Comparison of essential oils and hydromethanol extracts of cultivated and wild growing Thymus pannonicus All.

AB  - In the present research, the chemical composition of aerial parts of cultivated citral chemotype of Thymus pannonicus All. (Lamiaceae), growing at different soil conditions was studied. Two plant lineages were unfertilized or treated using either nitrogen or phosphorus fertilizers at different concentration levels. Essential oil (EO) content, EO composition, and composition of hydromethanol extracts (MEs) were compared to the results obtained for wild growing plants in order to evaluate the potential of T. pannonicus for its industrial production. EO content (%, V/m) in cultivated samples varied between 0.32-0.75% and 0.62-1.05% within two examined lineages, whereas the herbs from the natural habitat contained 0.49-1.29% of EO. GC-FID/MS analysis showed that citral, i.e. mixture of isomers geranial and neral, was the most abundant constituent in all EOs (51.90-81.96%). HPLC analysis revealed that total caffeic acid derivatives, with rosmarinic acid (RA) as the most abundant compound in all extracts, as well as total apigenin glycosides contents were significantly higher in the extracts originating from the cultivated herbs (112.15-184.94 mg/g and 5.08-15.29 mg/g, respectively), in comparison to the extracts of the herbs from the natural habitats (67.61-98.75 mg/g and 1.17-1.32 mg/g). In general, the composition of cultivated herbs varied less than that of samples originating from natural habitats, indicating that controlled cultivation of citral chemotype of T. pannonicus can provide herbal drug with favourable characteristics. Only minor differences were observed between cultivated samples treated with different fertilizers.

Potential use of deodorised water extracts: polyphenol-rich extract of Thymus pannonicus All. as a chemopreventive agent

AB  - Deodorised water extracts of aromatic plants are obtained as by-products of essential oil isolation and usually discarded as waste. However, phytochemical composition of these extracts encourages their further utilization as food additives or functional food ingredients. In this study we investigated phytochemical composition, antioxidant and in vivo antiproliferative activity of deodorised water extract of Thymus pannonicus All. (DWE). HPLC analysis revealed rosmarinic acid (RA) (71.11 +/- 1.54 mg/g) as the most abundant constituent of the extract, followed by salvianolic acid H (14.83 +/- 0.79 mg/g, calculated as RA). DWE exhibited pronounced antioxidant activity in vitro, in FRAP and DPPH tests (FRAP value: 7.41 mmol Fe/g and SC50: 3.80 mu g/g, respectively). Using the model of Ehrlich carcinoma cells in mice that were treated with DWE prior, at the time, and after tumour cells implantation, the tumour growth suppression and redox status of malignant cells (i.e., activities of antioxidant enzymes, level of glutathione and intensity of lipid peroxidation) were followed. DWE applied as pretreatment caused disturbance of antioxidant equilibrium as well as apoptosis/necrosis of up to 90% EAC cells. Results obtained in the present study revealed chemopreventive potential and possibility of T. pannonicus DWE usage. High content of RA and other phenolic compounds explains, at least in part, the observed effects.

Bioactivity of herbal tea of Hungarian thyme based on the composition of volatiles and polyphenolics

AB  - Hungarian thyme (Thymus pannonicus All., Lamiaceae) is an aromatic herb used as traditional remedy, a refreshing beverage and a food aromatizer. Herbal teas, i.e., infusions, of Hungarian thyme from eight localities in Serbia were analyzed regarding their polyphenolic and volatile composition, and tested for their antioxidant and antimicrobial activity. The total polyphenolics content, determined by the Folin-Ciocalteu method, ranged from 1122.25 to 1979.93 mg gallic acid/L. HPLC analysis revealed rosmarinic acid (367.42-1199.47 mg/L) and luteolin glucuronides as the main polyphenolics. The volatile fractions of the infusions, analyzed by static headspace extraction coupled with GC and GC-MS analyses, contained citral, 3-octanone, 1-octen-3-ol, linalool and 1,8-cineole as the dominant constituents. The antioxidant activity of the infusions was examined through the ferric reducing antioxidant power (FRAP) (68.09-124.58 mmol Fe2+/L) and the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical tests (SC50 1.32-2.96 mu L/mL). The antimicrobial activity was tested by the broth microdilution method against standard strains of Gram(+) Staphylococcus aureus, Staphylococcus epidermidis, Bacillus subtilis and Enterococcus faecalis and Gram(-) bacteria Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeruginosa, and one strain of yeast Candida albicans. The infusions inhibited microbial growth in the tested concentration range (31.25-500.00 mu L/mL) and the strongest activity was exhibited against the strain of C. albicans (MIC 31.25-62.50 mu L/mL). The antioxidant and antimicrobial properties of the infusions were, to some extent, in correlation with the composition and content of the polyphenolic compounds, whereas the volatiles noticeably influenced the exhibited antimicrobial activity.

