Sugars and Organic Acids in 25 Strawberry Cultivars: Qualitative and Quantitative Evaluation

AB  - (1) The nutritional quality of strawberry (Fragaria × ananassa Duch) fruits, among others, is largely maintained by the presence of soluble sugars and organic acids. As the primary products of photosynthesis, they are energy depots in plants, necessary for the construction of cell constituents, but also serve as precursors of aromatic compounds and signaling molecules. (2) In this study, fruits of 25 strawberry cultivars were qualitatively and quantitatively characterized concerning individual sugars and organic acids by HPLC, FT-ICR-MS, and MS imaging analysis. In addition, the total quality index (TQI), as a novel mathematical model, was used to compare all individual parameters evaluated to obtain a quantitative single score, as an indicator of overall fruit quality. (3) Regardless of a large number of cultivars and monitored parameters that were studded, several cultivars stood out in terms of selected primary metabolites, such as 'Rumba', 'Jeny', and 'Sandra', while the latter had the best TQI score. (4) Intercultivar variations in sugars and organic acids profiles, along with other bioactive compounds, should be considered for selection of promising cultivars with improved naturally occurring nutraceutical traits. Besides the search for a pleasant taste, increased awareness of healthy nutrition resulted in heightening consumer demand for high-quality fruit.

AB  - The UV-B represents the minor fraction of the solar spectrum, while UV-C is not contained in natural solar radiation,
but both radiation types can cause damaging effects in plants. Cell walls (CWs) are one of the targets for
external stressors. Juvenile P. omorika trees were treated either with 21 day-high doses UV-B or with 7 day- UV-C
in open-top chambers. Using spectroscopic and biochemical techniques, it was shown that the response to UV
radiation includes numerous modifications in needle CW structure: relative content of xylan, xyloglucan, lignin
and cellulose decreased; cellulose crystallinity changed; yield of lignin monomers with stronger connection of
C––
C in side chain with the ring increased; re-distribution of inter- and intra-polymer H-bonds occurred. The
recovery was mediated by an increase in the activities and changes in isoform profiles of CW bound covalent
peroxidases (POD) and polyphenol oxidases (PO) (UV–B), and ionic POD and covalent PO (UV–C). A connection
between activities of specific POD/PO isoforms and phenolic species (m- and p-coumaric acid, pinoresinol and
cinnamic acid derivatives) was demonstrated, and supported by changes in the sRNA profile. In vivo fluorometry
showed phenolics accumulation in needle epidermal CWs. These results imply transversal connections between
polymers and changed mechanical properties of needle CW as a response to UV. The CW alterations enabled
maintenance of physiological functions, as indicated by the preserved chlorophyll content and/or organization.
The current study provides evidence that in conifers, needle CW response to both UV-B and UV-C includes
biochemical modifications and structural remodeling.

The aim of this study was to examine the effect of genotype, microbiological fertilisers and season on the rosette fresh weight and components of antioxidant activity. Six cultivars (green ‘Kiribati', ‘Aleppo', ‘Aquino' and red ‘Murai', ‘Carmesi', ‘Gaugin‘) were grown in a greenhouse experiment during three consecutive seasons (autumn, winter and spring) with application of microbiological fertilisers (EM Aktiv, Vital Tricho and their combination). Green cultivars showed higher fresh weight than red in spring and winter. Green cultivar ‘Aleppo’ showed the highest fresh weight in control during spring. Microbiological fertilisers led to increased fresh weight in autumn. Mainly, red cultivars showed higher quality parameters compared to green (total antioxidant capacity, total phenolic content, total carotenoids and POD activity). Red cultivar ‘Carmesi' showed the highest total antioxidant capacity, total carotenoids, and total phenolic content in winter with combination of fertilisers, Vital Tricho, and in control. In spring, ‘Carmesi' and ‘Gaugin' showed the highest POD activity with Vital Tricho, and combination of fertilisers. The present study suggested that genotype, fertilisers and season jointly influenced quantity and quality parameters with emphasis on Vital Tricho, and/or combination of fertilisers.

