AB  - The aim of our study was to evaluate the use of Raman spectroscopy for pre-diagnostic estimation of weed response to bleaching herbicides. Model plants were Chenopodium album and Abutilon theophrasti treated with mesotrione (120 g a.i. ha−1). Raman single-point measurements were taken 1, 2, 3, and 7 days after herbicide application from different points on the leaves. Principal component analysis (PCA) was carried out on data normalized by the highest intensity band at 1522 cm−1 and using spectral region from 950 to 1650 cm−1 comprising mainly contributions of carotenoids. The carotenoids by intensive band at ∼1522 cm−1 and bands with lower intensity at ∼1155 and 1007 cm−1 in treated plants were confirmed. According to PC1 (the first principal component) and PC2 (the second principal component), the highest intensity bands responsible for treatment differentiation in C. album could be assigned to chlorophyll, lignin, and carotenes. According to PC1 in A. theophrasti leaves the treatment differences could be observed 7 days after mesotrione treatment and PC2 gave a clear separation between all control and treated leaf samples. Raman spectroscopy may be a good complement to invasive analytical methods, in assessing the plant abiotic stress induced by bleaching herbicides. © 2023 Taylor & Francis Group, LLC.

Procena uticaja mezotriona na sadržaj karotenoida u listovima primenom Ramanove spektroskopije

AB  - Reakcije biljaka na primenu i delovanje herbicida koji inhibiraju biosintezu karotenoida
i direktne promene sadržaja ovog pigmenta se mogu pratiti pomoću velikog broja
metoda koje podrazumevaju razaranje uzorka pre analize. Za razliku od tradicionalnih
analitičkih metoda, Ramanova spektroskopija se pokazala kao brza i nedestruktivna
metoda za detekciju hemijskog sastava biljnih uzorka. Cilj ovog istraživanja je bio da
se utvrdi da li se primenom Ramanove spektroskopije može detektovati uticaj
mezotriona na sadržaj karotenoida u listovima tretiranih korovskih biljaka,
Chenopodium album L. i Abutilon theophrasti Medik. kako bi se procenile mogućnosti
primene ove metode za praćanje promena u sadržaju ovih pigmenata pre nego što
promene postanu i vizuelno jasno uočljive. Preparat Skaut (a.s. mezotrion 480 g/l,
SC) u količini od 120 g a.s./ha uz dodatak okvašivača Alteox wet 40 (a.s. mineralno
ulje (500 g/l) i polioksi alkohol (500 g/l)) u količini 1 l/ha primenjen je kada su biljke Ch.
album bile u fazi 4 lista, a A. theophrasti u fazi 2 razvijena lista. Primenom Ramanove
spektroskopije (XploRA Raman spektrometar Horiba Jobin Yvon, laser Nd/YAG na
talasnoj dužini 532 nm) analiziran je sadržaj karotenoida u listovima tretiranih biljaka u
četiri termina: 1, 2, 3 i 7 dana nakon primene (DNP) mezotriona. Relativni sadržaj
karotenoida je dobijen izračunavanjem parametra intenziteta za trake koje ukazuju na
prisustvo dominantnih karotenoida u Spectragryph v1.2.13 softveru (Menges, 2018).
Dobijeni rezultati intenziteta traka u sledeća tri regiona: 1007, 1155-1160 i 1522-1525
/cm su pokazali da postoji statistički značajna razlika između analiziranih uzoraka
kontrolnih i biljaka tretiranih mezotrionom 3. i 7. DNP. Kod vrste Ch. album promene u
relativnom sadržaju karotenoida u listovima tretiranih biljaka zabeležene su već 3.
DNP u poređenju sa listovima kontrolnih biljka iste starosti, dok je kod vrste A.
theophrasti utvrđena statistički značajna razlika za isti parametar 7. DNP mezotriona.
Dobijeni rezultati ukazali su na kasniji uticaj mezotriona na promene relativnog
sadržaja karotenoida kod A. theophrasti, što se može pripisati specifičnoj građi
epidermisa lica lista, koji ima deblju kutikularnu prevlaku i brojne dlake koje
predstavljaju snažnu barijeru za apsorpciju herbicida preko lisne površine i posledično
kasniji uticaj na promene u sadržaju pigmenata. Rezultati ovog istraživanja pokazali
su da je Ramanova spektroskopija adekvatna metoda za brzu detekciju delovanja
mezotriona na promene u sadržaju karotenoida u listovima korovskih vrsta, posebno
kada one još nisu vizuelno jasno uočljive.

