Antibacterial effect of Juniperus communis and Satureja montana essential oils against Listeria monocytogenes in vitro and in wine marinated beef

AB  - Juniperus communis and Satureja montana essential oils (EOs) were chemically characterized and their activity against Listeria monocytogenes was evaluated. Antilisterial effect was determined in vitro and in situ, in red winemarinated beef, where the effect against common meat contaminants (aerobic heterotrophic mesophyll bacteria, Enterobacteriaceae and lactic acid bacteria) was also monitored. Microdilution assay on L. monocytogenes (strain ATCC19111 and three primoisolates) showed that minimal inhibitory concentrations (MICs) varied in the range 0.5-1% for both EOs. Effect of EOs mixtures was monitored in checkerboard assay revealing that all types of interactions including synergism were obtained only for isolates. In vitro time kill assay confirmed the mode of interactions: synergism for isolate from beef carcass (LMB), and its absence for ATCC19111 strain. The analysis of concentration dependent growth rates showed that estimated curve MIC values were notably lower than in microdilution assay (0.03-0.14%). In situ time kill assay against inoculated L. monocytogenes (ATCC19111 and LMB strains) and pre-existing meat spoiling contaminants was performed on marinated beef using EOs concentrations that were confirmed to be sensory acceptable. Basic red wine marinades and the ones containing each EO or their combination remarkably decreased the counts of all monitored groups comparing to saline control. The most pronounced effect was obtained with the marinade containing EOs mixture. For all monitored bacteria, the bactericidal effect during marination was followed by bacteriostatic effect during subsequent meat storage. Our results demonstrate for the first time the in situ efficacy of J. communis EO against meat contaminants and provide additional data on the preservation potential of S. montana EO.

AB  - The success of antioxidant therapy in hyperthyroidism implies that disease is mediated by oxidative stress, which is known as one of the causing agents of agemg, degenerative diseases, and cancer. The main objective of our study was to determine possible protective effects of methanolic extract of N. rtanjensis in triiodothyronine (T-3)-induced DNA breaks of human lymphocytes under in vitro conditions, based upon plant antioxidant capacity related to its phytochemical profile, mainly its polyphenolic complex. The total phenolic and flavonoid content and the antioxidant activity using in vitro 1,1-dyphenyl-2-picrylhydrazyl reagent (DPPH) was determined in methanolic extracts of plant leaves and flowers. The phenolic compound content of 62.73 +/- 1.80 mg of GaA/g, exhibited solid antioxidant activity (IC50= 112.59 +/- 0.95 g/ml). The antigenotoxic activity of 0.2, 0.5 and 1.0 mg/ml N. rtanjensis methanol extracts mixture with 100 mu M of T-3 was studied in human lymphocytes in vitro using the Comet assay. It is supposed that the antigenotoxicity of N. rtanjensis methanol extracts was caused by high presence of chlorogenic acid, rosmarinic acid and rutin, all known as efficient antioxidant bioactive compounds, which were determined by ultrahigh-pressure liquid chromatograph with MS/MS Mass Spectroscopy (UHPLC/-HESI-MS/MS).

Differences in symbiotic N-2 fixation of alfalfa, Medicago sativa L. cultivars and Sinorhizobium spp. strains in field conditions

AB  - In a two-year field trial, four alfalfa cultivars grown in all combinations with five strains of Sinorhizobium spp. were assessed for N-2-fixing potential and compatibility of symbionts with the aim to co-select plant and rhizobia as alternative approach to symbiotic N-2 fixation improvement. It was found that rhizobial strains, host cultivars and interactions between them had a highly significant effects on shoot dry weight (SDW), total and fixed N content of alfalfa. A significant variation in N-2 fixation effectiveness of rhizobial strains, from ineffective to significantly effective depending on cultivars and year of utilization was detected. Effective strains increased SDW by 49-114% (2010) and 35-86% (2011). In the first year the highest effectiveness had strain L5 with cultivars K-28 and BL-88 and strain L3Si with BL-88 while in the following year all applied strains were effective with different extent. Only one strain (L5) was effective with most of alfalfa cultivars over a two-year period. Percentage of N-2 fixed in plant mass for strain L5 was 33% in 2010, and 38% in 2011. In addition, one cultivar (BL-88) was identified to have superior symbiotic performances, exhibiting its high capacity for N-2 fixation. Results indicate that simultaneous selection of both symbionts should be applied for improvement of N-2 fixation in alfalfa.