AB  - Phenolic compounds of 12 strawberry cultivars were profiled using spectrophotometry, spectrofluorometry, and high-performance liquid chromatography-mass spectrometry, coupled with multivariate regression analysis. Total phenolic content, total anthocyanin content, and total antioxidant capacity (TPC, TACY, and TAC, respectively) and concentrations of individual phenolics were evaluated, and the multivariate statistic was employed to identify the most promising cultivars based on their phenolic content. According to the principal component analysis, TAC was strongly correlated with the TPC (0.81), pointing out its importance in overall antioxidant activity. 'Joly', 'Laetitia', and 'Asia' cultivars were distinguished from others as the richest in concerning identified anthocyanins, almost all flavonoids and phenolic acids. Multivariate curve resolution with alternating least squares indicated the presence of two main types of fluorophores assigned to anthocyanins and phenolics, in which emission spectral ratios also showed the highest values in the referred cultivars. These cultivars could be recommended to consumers in terms of the health functionality of fruit.

Variation in health promoting compounds of blueberry fruit associated with different nutrient management practices in a soilless growing system

AB  - The objective of this study was to determine and compare the content of total anthocyanins (TACY), total phenolics (TPC) and total antioxidant activity (TAA) of the fruit of 'Bluecrop' highbush blueberry grown under different nutrient management practices in a soilless production system. A field study was carried out in a highbush blueberry plantation situated near Belgrade (Serbia), during the period of 2016-2017. The orchard was planted in the spring of 2016 in 50 l polypropylene pots with 5-year-old nursery plants. Each pot was filled with the mix of pine sawdust (60%), white peat (30%) and perlite (10%), and placed at a distance of 0.8 m within the row and 3.0 m between the rows (4,170 bushes ha-1 ). The following fertilizer treatments were evaluated: 1. organic fertilizers (Org); 2. mineral fertilizers (Min) and 3. a combination of organic and mineral fertilizers (Org-Min). Soluble NPK fertilizers were applied with irrigation water, whereas granulated mineral and pelleted organic fertilizers were mixed with the substrate. Fruit samples were collected in triplicate at the beginning of ripening, full maturity and the end of the harvest season. No significant effect of harvest time on each of the tested parameters was observed, whereas the content of TACY did not even differ under various fertilizer treatments. TPC in the fruit significantly increased in Org and Org-Min treatments (139.8 and 139.3 mg eq GA 100 g-1 FW, respectively) compared to Min treatment (122.7 mg eq GA 100 g-1 FW), while a considerably high TAA level was found only in berries under Org-Min treatment (0.53 mg asc g-1 FW).
AB  - Cilj ovog istraživanja bio je da se odredi i uporedi sadržaj ukupnih antocijana i ukupnih fenola, kao i antioksidativna aktivnost u plodu sorte visokožbunaste borovnice 'Bluecrop' gajene u supstratu pod uticajem različitih načina ishrane. Poljsko istraživanje je izvedeno u proizvodnom zasadu borovnice koji se nalazi u blizini Beograda (Srbija), tokom perioda 2016-2017. godine. Zasad je podignut u proleće 2016. godine sadnjom petogodišnjih biljaka u saksije od polipropilenske folije zapremine 50 l. Svaka saksija je ispunjena supstratnom smešom sastavljenom od strugotine četinara (60%), belog treseta (30%) i perlita (10%). Primenjeno rastojanje sadnje je 0,8 m u redu i 3,0 m između redova (4.170 žbunova po ha). Ispitivana je primena sledećih đubriva: 1. organska đubriva (Org); 2. mineralna đubriva (Min) i 3. kombinovana primena organskih i mineralnih đubriva (OrgMin). Rastvorljiva NPK đubriva su primenjivana kroz sistem za navodnjavanje, dok su granulisana mineralna i peletirana organska đubriva mešana sa supstratom. Uzorci plodova su uzimani u 3 ponavljanja na početku, sredini i na kraju sezone berbe. Vreme berbe nije ispoljilo značajan uticaj na testirane parametre, dok se sadržaj ukupnih antocijana nije razlikovao čak ni pod uticajem primenjenih đubriva. Sadržaj ukupnih fenola u plodu bio je značajno povećan u Org i Org-Min tretmanima (139,8, odnosno 139,3 mg ekv. gal. kis./100 g sv.m.pl.), dok je značajno veća ukupna antioksidativna aktivnost ustanovljena u plodovima iz OrgMin tretmana (0,53 mg ask./g sv.m.pl.).