AB  - Raman spectroscopy has so far proved to be a suitable method for the analysis of carotenoids in a wide range of plant samples. This study aimed to determine whether the influence of mesotrione on the changes in carotenoid content of leaves in treated weed species Chenopodium album and Abutilon theophrasti can be detected using Raman spectroscopy. When the plants reached the targeted growth stages (C. album: 4 leaves, A. theophrasti: 2 leaves), Skaut herbicide (a.i. mesotrione 480 g l-1, SC) was applied in the amount of 120 g a.i. ha-1 in the tank mix with the adjuvant Alteox wet 40 (a.i. mineral oil (500 g L-1) + polyoxy alcohol (500 g L-1)), in the amount of 1 L ha-1. Leaf analysis, using Raman spectroscopy, was performed four times: on the 1st, 2nd, 3rd, and 7th day after the application of mesotrione. The intensity of the bands in the obtained Raman spectra, at positions 1007, 1155, and 1522 cm-1 in untreated and treated plants was analysed to determine the relative carotenoid content. The obtained results have shown that changes in the relative carotenoid content in the leaves of treated C. album plants can be detected as early as 3 days after the application of mesotrione, compared to the leaves of untreated plants of the same age. Meanwhile, a statistically significant difference for the same parameter in the leaves of treated A. theophrasti plants was evident at a later stage than in C. album, on the 7th day after the application of mesotrione. Consequently, this study has confirmed that Raman spectroscopy can be used as an adequate method for the rapid detection of the effect of herbicides on the content of carotenoids in the leaves of treated weed species.
AB  - Ramanova spektroskopija se do sada pokazala kao pogodna metoda za analizu karotenoida u širokom spektru biljnih uzoraka. Cilj ovog istraživanja je bio da se utvrdi da li se primenom Ramanove spektroskopije može detektovati uticaj mezotriona na promene sadržaja karotenoida u listovima tretiranih korovskih biljaka, Chenopodium album i Abutilon theophrasti. Kada su biljke dostigle odgovorajuću fenofazu (C. album: četiri lista, odnosno A. theophrasti: dva lista), primenjen je preparat Skaut (a.s. mezotrion 480 g L-1, SC) u količini od 120 g a.s. ha-1 uz dodat okvašivač Alteox wet 40 (a.s. mineralno ulje (500 g L-1) i polioksi alkohol (500 g L-1)) u količini 1 L ha-1. Analiza listova, primenom Ramanove spektroskopije, je urađena u četiri termina: prvog, drugog, trećeg i sedmog dana nakon primene (DNP) mezotriona. U dobijenim Ramanovim spektrima analiziran je intenzitet traka na pozicijama 1007, 1155 i 1522 cm-1 kod kontrolnih i tretiranih biljaka u cilju utvrđivanja relativnog sadržaja karotenoida. Dobijeni rezultati su pokazali da se kod vrste C. album mogu utvrditi promene u relativnom sadržaju karotenoida u listovima tretiranih biljaka već trećeg DNP mezotriona u poređenju sa listovima kontrolnih biljka iste starosti, dok je kod vrste A. theophrasti utvrđena statistički značajna razlika u istom parametru nešto kasnije u poređenju sa C. album, sedmog DNP mezotriona. Ovim istraživanjem je potvrđeno da se Ramanova spektroskopija može koristiti kao adekvatna metoda za brzu detekciju delovanja herbicida na sadržaj karotenoida u listovima tretiranih korovskih vrsta.

Study of the anatomical structure of Chenopodium album leaves in relation to susceptibility to herbicides

AB  - As the plant matures numerous changes at the morphological, physiological and anatomical level occur, leading to changes in the susceptibility to herbicides. Anatomical characteristics of apical fully developed leaves of Chenopodium album were studied in this paper. From plants at 2, 4, 6, 8 and 10 leaves stage, fully developed leaves were sampled and used to make permanent slides for light microscopy. The following parameters were measured on permanent microscope slide samples: thickness of the leaf, epidermal adaxial and abaxial, thickness of the mesophyll, palisade and spongy tissue. The obtained results have shown that maturing of the C. album plants can affect the anatomical structure of its fully developed leaves. Hence, it has been confirmed that the tissue thickness in leaves collected from plants in younger stages of development is significantly different from the tissue thickness of corresponding leaves collected from older plants.
AB  - Sa starenjem biljke najčešće dolazi do promena na morfološkom, fiziološkom i anatomskom nivou, što dovodi do promena u osetljivosti na herbicide. Ova istraživanja obavljena su sa ciljem da se utvrdi da li se sa starenjem biljaka menjaju anatomske karakteristike gornjih fiziološki zrelih listova Chenopodium album. Sa biljaka koje su bile u fazi 2, 4, 6, 8 i 10 listova uzorkovani su listovi u punoj fiziološkoj zrelosti i napravljeni trajni preparati za svetlosnu mikroskopiju, na kojima su mereni: debljina lista, debljina epidermalnih ćelija lica i naličja lista, debljina mezofila, palisadnog i sunđerastog tkiva. Dobijeni rezultati su pokazali da starenje vrste C. album može uticati na anatomsku građu fiziološki zrelih listova. Potvrđeno je da se debljina tkiva listova uzetih sa mlađih biljaka značajno razlikuje od debljine istih listova uzetih sa starijih biljaka.

