Risk assessment of dietary exposure to aflatoxin B1 in Serbia

AB  - Classified as Group 1 carcinogens aflatoxins (AFs) are of major concern for food safety and public health. Within the AFs group, aflatoxin B1 (AFB1) occurs at the highest levels in different food products and is considered as the most potent representative. A total of 463 samples of products susceptible to contamination with AFB1 and commonly consumed in Serbia were collected and analyzed. Consumption surveys were performed using a food frequency questionnaire (FFQ) and 24-h recall-based method. Monte Carlo simulation of 100,000 iterations was performed to estimate the intake of AFB1 through each food category, while a simple distribution approach was performed to estimate overall exposure of tested population groups. Risk characterization was performed by calculation of the Margin of Exposure (MOE) and by calculation of the number of possible hepatocellular carcinoma (HCC) cases. The highest exposure was observed in children with the mean overall estimated daily intake (EDI) in the range of 0.79–1.10 and 1.20–1.66 ng kg−1 bw day−1, followed by adolescents 0.56–0.81 and 0.94–1.32 ng kg−1 bw day−1, adult females 0.52–0.72 and 0.56–0.76 ng kg−1 bw day−1 and adult males with the mean overall EDI in the range of 0.39–0.56 and 0.47–0.66 ng kg−1 bw day−1 depending on consumption survey method and for the lower (LB) and upper bound (UB) scenario. MOE values that resulted from the mean values of the overall EDI of AFB1 were low, indicating high concern for all population groups. Based on obtained mean values of the overall EDI higher estimates of possible HCC cases caused by exposure to AFB1 were in the range of 0.01–0.02 cases/year/105 individuals, depending on the population group.

How changes in water quality under the influence of land-based trout farms shape chemism of the recipient streams-case study from Serbia

AB  - A total of eight trout farms with different production capacities located in highland regions of Serbia were selected for testing of their influence on water parameters of the recipient streams at 46 sites seven times in the course of a year. All of the trout farms can be divided into two groups with respect to the relationship of the values of the parameters recorded at their outlets and downstream sites and those recorded at the control sites. The study showed that trout farms most often cause a statistically significant increase in the contribution of fine particulate organic matter (FPOM %), in the concentrations of NH3, NH4+, and PO43- and in the values of total phosphorous (P-t) and water temperature (t(w)). On the other hand, they cause a statistically significant decrease in the values of dissolved oxygen. In our study, the parameter defined as the ratio of the annual production of a trout farm to the minimal water discharge (Pr/Q(min)) showed a high degree of correlation with the majority of analysed parameters, proven to be a good predictive model for testing the farm's influence on the recipient stream.

Oxygen Consumption Rate Analysis of Mitochondrial Dysfunction Caused by Bacillus cereus Cereulide in Caco-2 and HepG2 Cells

AB  - The emetic syndrome of Bacillus cereus is a food intoxication caused by cereulide (CER) and manifested by emesis, nausea and in most severe cases with liver failure. While acute effects have been studied in the aftermath of food intoxication, an exposure to low doses of cereulide might cause unnoticed damages to the intestines and liver. The toxicity which relies on the mitochondrial dysfunction was assessed on Caco-2 and HepG2 cells after exposure of one, three and ten days to a range of low doses of cereulide. Oxygen consumption rate analyses were used to study the impact of low doses of CER on the bioenergetics functions of undifferentiated Caco-2 and HepG2 cells using Seahorse XF extracellular flux analyzer. Both Caco-2 and HepG2 cells experienced measurable mitochondrial impairment after prolonged exposure of 10 days to 0.25 nM of cereulide. Observed mitochondrial dysfunction was greatly reflected in reduction of maximal cell respiration. At 0.50 nM CER, mitochondrial respiration was almost completely shut down, especially in HepG2 cells. These results corresponded with a severe reduction in the amount of cells and an altered morphology, observed by microscopic examination of the cells. Accurate and robust quantification of basal respiration, ATP production, proton leak, maximal respiration, spare respiratory capacity, and non-mitochondrial respiration allowed better understanding of the effects of cereulide in underlying respiratory malfunctions in low-dose exposure.