AB  - Aluminium (Al) toxicity in acid soils is a global problem. Here, we investigated Al tolerance in high yielding winter wheat (Triticum aestivum L.) cultivars bred in Serbia. The common relative mot length (RRL) test for Al tolerance, and both physiological (malate efflux) and molecular (Aluminium-Activated Malate Transporter 1 [TaALMT1] expression) approaches were used for this characterization. Both moderately Al-tolerant cvs. Ljiljana and Arabeska showed significantly higher malate efflux rate from the root tips in comparison to moderately Al-sensitive cv. Pobeda and followed the RRL pattern. Irrespectively of Al supply, moderately Al-tolerant cultivars showed significantly higher relative TaALMT1 expression than the Al-sensitive ones. A considerably high level of Al tolerance was found in cv. Ljiljana, which showed the highest Al-induced malate efflux along with the highest constitutive expression level of TaALMT1 transcripts. Our results also demonstrate that Al tolerance is based on a constitutive trait of high TaALMT1 expression and malate efflux in wheat roots, resulting in a decrease in root length reduction.

Correlation of TPC with total antioxidant capacity (TAC) showed higher free-radical scavenging properties of Autumn Bliss (r(2) = 0.9999) compared to Polka (r(2) = 0.8972). Correlation coefficient between TACY and TAC were higher in Autumn Bliss (r(2) = 0.9939) compared to Polka fruits (r(2) = 0.8419). Although total protein concentrations were similar in both cultivars (similar to 0.35 mg mL(-1)), activities of peroxidases and polyphenol oxidases were much higher in Polka, which were confirmed with isoelectric focusing in cationic (pl 9.3) and anionic (pl 3.6) range. HPLC detection showed that among detected flavonoids (catechin, epicatechin, epicatechin gallate, rutin, myricetin, resveratrol, quercetin and kaempferol) epicatechin appears to be the most abundant compound. Chlorogenic, caffeic and p-coumaric acid were also detected. The results indicate that the changes in enzymes activities related to content of substrates play an important role in nutrient quality definition of raspberry fruits.

AB  - The objective of this work was to evaluate the biochemical composition of six berry types belonging to Fragaria, Rubus, Vaccinium and Ribes genus. Fruit samples were collected in triplicate (50 fruit each) from 18 different species or cultivars of the mentioned genera, during three years (2008 to 2010). Content of individual sugars, organic acids, flavonols, and phenolic acids were determined by high performance liquid chromatography (HPLC) analysis, while total phenolics (TPC) and total antioxidant capacity (TAC), by using spectrophotometry. Principal component analysis (PCA) and hierarchical cluster analysis (CA) were performed to evaluate the differences in fruit biochemical profile. The highest contents of bioactive components were found in Ribes nigrum and in Fragaria vesca, Rubus plicatus, and Vaccinium myrtillus. PCA and CA were able to partially discriminate between berries on the basis of their biochemical composition. Individual and total sugars, myricetin, ellagic acid, TPC and TAC showed the highest impact on biochemical composition of the berry fruits. CA separated blackberry, raspberry, and blueberry as isolate groups, while classification of strawberry, black and red currant in a specific group has not occurred. There is a large variability both between and within the different types of berries. Metabolite fingerprinting of the evaluated berries showed unique biochemical profiles and specific combination of bioactive compound contents.

Identification of fruit taste and antioxidant compounds in wild strawberry (F. Vesca L.) and everbearing varieties

Citric acid was determined to be a major organic acid in F. vesca and 'Regina' (11,04 i 15,09 mg g-1 FW, respectively). However, content of citric and malic acid exhibited almost identical values in everbearing variety 'Irma'. Among the phenolics measured, free ellagic acid was most abundant in achenes of strawberry variety 'Regina' (151,36 μg g-1 FW), whereas the highest amounts of pelargonidin-3-glucoside i cyanidin- 3-glucoside were detected in fruits of F. vesca. Similar trend was observed with total phenolic content, which expressed the highest value in achenes of strawberry variety 'Regina' (128,19 mg eqv. GA g-1 FW), but by analysing data obtained only for fruit flesh the highest value of total phenolics was obtained in F. vesca (10,30 mg eqv. GA g-1 FW). AB  - Rad prikazuje rezultate ispitivanja važnih parametara kvaliteta ploda i ilustruje razlike u sadržaju komponenti ukusa i izvesnih zdravstveno-promotivnih komponenti prisutnih u plodovima šumske jagode (F. vesca L.), sorte 'Regina' selekcionisane od stalnorađajuće forme F. vesca var. semperflorens i komercijalne stalnorađajuće sorte 'Irma' dobijene planskom hibridizacijom. Sorta 'Regina' je ispoljila najveći sadržaj fruktoze (14,44 mg g-1 sv. m. pl.) i glukoze (10,85 mg g-1 sv. m. pl.), dok je najveći sadržaj saharoze imala šumska jagoda (19,04 mg g-1 sv. m. pl.). A comparison of major taste- and health-related compounds of Vaccinium berries