abstract = "As the plant matures numerous changes at the morphological, physiological and anatomical level occur, leading to changes in the susceptibility to herbicides. Anatomical characteristics of apical fully developed leaves of Chenopodium album were studied in this paper. From plants at 2, 4, 6, 8 and 10 leaves stage, fully developed leaves were sampled and used to make permanent slides for light microscopy. The following parameters were measured on permanent microscope slide samples: thickness of the leaf, epidermal adaxial and abaxial, thickness of the mesophyll, palisade and spongy tissue. The obtained results have shown that maturing of the C. album plants can affect the anatomical structure of its fully developed leaves. Hence, it has been confirmed that the tissue thickness in leaves collected from plants in younger stages of development is significantly different from the tissue thickness of corresponding leaves collected from older plants., Sa starenjem biljke najčešće dolazi do promena na morfološkom, fiziološkom i anatomskom nivou, što dovodi do promena u osetljivosti na herbicide. Ova istraživanja obavljena su sa ciljem da se utvrdi da li se sa starenjem biljaka menjaju anatomske karakteristike gornjih fiziološki zrelih listova Chenopodium album. Sensitivity of Chenopodium album and Abutilon theophrasti to mesotrione depending on the growth stage

AB  - Growth stage of weeds can significantly affect the level of control at the moment of herbicide application. Maturation of plants usually lead to decreased sensitivity of plants. The aim of this research was to determine whether growth stage of weed species can affect the sensitivity of Chenopodium album and Abutilon theophrasti to mesotrione at the moment of application. Seeds of these species were sown in 5 days interval to ensure that at the moment of herbicide application Ch. album would be in stages of 2, 4, 6, 8 and 10 leaves and A. theophrasti in stages of 1, 2, 3, 4 and 5 leaves. When plants reached this stages herbicide Skaut (mesotrione 480 g/l) was applied in recommended dose rate 0.25 l/ha in tank-mix with adjuvant Alteox wet 40 (mineral oil (500 g/l) and polyoxyethylene alcohol (500 g/l)) in the dose rate 1 l/ha. The results showed that growth stage at the moment of application of mesotrione can significantly affects the sensitivity of both species to herbicide. The application of herbicides in the early growth stages (A. theophrasti - 1 leaf, Ch. Album - 2 and 4 leaves) resulted in total control of plants. With further maturation, sensitivity has decreased, in terms that the lowest effect was noted with A. theophrasti in growth stage of 5 leaves and Ch. album in growth stage of 10 leaves.
AB  - Fenofaza razvoja korovskih biljaka u momentu primene herbicida može značajno uticati na njihovu osetljivost. Sa starenjem biljke najčešće dolazi do smanjenja osetljivosti biljaka. Obavljena su istraživanja kako bi se utvrdilo da li fenofaza razvoja biljaka u momentu primene mezotriona utiče na osetljivost Chenopodium album i Abutilon theophrasti. Setva semena ovih vrsta je obavljena u razmacima od po 5 dana kako bi se obezbedilo da u momentu primene herbicida biljke Ch. album budu u fazi 2, 4, 6, 8 i 10 listova, odnosno A. theophrasti u fazi 1, 2, 3, 4 i 5 listova. Kada su biljke dostigle navedene fenofaze razvoja primenjen je preparat Skaut (mezotrion 480 g/l) u količini primene 0,25 l/ha, kome je dodat ađuvant Alteox wet 40 (mineralno ulje (500 g/l) + polioksi alkohol (500 g/l)) u količini 1 l/ha. Dobijeni rezultati su pokazali da fenofaza razvoja u vreme primene mezotriona značajno utiče na osetljivost obe ispitivane vrste. Primena herbicida u najranijim fazama razvoja (A. theophrasti - 1 list; Ch. album - 2 i 4 lista) dovela je do potpunog propadanja biljaka. Sa daljim starenjem biljaka osetljivost se smanjivala, tako da je najslabiji efekat utvrđen kod biljaka A. theophrasti u fenofazi 5 listova, a kod biljaka Ch. album u fenofazi 10 listova.
