important part of fruit growing in Serbia and provides significant profit to fruit growers. How ever, the profit can be compromised by various plant diseases. Those caused by bacteria or phytoplasma can be very difficult to control due to the pathogen nature and lack of efficient treatments. Control of Pseudomonas syringae pvs. syringae and morsprunorum is a serious problem for fruit growers in our country. 'Crown gall' pathogen Agrobacterium tumefaciens often infects rootstocks and young plants causing great loss in nurseries. Diseases not present in our country, such as bacterial canker of peach, caused by P. s. pv. persicae and bacterial leaf spot of stone fruits caused by Xanthomonas arboricola pv. pruni, are also listed here, since they are described in close proximity and there is a great risk for their introduction. In Serbia, presence of European Stone Fruit Yellows phytoplasma disease on plum and apricot has been confirmed. How ever, it causes less damage than other pathogens. Aim of this paper is to contribute to the knowledge of symptomatology and epidemiology of stone fruit bacterial and phytoplasma diseases. This should facilitate proper and timely diagnosis and there fore successful protection measures. .
AB  - U voćarstvu naše zemlje koštičave voćke zauzimaju značajno mesto i njihovo gajenje predstavlja profitabilnu delatnost. Međutim, rentabilnost voćarske proizvodnje mogu ugroziti različite biljne bolesti. Po značaju, raširenosti, ali i nemogućnosti uspešne kontrole, izdvajaju se oboljenja bakteriozne i fitoplazmozne prirode. Bakteriozno izumiranje, koje prouzrokuje Pseudomo nas syringae sa svoja dva patogena varijeteta syringae i morsprunorum, zadaje ozbiljne probleme voćarima u našoj zemlji. Značajan prob lem, takođe, predstavlja i Agrobacterium tumefaciens, prouzrokovač bakterioznog raka korena, koji nanosi štete u rasadničarskoj proizvodnji. U radu su pred stavljena i oboljenja koja nisu prisutna u našoj zemlji, a u svetu su opisana kao izuzetno destruktivna. To su bakteriozno izumiranje breskve, koje prouzrokuje P. s. pv. persicae i bakteriozna pegavost lišća koštičavih voćaka, čiji je prouzrokovač Xanthomonas arboricola pv. pruni. U Srbiji je prisutno i fitoplazmozno oboljenje Evropsko žutilo koštičavih voćaka na kajsiji i šljivi, koje za sada ne predstavlja ozbiljan problem. Cilj rada je da doprinese poznavanju simptomatologije i epidemiologije bakterioznih i fitoplazmoznih oboljenja koštičavih voćaka, kako bi se donela pravilna i pravovremena dijagnoza od koje zavisi uspešnost mera zaštite.

abstract = "Stone fruit production represents an important part of fruit growing in Serbia and provides significant profit to fruit growers. How ever, the profit can be compromised by various plant diseases. Those caused by bacteria or phytoplasma can be very difficult to control due to the pathogen nature and lack of efficient treatments. Control of Pseudomonas syringae pvs. syringae and morsprunorum is a serious problem for fruit growers in our country. 'Crown gall' pathogen Agrobacterium tumefaciens often infects rootstocks and young plants causing great loss in nurseries. Diseases not present in our country, such as bacterial canker of peach, caused by P. s. pv. persicae and bacterial leaf spot of stone fruits caused by Xanthomonas arboricola pv. pruni, are also listed here, since they are described in close proximity and there is a great risk for their introduction. In Serbia, presence of European Stone Fruit Yellows phytoplasma disease on plum and apricot has been confirmed. How ever, it causes less damage than other pathogens. Aim of this paper is to contribute to the knowledge of symptomatology and epidemiology of stone fruit bacterial and phytoplasma diseases. This should facilitate proper and timely diagnosis and there fore successful protection measures. ., U voćarstvu naše zemlje koštičave voćke zauzimaju značajno mesto i njihovo gajenje predstavlja profitabilnu delatnost. Međutim, rentabilnost voćarske proizvodnje mogu ugroziti različite biljne bolesti. Po značaju, raširenosti, ali i nemogućnosti uspešne kontrole, izdvajaju se oboljenja bakteriozne i fitoplazmozne prirode. Bakteriozno izumiranje, koje prouzrokuje Pseudomo nas syringae sa svoja dva patogena varijeteta syringae i morsprunorum, zadaje ozbiljne probleme voćarima u našoj zemlji. Značajan prob lem, takođe, predstavlja i Agrobacterium tumefaciens, prouzrokovač bakterioznog raka korena, koji nanosi štete u rasadničarskoj proizvodnji. U radu su pred stavljena i oboljenja koja nisu prisutna u našoj zemlji, a u svetu su opisana kao izuzetno destruktivna. To su bakteriozno izumiranje breskve, koje prouzrokuje P. s. pv. persicae i bakteriozna pegavost lišća koštičavih voćaka, čiji je prouzrokovač Xanthomonas arboricola pv. pruni. AB  - Hazelnut production is a fast growing production in Serbia lately. Bacterial diseases, resulting in premature dieback of plants in commercial orchards, are well known problem in countries with long hazelnut growing tradition. Consequently, it reduces the yield and quality of nuts, but in a long term it causes reduction of the exploitation period end serious economical loss. Last years, symptoms resembling those caused by bacteria, were observed in hazelnut orchards grown in Serbia. Due to low disease intensity and sporadic appearance bacterial diseases of hazelnut received no adequate attention from our plant pathologists and therefore were not studied in details. Aim of this paper is to point attention on economical importance of hazelnut bacterial diseases, their spread, characteristics of the pathogen, and factors contributing to the disease occurrence. Information on symptom differentiation and pathogen biology are important for accurate disease diagnosis and pathogen identification, which is ground for effective and timly protection strategy.
AB  - Proizvodnja leske intenzivno se širi poslednjih godina u Srbiji. U područjima sa dugom tradicijom u gajenju ove jezgraste voćke, dobro su poznate pojave prevremenog sušenja i izumiranja biljaka, usled pojave bakterioza u komercijalnim zasadima. To za posledicu ima smanjenje prinosa i kvaliteta plodova, a dugoročne posledice nastaju usled skraćenja eksploatacije zasada, kada i nastaju najveće ekonomske štete. Poslednjih godina, u zasadima leske na teritoriji Srbije, zapaženi su simptomi koji ukazuju na potencijalnu bakterioznu prirodu oboljenja. Nizak intenzitet simptoma, sporadična pojava, ali i nedovoljno interesovanje za fitopatološke probleme ove biljne vrste, razlog su slabog poznavanja pomenute problematike u nas. Cilj rada je da se skrene pažnja na ekonomski značaj bakterioza leske, rasprostranjenost, osnovne karakteristike patogena, kao i činioce koji doprinose pojavi i širenju bolesti. Poznavanje simptomatologije i biologije patogena od značaja je za pravilnu dijagnozu oboljenja i identifikaciju prouzrokovača, a predstavlja, takođe, osnovni preduslov za pravovremenu i uspešnu zaštitu.