AB  - Biomasa (produkcija) makrozoobentosa je odabrana kao osnovni pokazatelj za praćenje promena kvantitativnog sastava naselja dna na lokalitetima uzvodno i nizvodno od pastrmskog ribnjaka u reci Rači.
Istraživanje sekundarne produkcije makrozoobentosa reke Rače sa sedam lokaliteta, obavljeno je u periodu 2011. (april, jun, septembar, oktobar, decembar) i 2012. godine (februar i maj). Dominantne grupe u biomasi makrozoobentosa su Hirudinea (Annelida), Mollusca, Gammaridae (Crustacea) i Trichoptera (Insecta).
Vrednosti biomase zoobentosa kretale su se u svim mesecima istraživanja u intervalu od 3,2001 g/m2, na lokalitetu RČ2 (u februaru) ,do 216,7120 g/m2, na lokalitetu RČ3 (u februaru).
Biomasa faune dna najveća je u svim mesecima istraživanja na lokalitetu RČ3, koji je lociran nizvodno od pastrmskog ribnjaka. Na ovom lokalitetu biomasa makroinvertebrata se kretala od 87,8643 g/m2 (u aprilu 2011. godine) do 216,7120 g/m2 (u februaru 2012. godine).

AB  - The isothermal kinetics curves of the swelling of a poly(acrylic acid) hydrogel in buffer solutions of different pH values (5, 7, and 9) at temperatures ranging from 30 to 40 degrees C were determined. The possibilities of applying the Fick kinetics model and the Peppas equation were examined. It was found that the applicability of these models were limited. The kinetics model of a first-order chemical reaction was found to describe the swelling kinetics of the PAA hydrogel in all the investigated buffer solution at all the investigated temperatures. Swelling kinetics is determined by the rate of expansion of the network. The kinetic parameters (E(a), In A) of the swelling of the PAA hydrogel in buffer solutions of different pH values were determined. The activation energy and the pre-exponential factor of the swelling of the PAA hydrogel in buffer medium decreased with increasing pH value of the swelling medium.

Comparison of the swelling kinetics of a partially neutralized poly(acrylic acid) hydrogel in distilled water and physiological solution

AB  - The isothermal kinetics curves of the swelling of a poly(acrylic acid) hydrogel in distilled water and physiological solution at temperatures ranging from 20 to 40 ºC were determined. The possibility of applying both the Fick’s kinetics model and kinetics model of the first order chemical reaction to the swelling kinetics of the PAA hydrogel in distilled water and physiological solution were examined. It was found that the possibilities of applying these models were limited. The new model of the kinetics of swelling in distilled water and physiological solution was established. The kinetic parameters (Ea, ln A) for the swelling in distilled water and physiological solution were determined. The decrease of the equilibrium degree of swelling and the saturation swelling rate of the swelling of the PAA hydrogel in physiological solution compared to swelling in distilled water could be explained by the decreased differences in the ionic osmotic pressures between the hydrogel and the swelling medium. The increase of the initial swelling rate in the physiological solution might be caused by an increased density of charges at the network and by an increased affinity of the network towards the water molecules. The increase of the activation energy of the swelling of the PAA hydrogel in the physiological solution is a consequence of its additional "ionic crosslinking".
AB  - U radu su ispitivane izotermalne kinetičke krive bubrenja hidrogela delimično neutralisane poli(akrilne) kiseline u destilovanoj vodi i fiziološkom rastvoru u temperaturnom opsegu od 20 do 40 °C. Ispitivana je mogućnost primene Fikovog kinetičkog modela kao i kinetike i reda hemijskih reakcija na kinetiku bubrenja poliakrilnog hidrogela.Utvrđeno je da su mogućnosti za njihovu primenu vrlo ograničene. Iz tih razloga primenjen je novi model kinetike bubrenja. Određeni su kinetički parametri (Ea, lnA) za procese bubrenja u destilovanoj vodi i fiziološkom rastvoru. Smanjenje ravnotežnog stepena bubrenja i saturacione brzine bubrenja hidrogela delimično neutralisane poli(akrilne) kiseline u fiziološkom rastvoru u odnosu na destilovanu vodu može se objasniti smanjenjem razlike u jonskom osmotskom pritisku između hidrogela i medijuma za bubrenje. Povećanje inicijalne brzine bubrenja u fiziološkom rastvoru u odnosu na destilovanu vodu prouzrokovano je povećanjem gustine naelektrisanja na polimernoj mreži i povećanim afinitetom prema molekulima vode. Povećanje energije aktivacije hidrogela delimično neutralisane poli(akrilne) kiseline pri bubrenju u fiziološkom rastvoru se objašnjava dodatnim "jonskim umreženjem" hidrogela u fiziološkom rastvoru.