In vitro antioxidant activity of honeydew and multifloral types of honey from Serbia

AB  - Antioxidant properties of fifteen multifloral and honeydew types of honey from Serbia were assessed by determination of ferric-reducing antioxidant power (FRAP assay) and 2,2- diphenyl-1-picrylhydrazyl free radical scavenging ability (DPPH assay), with respect to their total polyphenol content and colour intensity. The results of this study showed that total polyphenol content, antioxidant activity and colour intensity varied widely among different samples, even within the same type of honey. All investigated parameters were strongly dependent on geographic origin of samples, further emphasizing the importance of their detailed chemical characterisation. In general, polyphenolic content in investigated samples of honey, expressed as catechin equivalents, ranged from 480.2 to 1861.1 mg/kg. The ferric-reducing antioxidant power of honey tended to be lower in brighter and higher in darker samples, varying between 489.6 and 3089.8 μmol Fe(II) per 100 g of honey. The correlation between the colour and antioxidant activity never reached statistical significance in the case of honeydew type of honey, which was opposed to a trend observed in the case of multifloral honey.
AB  - U ovom radu, ispitivana su antioksidantna svojstva petnaest uzoraka livadskog i šumskog meda, primenom Ferric-Reducing/Antioxidant Power i DPPH-free radical scavenging ability testa, u odnosu na sadržaj ukupnih polifenolnih sastojaka i intenzitet boje meda. Rezultati ovog ispitivanja su pokazali da su sadržaj ukupnih polifenola, antioksidantna aktivnost i intenzitet boje izrazito varirali od jednog uzorka do drugog, čak i u okviru svake vrste meda ponaosob. Svi ispitivani parametri su bili vrlo zavisni od geografskog porekla uzoraka, naglašavajući važnost detaljne hemijske karakterizacije meda. Generalno, sadržaj ukupnih polifenola u ispitivanim uzorcima meda, izražen u ekvivalentima katehina, kretao se u intervalu od 480,2 mg/kg do 1861,1 mg/kg. Sposobnost redukcije gvožđa bila je niža u svetlijim vrstama meda, a viša u tamnijim, varirajući između 489,6 i 3089,8 μmol Fe(II) na 100 g meda. Korelacija između intenziteta boje i antioksidantne aktivnosti nije imala statističku značajnost kod uzoraka šumskog meda, nasuprot trendu koji je uočen u slučaju livadskog meda.

