Effect of zeolite on growth of some medical industrial and mushrooms

AB  - We studied the influence of natural zeolite Minazel (Z) on the dynamics of growth of mycelium and fruiting bodies of several important edible and medicinal mushrooms. Phenotypic and biological efficacy as well as the content of selenium in carpophores Grifola frondosa mushrooms. In order to more fully assessed the effect of supplementation was done and comparative testing with the addition of some mineral raw materials: apatite and bentonite, for which there are states that can induce an increase in the yield of different crops. It was found that the addition of natural zeolite from Serbia has a positive effect on all monitored parameters, and that the most effective concentration of 1%. Achieved effects can be attributed to the properties of zeolites and ion exchange capacity, sorption, buffering operation. Actual results indicate that the zeolites are very suitable as supplements in industrial production conditions mushrooms. Their use allows for faster growth and change of the functional properties of the obtained mushrooms.
AB  - Ispitivan je uticaj prirodnog zeolita Minazel (Z) na dinamiku rasta micelijuma i plodonosnih tela nekoliko važnih jestivih i medicinskih gljiva. Praćena je i biološka efikasnost kao i sadržaj selena u karpoforima gljive Grifola frondosa. Da bi se potpunije ocenio efekat suplementacije, rađeno je i uporedno ispitivanje uz dodatak nekih mineralnih sirovina: apatita i bentonita, za koj postoje navodi da mogu usloviti povećanje prinosa pojedinih poljoprivrednih kultura. Utvrđeno je da dodatak prirodnog zeolita sa područja Srbije ima pozitivno delovanje na sve praćene parametre, a da je najefikasnija koncentracija 1%. Postignuti efekti mogu se pripisati osobinama zeolita: jonoizmenjivačka sposobnost, sorpcija, pufersko delovanje. Ostvareni rezultati ukazuju na to da su zeoliti veoma pogodni kao suplementi u industrijskim uslovima proizvodnje gljiva. Njihova primena omogućava brži rast i izmenu funkcionalnih osobina dobijenih pečuraka.

AB  - The aim of this study was to investigate the potential influence of selenized yeast (Sel Plex, Alltech Inc., Lexington, USA) on chemical composition and flavour of solid state grown mushroom Pleurotus ostreatus. Amino acid composition that influences the flavour of selenium-enriched P.ostreatus P80 (137.84 ppm of selenium in d.w.) and non enriched cultivated strains with particular emphasis on selenomethionine was determined by HPLC method, after complete hydrolysis. Volatile flavour compounds of mushroom cultures were analyzed by GC-MS using Headspace sampler. In mushrooms with high selenium content, selenium in the form of L-selenomethionine was present. High selenium concentration in fruit body did not significantly change the amino acid composition of mushrooms. The major amino acids of fruit body sample were glutamic acid, alanine, aspartic acid and tryptophan. In the fruit body of P.ostreatus P80 control, 30 volatile compounds were detected, and in selenium enriched sample number of detected compounds was 25. Compounds that were detected in control sample, but not in eriched fruit body are 2-amino-N-ethylpropanamide, 2-methylpropyl pentan-2-yl sulfite, 2,3-pentanedione, heptan-2-one, pentan-1-ol, 2-methyldihydrofuran-3(2H)-one,1-hydroxyacetone, 3-hydroxy-2-butanon, 2,5-dimethylpyrazine, 2,6-dimethylpyrazine and benzaldehyde. Compounds that were detected in enriched P.ostreatus P80, but not in control are acetone, ethylacetate, 2-propanol, acetonitrile, 2-methylbenzaldehyde and 2-hydroxyethylmethacrylate. Selenized yeast presents good source of selenium for selenium enriched mushroom production with good flavour, which is probably safety for consumption.

AB  - The activity of organic selenium against pathogenic molds and its use as a potential selenium source in the production of enriched mushrooms were examined. The effect of commercial selenized yeast on mycelia growth was examined using a method with mycelia disks and a well diffusion method. For mushroom enrichment, different concentrations of selenium were added to a growth substrate. The results presented in this paper suggest that the most suitable concentration of selenized yeast that inhibits the growth of the mycopathogenic molds is 70-100 mg/kg of selenium. With the addition of this concentration to the substrate, mushroom fruit bodies will uptake a high level of selenium, about 100 μg/g for Pleurotus spp., and 200 μg/g for Agaricus bisporus in dry weight of the mushroom. Thereby a double effect in the cultivation of mushrooms is achieved.