AB  - The chemical fruit composition, phenolic profile, and corresponding total antioxidant activity of bilberry (Vaccinium myrtillus) and 2 commercial blueberry cultivars (Vaccinium corymbosum) were analyzed. Results from this study showed that cultivar Berkeley yielded the highest glucose and fructose contents (70.8 and 88.8 mg/g FW, respectively), and the sweetness index expressed a similar trend, achieving the highest value for cultivar Berkeley (279.2). Citric acid was the major organic acid in the berries tested. V myrtillus yielded a total organic acids value 2-fold higher (0.35 mg/g FW) as well as the highest vitamin C content (25.8 mg/100 g FW). Among the flavonols measured, myricetin was most abundant in V myrtillus (10.7 mu g/g FW), whereas the highest amounts of kaempferol and quercetin were detected in the blueberry cultivar Bluecrop (4.75 and 9.11 mu g/g FW, respectively). Despite the challenge of characterizing phenolic acids due to the complexity and natural variation in fruit composition, this study confirms that cultivars of V corymbosum are rich sources of chlorogenic acid, particularly cultivar Bluecrop (71.2 mu g/g FW). Nevertheless, total phenolics were higher in V myrtillus than in the cultivated blueberries, and consequently the highest level of total antioxidant capacity was recorded in wild bilberry (6.16 mg asc/g FW). This semicomprehensive study characterizes the fruit quality attributes and illustrates differences in the content of taste- and health-related compounds present in these Vaccinium berries.

Ellagic acid content was higher in F. vesca, R. idaeus and R. fruticosus (122.5 mu g/g FW, 12.71 mu g/g FW and 61.7 mu g/g FW, respectively) than that obtained in analyzed cultivars. Overall, TPH expressed higher values in the wild strawberry and blackberry species in comparison to the studied cultivars, and consequently, the highest levels of TAC were recorded in F. vesca (5.78 mg asc/g FW), followed by R. fruticosus (4.95 mg asc/g FW).

Pomological and antioxidant properties of red currant cultivars (Ribes rubrum L.)