AB  - The objective of this research was to study the effects that the extract of rhizome and root of Helleborus odorus W. et K. (Ranunculaceae) can have on modifications in the parameter values of white blood cells count and degree of phagocytosis by peritoneal macrophages and neutrophil granulocytes in Wistar rats. The trial was conducted on 28 rats divided into 4 groups with 7 animals in each group. To the control group of rats sterile physiological solution in the quantity of 0.25 mL/100 g BW was applied intramuscularly. For the purpose of monitoring the effect of the extract of rhizome and root of hellebore (HE) during a time period, the HE was applied intramuscularly to rats in a dose of 10 mg/100 g BW, while the blood samples for analysis were taken after 24h, 48h and 72h. The consequence of intramuscular application of HE was an increased count of total leukocytes in all trial groups, the most expressed leukocytosis being registered 24h after application of HE. Statistically significant higher value in the count and percent of neutrophil granulocytes in the blood was recorded 24h after treatment in relation to the control and two other trial groups (p lt 0.001), among which a statistical significance was not established. The extract of hellebore rhizome and root has led to lymphopenia, resulting in the increase of the neutrophil/limphocyte index in the trial groups 24h and 48h after treatment. The application of HE had no significant effect on the count of monocytes in treated animals. The applied extract has caused a significant increase in the degree of phagocytosis by residing peritoneal macrophages and neutrophil granulocytes in blood.
AB  - Cilj ovog rada je bio da se ispita uticaj ekstrakta rizoma i korena H. odorus W. et K. na promenu vrednosti parametara bele krvne slike i stepen fagocitoze od strane peritonealnih makrofaga i neutrofilnih granulocita kod pacova soja Wistar. Ogled je izveden na 28 pacova podeljenih u 4 grupe po 7 jedinki. Kontrolnoj grupi pacova je intramuskularno aplikovan sterilan fiziološki rastvor u količini od 0,25 ml/100 g TM. U cilju praćenja efekta ekstrakta rizoma i korena kukureka (EK) u toku vremena, pacovima je intramuskularno aplikovan EK u dozi od 10mg/100g TM, a krv za analizu je uzimana posle 24h, 48h i 72h. Intramuskularna aplikacija EK imala je za posledicu povećanje broja ukupnih leukocita u svim oglednim grupama, pri čemu je najizraženija leukocitoza registrovana 24h nakon aplikovanja EK. Statistički značajno veća vrednost broja i procenta neutrofilnih granulocita u krvi zabeležena je 24h posle tretmana u odnosu na kontrolnu i ostale dve ogledne grupe (p lt 0,001), između kojih nije utvrđena statistička značajnost. Ekstrakt rizoma i korena kukureka doveo je do nastanka limfopenije, što je imalo za posledicu povećanje neutrofilno/limfocitnog indeksa u oglednim grupama 24h i 48h nakon tretmana. Aplikacija EK nije značajno uticala na broj monocita kod tretiranih životinja. Upotrebljeni ekstrakt doveo je do značajnog povećanja stepena fagocitoze od strane rezidentnih peritonealnih makrofaga i neutrofilnih granulocita krvi.

Influence of Helleborus odorus W. et K. underground parts extracts on red blood pictures of Wistar rats

AB  - This paper presents the results of investigations of parameter values of the red blood cell count in Wistar rats, after intramuscular application of rhizome and root extract of Helleborus odorus W. et K (HE). The trial was conducted in experimental conditions on 49 Wistar rats divided into 7 groups with 7 animals per unit (control and six experimental groups). The control group was treated by an intramuscular injection of sterile saline (0.25 ml/100 g BM). For the purpose of determing the duration of the HE effects, the animals were injected with a dose of 10 mg/100 g BMand blood was collected by cardial taps after 24, 48 and 72 hrs. Moreover, we investigated the influence of elevated temperatures on the noted HE extract effects, 24 hrs after the application of the inactivated (56oC 30 min) or denaturized (100oC 5 min) extract, as well as the influence of dialyzed HE. The obtained results of haematological analyses have pointed out the weak hemolyses activity of the extract. In all experimental groups, the erythrocyte count was decreased, but remained within reference values. Haemoglobin concentration and haematocrit values in the experimental and the control groups of rats were decreased in comparison to the physiological values, while the applied extract did not significantly affect the examined parameters. .
AB  - U ovom radu su prikazani rezultati ispitivanja vrednosti parametara crvene krvne slike pacova soja Wistar, nakon intramuskularne aplikacije ekstrakta rizoma i korena Helleborus odorus W. et K. Ogled je izveden u eksperimentalnim uslovima na 49 pacova soja Wistar, podeljenih u 7 grupa sa po 7 jedinki (jedna kontrolna i šest oglednih grupa). Kontrolnoj grupi pacova je intramuskularno aplikovan fiziološki rastvor (0,25 ml/100 g t.m.). U cilju određivanja dužine trajanja efekata ekstrakta, pacovima je aplikovan tečni ekstrakt podzemnih organa kukureka u koncentraciji od 10 mg/100 g t.m., a krv je uzorkovana kardijalnom punkcijom nakon 24h, 48h i 72h. Ispitivan je i uticaj ekstrakta izlaganog povišenoj temperaturi na uočene efekte, 24h posle aplikacije inaktivisanog (56oC 30 min) ili denaturisanog ekstrakta (100oC 5 min), kao i efekat dijalizata ekstrakta. Dobijeni rezultati hematoloških analiza su ukazali na slabu hemolitičku aktivnost ekstrakta. Kod svih oglednih grupa, prosečan broj eritrocita bio je smanjen, ali se održao u granicama referentnih vrednosti. Zabeležene koncentracije hemoglobina i hematokrita u krvi pacova kontrolne i oglednih grupa bile su niže u odnosu na fiziološke vrednosti, pri čemu korišćeni ekstrakt nije značajno uticao na ispitivane parametre.

