Morphological and molecular identification of potato cyst nematode populations in Serbia

AB  - Quarantine species such as potato cyst nematodes Globodera rostochiensis and G. pallida are present in Serbia since 1999 and 2005, respectively. These nematodes are sibling species and their morphological identification is complex due to their morphometric overlap. The cysts from the localities of Kladnica, Šanac, Gojna Gora and Milatovići were grown on susceptible potato varieties and their morphological differences have been discussed. To avoid ambiguities in species morphological designation a duplex PCR method was chosen for a rapid and accurate species identification. The whole procedure, from DNA extraction to DNA isolation, can be performed in a single day.
AB  - Karantinske vrste kao što su cistolike nematode krompira Globodera rostochiensis i G. pallida su prisutne u Srbiji od 1999. god. odn. 2005. god. Ove nematode su sestrinske vrste i njihova morfološka identifikacija je složena zbog morfometrijskog preklapanja. Ciste sa lokaliteta Kladnica, Šanac, Gojna Gora i Milatovići su gajene na osetljivim sortama krompira i njihove morfološke razlike su analizirane. Da bi se izbegle nejasnoće u morfološkom određivanju vrsta, izabran je duplex PCR metod za brzu i preciznu specijsku identifikaciju. Celokupna procedura, od ekstrakcije DNK do njene vizualizacije, može biti urađena za jedan dan.

Genital organ anomalies in Xiphinema dentatum (Nematoda : Longidoridae)

AB  - Seven specimens of Xiphinema dentatum with abnormal genital organs were found in the same population. Normal females are amphidelphic with the vulva located slightly anterior to the mid-body. Of some 24 000 females examined, six were found to be abnormal. One has a normally developed genital tract, but located in an abnormally posterior position. Two females have two complete systems each, one system in the normal position, and the other well posterior. Another female has a normal posterior branch, while the anterior one is abnormally branched at the pars dilatata uteri into two oviducts and ovaries, one ovary being degenerate. Another two females lack central parts of the system, the remaining distal parts being fused proximally. Males are rare in this species, sometimes having a fully developed diorchic genital tract, but usually with only the copulatory apparatus developed. Of the 137 males examined, one has a normal copulatory apparatus, lacks the seminal duct and has two abnormal masses of tissue corresponding to the position where the testes would normally be located.

Observations on spear replacement in Xiphinema dentatum (Nematoda : Longidoridae)

AB  - The process of spear (odontostyle + odontophore) replacement was determined by studying juveniles of Xiphinema dentatum in different phases of moulting. The process is subdivided into eight distinct phases, each readily identified by characteristic morphological features. The replacement odontostyle takes the functional position by assembling the new stoma behind the lining of the old cheilostoma, at the level where the replacement odontostyle inserts into the old odontophore during early moulting. A new spear is produced by the formation of the new odontophore behind the base of the new functional odontostyle, which was formed either during the previous moulting, or during embryogenesis (spear of the second stage juveniles). The most obvious event during spear replacement is development of a large space, void of tissues, in the region of the old cheilostoma, where a delicate sleeve-like membranous structure, coaxial with the old sternal was observed. This extends posteriorly from about the old amphids to the lit! region border of the newly forming stage. There are two, dorsal and ventral, Rap-like cuticular extensions of the base of the cheilostoma lining. In the postlabial region thre are, most likely four, sabre blade or carrot-like cuticular extensions.