Fungi shiitake enriched with organic source selenium

AB  - Selenium (Se) is a trace mineral that is essential to good health but only in small quantities. Soils, grains and forage have very low amounts of selenium. In most countries, selenium content in soil ranges between 0,1 and 0,2 ppm. There are many ways for selenium supplementation but it is considered that the best way is to increase selenium content in the plants. Mushrooms contain relatively high protein levels and they can accumulate large amounts of selenium. It is reasonable to expect that Se could be incorporated into the proteins as selenomethionine and selenocysteine. Lentinus edodes (Shiitake) is an edible mushroom native to East Asia, which is cultivated and consumed in many Asian countries for over 2,000 years. Shiitake Mushroom is revered for its health-promoting activity; it boosts the immune system, lowers cholesterol, works as an anticoagulant and is helpful in cancer treatment. Bioaccumulation of selenium in industrial mushroom was investigated. Vegetative growth of mycelium were measured as colony diam in pure cultures supplemented with organic selenium from new synthetic compound Zn(II) complex with ligand 2,6-diacetylpyridines bis(selenosemicarbazone), (Zn(dapsesc)). Accumulation of selenium in the fruit body was determined by the use of ICP-OES (Inductively coupled plasma optical emission spectrometry). Selenium as a complex, was effectively taken up from substrates and accumulated in fruit bodies. Highest selenium content was in the cap (60 ppm). In mushrooms cultivated without selenium supplement, selenium content was approximately 1 ppm. .
AB  - Selen je esencijalni mikroelement, neophodan za normalno funkcionisanje organizma ljudi i životinja. Rezultati prvih istraživanja o obezbeđenosti zemljišta, žitarica i stočne hrane ukazali su na vrlo nizak nivo selena. U većini zemljišta sveta sadržaj ukupnog selena je u granicama od 0,1 do 2 ppm. Postoji više mogućnosti suplementacije selena (selenizovana kuhinjska so, pelete, inekcije), ali se smatra da je najbolji način povećanjem sadržaja selena u biljkama. S obzirom na to da gljive sadrže visok procenat proteina, očekuje se da će apsorbovati i inkorporirati selen u proteinima, u obliku kao što su selenometionin i selenocistein. Gljiva shiitake (Lentinus edodes) vodi poreklo iz Istočne Azije - Kina, Japan, a prvi pisani podaci govore da je poznata više od 2000 godina. Pregled literature ukazuje da ova gljiva smanjuje nivo holesterola, jača imunitet, ima antikoangulantnu ulogu i da se koristi u prevenciji nastanka tumora. Praćen je uticaj novosintetisanog organskog kompleksa Zn (II) sa ligandom 2,6-diacetilpiridin bis-(selenosemicarbazon) (H2dapsesc) - (Zn (dapsesc)) sa Hemijskog fakulteta Univerziteta u Beogradu na porast micelijuma gljiva i mogućnost akumulacije selena u plodonosnim telima. Ukupan sadržaj selena je određivan optičkim emisionim spektrometrom sa indukovano spregnutom plazmom, ICP-OES. Dobijeni rezultati govore da je selen iz organskog izvora uspešno usvojen. Sadržaj selena je bio očekivano najveći u šeširu, 60 µg/g suve mase. U kontrolnim pečurkama, ova vrednost se kretala u proseku oko 0.4 µg/g suve mase. .