AB  - BACKGROUND: Knowledge about the chemical and therapeutic properties of Thymus glabrescens Willd. (Lamiaceae) is scarce and inconsistent. Therefore the main objectives of this study were to determine the yield and chemical composition of essential oils from wild-growing T. glabrescens populations, to assess their radical scavenging activity and to correlate the results with published data in order to deduce which components are responsible for the activity. RESULTS: The plant material yielded between 4.0 and 8.0 mL kg(-1) of essential oil. All samples contained considerable but variable concentrations of thymol (22.3-55.1%), depending on the source. Radical-scavenging activities of the oils were estimated by 1,1-diphenyl-2-picrylhydrazyl free radical (DPPH) assay against butylated hydroxytoluene (BHT) and thymol as positive controls. The observed activities (IC50 values ranged from 94 to 230 mu gmL(-1)) were strongly influenced by thymol concentration, as verified by rapid screening for DPPH radical-scavenging activity on thin layer chromatography (TLC) plates and regression analysis. CONCLUSION: These results represent the first report on the free radical-scavenging activity of T. glabrescens essential oil and one of the first comprehensive reports on its composition. Thymus glabrescens could be used in the food industry for seasoning purposes or for preserving processed foods from oxidative degradation.

AB  - In the prevention and treatment of milder forms of diseases, chronic diseases and recurrent infections in farm animals the application of individual or combined medicinal herb preparations has long been used. The application of some herbal preparations can initiate the function of the effectors defensive cells and potentiate their defensive capacities. On the other hand, some immuno-suppresive phytopreparations are used in order to reduce or prevent undesired activities of immune system (allergic reactions and autoimmune disease). By the regulations of the European Union from 2006 the use of antibiotics as growth stimulators in livestock production was forbidden so aromatic herbs and their extracts have recently been more and more used for that purposes. .
AB  - U prevenciji i lečenju blažih oblika bolesti, bolesti hroničnog toka i rekurentnih infekcija domaćih životinja, već dugo se primenjuju pojedinačna ili kombinovana biljna lekovita sredstva. Primena pojedinih biljnih preparata podstiče funkcije efektorskih odbrambenih ćelija organizma i potencira njegove odbrambene sposobnosti. Sa druge strane, neki imunosupresivni fitopreparati se upotrebljavaju u cilju smanjenja ili sprečavanja neželjenih aktivnosti imunog sistema (alergijskih reakcija i autoimunih bolesti). Propisima Evropske Unije zabranjena je od 2006. godine upotreba antibiotika kao stimulatora rasta u stočarskoj proizvodnji, pa se poslednjih godina u te svrhe sve više koriste aromatične biljke i njihovi ekstrakti. .

By the regulations of the European Union from 2006 the use of antibiotics as growth stimulators in livestock production was forbidden so aromatic herbs and their extracts have recently been more and more used for that purposes. ., U prevenciji i lečenju blažih oblika bolesti, bolesti hroničnog toka i rekurentnih infekcija domaćih životinja, već dugo se primenjuju pojedinačna ili kombinovana biljna lekovita sredstva. Primena pojedinih biljnih preparata podstiče funkcije efektorskih odbrambenih ćelija organizma i potencira njegove odbrambene sposobnosti. Sa druge strane, neki imunosupresivni fitopreparati se upotrebljavaju u cilju smanjenja ili sprečavanja neželjenih aktivnosti imunog sistema (alergijskih reakcija i autoimunih bolesti). Propisima Evropske Unije zabranjena je od 2006. godine upotreba antibiotika kao stimulatora rasta u stočarskoj proizvodnji, pa se poslednjih godina u te svrhe sve više koriste aromatične biljke i njihovi ekstrakti. .",

AB  - Chemical analysis and antimicrobial activity of essential oil isolated by steam distillation from Ambrosia artemisiifolia L., Asteraceae dried flowering herb were performed. Principal constituents were germacrene D (24.1%), limonene (16.83%), alpha-pinene (8.0%) and myrcene (7.4%), A significant bactericidal and fungicidal activity was observed against all microorganisms tested.