AB  - Three red currant cultivars ('Heinemanns Rote Spatlese', 'Rondom' and 'Red Versailles') were assayed for pomological and antioxidant properties in the period of 2004-2006. It was found out that cv 'Rondom' was superior with regard to physical properties of bunch and fruit, whereas cv 'Heinemanns Rote Spatlese' expressed significantly lower values for these traits. However, the best chemical fruit composition was obtained by cv 'Heinemanns Rote Spatlese'. Regarding the content of some individual phenolic compounds (kaempferol, myricetin, quercetin, and ellagic acid) as well as the total phenolics, great variability was observed among the studied cultivars. Total phenolics expressed the highest value in cv 'Heinemanns Rote Spatlese' (1,70 mg GA g-1), and consequently the highest level of antioxidant capacity was recorded in this cultivar (2,13 mg ask g-1). AB  - Rad prikazuje rezultate trogodišnjih ispitivanja pomoloških osobina i antioksidativnog kapaciteta ploda kod tri sorte crvene ribizle (Industrija, Rondom i Versajska crvena). Ustanovljene su značajno niže vrednosti mase grozda (4,73 g), dužine grozda (6,85 cm), broja bobica u grozdu (9,1) i mase bobice (0,60 g) kod sorte Industrija, dok u pogledu hemijskog sastava ploda, odnosno sadržaja ukupnih prostih šećera (208,7 mg g-1) i ukupnih kiselina (0,45 mg g-1), ova sorta je pokazala superiornost. Rezultati sadržaja individualnih fenolnih jedinjenja (flavonola i elaginske kiseline) ukazuju da je sorta Versajska crvena ispoljila najviše prosečne vrednosti sadržaja miricetina (1,28 μg g-1), kvercetina (3,14 μg g-1) i elaginske kiseline (6,02 μg g-1), ali istovremeno i najni- ži prosečni sadržaj ukupnih fenola (0,82 mg GA g-1). AB  - The content of some important phenolic compounds (kaempferol, myricetin, quercetin, and ellagic acid), total phenolics (TPH), vitamin C, and total antioxidant capacity (TAC) in the fruit of three black currant cultivars (Ben Lomond, Ben Sarek and Malling Juel) were studied. The content of individual phenolic compounds showed variability among the studied cultivars. The main flavonol found in cv. Malling Juel was kaempferol (1.60 μg g-1 FW), whereas the lowest values of myricetin (5.46 μg g-1 FW) and quercetin (7.65 μg g-1 FW) were also observed in the fruit of this cultivar. Ellagic acid content was the highest in cv. Ben Lomond (12.90 μg g-1 FW), which is also characterized by lower values of individual flavonols compared to other cultivars tested. However, the highest TPH level was recorded in cv. Ben Lomond (4.71 mg g-1FW). Vitamin C content in fruit of cv. Malling Juel was at a very high level (141.4 mg 100g-1), whereas cv. Ben Lomond expressed the highest level of TAC (7.60 mg asc g-1 FW). Overall, the content and profile of phenolic compounds in the black currant cultivars showed significant contribution to the expressed antioxidant capacity which was confirmed by positive correlation between TPH and TAC obtained in this study. Besides total phenolics, vitamin C is also a significant contributor to the measured antioxidant capacity in black currant fruit indicating that some cultivars with lower phenolic content may express high TAC.
AB  - Rad prikazuje rezultate trogodišnjih ispitivanja sadržaja važnih fenolnih jedinjenja (flavonola - kampferola, miricetina i kvercetina, elaginske kiseline i ukupnih fenola), vitamina C, kao i vrednosti antioksidativnog kapaciteta registrovane u plodu tri sorte crne ribizle Ben lomond, Ben sarek i Moling džuel. Istraživanja su realizovana u periodu od 2004. do 2006. godine u laboratorijama Katedre za voćarstvo Poljoprivrednog fakulteta Univerziteta u Beogradu i Instituta za multidisciplinarna istraživanja iz Beograda. Ustanovljeno je variranje u sadržaju pojedinih individualnih fenolnih komponenti između ispitivanih sorti crne ribizle, pri čemu je kod sorte Moling džuel registrovan najveći prosečni sadržaj kampferola (1,60 μg g-1), ali i najniže prosečne vrednosti sadržaja miricetina (5,46 μg g-1) i kvercetina (7,65 μg g-1). Među ispitivanim sortama crne ribizle najveći sadržaj elaginske kiseline imala je sorta Ben lomond (12,90 μg g-1), koja se odlikuje nešto nižim vrednostima sadržaja flavonola u plodu, ali i najvećim sadržajem ukupnih fenola za ispitivani period (4,71 mg g-1). Pored ukupnih fenola, značajan doprinos antioksidativnom kapacitetu ploda daje i vitamin C. To objašnjava pojavu da su neke sorte posedovale visok antioksidativni kapacitet ploda uz, istovremeno registrovan, umeren sadržaj fenolnih jedinjenja.

Sugar and organic acids profile in the fruits of black and red currant cultivars

AB  - In this study, three black currant cultivars (Ben Sarek, Ben Lomond and Malling Juel) and three red currant cultivars (Industrija, Rondom and Red Versailles) were compared in terms of fruit quality by high-performance liquid chromatography analysis in order to determine sugars and organic acids profiles in their fruits. Among the analyzed individual sugars, fructose was determined to be the major component in all studied cultivars. Cv Ben Lomond exhibited the highest levels of glucose (78,8 mg g-1 FW), fructose (117,6 mg g-1 FW) and sucrose (10,1 mg g-1 FW) in comparison to the other tested black currant cultivars. Among investigated red currant cultivars the highest amounts of glucose (81,1 mg g-1 FW), fructose (113,1 mg g-1 FW) and sucrose (14,5 mg g-1 FW) were recorded in the fruits of cv Industrija, as well as the richest content of total sugars (208,7 mg g-1 FW). The same tendency was noticed considering the organic acids content (citric and malic), that is, the highest levels were registered with the cv Industrija (0,32 mg g-1 FW and 0,13 mg g-1 FW, respectively). Black currant cv Ben Sarek displayed the highest contents of citric and malic acids (0,68 mg g-1 FW and 0,14 mg g-1 F, respectively). Generally, dominant share of total acids was exhibited by citric acid in all studied black and red currant cultivars. These results point to the fact that the chemical fruit composition of currants significantly varied among the genotypes of each species analyzed.