AB  - Selenium is an essential nutrient, needed in the diet on a daily basis, but only in very small amounts (50 micrograms or less). Selenium helps prevent oxidative stress by working together with a group of nutrients that prevent oxygen molecules from becoming too reactive. It is a critical mineral for maintaining proper function of the thyroid gland. Selenium is incorporated at the active site of many proteins, including glutathione peroxides, which is particularly important for cancer protection. In this experiment, selenium has been studied for its effects on fungal growth and development in order to improve the pharmacological qualities of the industrial mushrooms. The growth of mycelia and fruit body formation of different medicinal mushroom strains of Lentinus edodes and Ganoderma lucidum over the wide range of concentrations and organic form of selenium were examined. Vegetative growths of mycelium were measured as colony diam in pure cultures supplemented with organic selenium from new synthetic compound Zn(II) complex with ligand 2,6-diacetylpyridine bis(selenosemicarbazone) (H2dapsesc)- [Zn(dapsesc)] in concentrations of 1, 10, 15, 20, 25, 30 and 50 μg/ml. Organic supplements were prepared with 5 ml of dimethyl sulfoxide, DMSO to dissolve it. For comparation, experiment was also performed with DMSO and ligand H2dapsesc. Compounds showed moderate activity, the complex being more active than the ligand and dissolver. Organic selenium in concentration 1-50 μg/ml did not significantly influence the growth of mycelia on agar base media. Media supplemented with different concentrations of Se complex showed different level of toxicity according to the different strains and species. Mycelia of both strains of L.edodes showed heatlthy growth at 10 μg/ml, while this concentration for G. lucidum G.L.-I strain was 1μg/ml. The most inhibiting concentration for L. edodes L31 was 25μg/ml, and for L. edodes L37 and G.lucidum G.L.-I 30μg/ml. Not any of tested concentrations inhibited mycelial growth by 50%. Control and complex-treated colonies both grew healthy. No toxic effect on mushroom species at any of trested concentrations of comlex. .
AB  - Selen je esencijalni nutrijent neophodan u svakodnevnoj ishrani, u malim količinama (svega oko 50 mikrograma). Selen u kombinaciji sa drugim nutrijentima pomaže u prevenciji oksidativnog stresa sprečavajući da molekuli kiseonika postanu suviše reaktivni. Selen je neophodan za normalnu funkciju tiroidne žlezde. U proteinima je inkorporiran kao aktivan deo, uključujući glutation-peroksidazu, što je značajno u prevenciji raka. Predmet istraživanja je ispitivanje uticaja selena na porast i razvoj gljiva, radi obogaćenja industrijskih jestivih gljiva - pečuraka. Ispitivan je uticaj većeg broja različitih koncentracija organskih jedinjenja selena na porast medicinski značajne gljive Lentinus edodes i Ganodrerma lucidum.Ispitivanje je obavljeno na sladnom agaru i porast micelijuma praćen je merenjem prečnika kolonije čiste kulture obogaćene selenom iz novosintetizovanog kompleksnog jedinjenja Zn(II) sa ligandom 2,6-diacetilpiridin bis(selenosemicarbazon) (H2dapsesc)- [Zn(dapsesc)] koncentracije 1, 10, 15, 20, 25, 30 and 50 μg/ml. Organski suplement je pripremljen rastvaranjem u 5 ml dimetil sulfoksida, DMSO. Zbog poređenja rezultata, obavljeno je i ispitivanje uticaja DMSO i liganda na porast micelijuma. Kompleks je pokazao bolji uticaj na porast micelijuma od liganda i rastvarača. Organski kompleks koncentracije 1-50 μg/ml nije značajno uticao na porast micelijuma. Rezultati su varirali u zavisnosti od koncentracije dodavanog jedinjenja i soja ispitivane gljive. Najbolji porast gljive L.edodes primećen je pri koncentraciji 10 μg/ml, dok je ova koncentracija za G.lucidum iznosila 1 μg/ml. Koncentracija koja je najviše inhibirala porast soja L.edodes L31 iznosi 25 μg/ml, dok su najnepovoljnije koncentracije za soj L.edodes L31 i G.lucidum 30 μg/ml. Nijedna od ispitivanih koncentracija nije inhibirala porast gljive 50%. Kontrolne i kolonije sa dodatkom selena su obe rasle normalno. Toksičnog efekta na sojeve nije bilo. .

AB  - Mushrooms are a popular and valuable functional food, low in calories and high in minerals, essential amino acids, vitamins and fibers. More than 270 mushroom species are high valuable nutritive food, but they also have different pharmacologicaly effects, what classify them in the group of the medicinal mushrooms. These mushrooms show immunomodulating and anticancerous properties, they act like antioxidants and antihypertensive substances, have ability to lower cholesterol and protect the liver, possess antiinflammatory, antidiabetic, antiviral and antimicrobial properties. Medicinal mushrooms are very reach sources of new biomedicinal molecules which can be used in treating different disorders in human organism. Among these biomolecules the most important are protein bounded polysaccharides which are made by basidiomycetes; thanks to their extraordinary properties these biomolecules are classified like antitumorous substances by American National Cancer Institute. .
AB  - Pečurke su popularna i vredna funkcionalna hrana, zahvaljujući niskom sadržaju kalorija i sa druge strane visokom sadržaju minerala, esencijalnih amino kiselina, vitamina i vlakana. Pored nutritivnog aspekta, dokazano je da više od 270 vrsta gljiva ima i različita farmakološka dejstva, što ih svrstava u grupu lekovitih gljiva. Ove gljive pokazuju imunomodulatorska i antikancerogena svojstva, deluju kao antioksidanti, antihipertenzivne supstance, snižavaju holesterol, štite jetru, pokazuju antiinflamatorna, antidijabetska, antivirusna i antimikrobna svojstva. Lekovite gljive su bogat izvor niza novih biomedicinskih molekula koji mogu da se koriste u tretiranju različitih poremećaja u ljudskom organizmu. Među ovim biomolekulima najznačajniji su polisaharidi vezani za proteine, koje stvaraju bazidiomicete, a zbog izuzetnih svojstava koje imaju Američki nacionalni institut za kancer klasifikovao ih je kao antitumorne supstance. .
