Clostridium botulinum spores in European honey bees from Serbia

AB  - A total of 61 honey bees from different regions of the Republic of Serbia were analyzed for Clostridium botulinum (C. botulinum) spores. The microbiological methods and molecular methods (multiplex PCR/mPCR and PCR method) were utilized to examine multiple subunits of each honey bees samples. The C. botulinum spores in PCR-positive samples were estimated by the most probable number method (MPN). The presence of C. botulinum spores, by applying mPCR and PCR methods, was detected in 1 of the 61 honey bees (1.64%). Using MPN method, the number of spores in positive sample was 110/kg. Detection of C. botulinum spores directly from untreated honey bees, without prior enrichment, is impossible by applying PCR. Using conventional microbiological methods, detection of C. botulinum spores in dead honey bees is not possible without preenrichment. Therefore, conventional, microbiological methods are not suitable for the detection of C. botulinum spores in honey bees. In order to detect C. botulinum spores in honey bees using PCR methods, due to the small and/or unequal distribution of spores in the samples, it is desirable to use multiple subunits/replicates for each sample examined.

Physicochemical parameters and microbiological status of honey produced in an urban environment in Serbia

AB  - Honey is a natural substance produced by honey bees (the genus Apis) enjoyed by people due to its unique nutritional and medicinal properties. The aim of this study was to determine the physicochemical parameters (moisture, ash, water-insoluble content, reducing sugars, sucrose, five acidity, diastase activity, hydroxymethylfurfural, and electrical conductivity) and microbiological status (total number of aerobic mesophilic bacteria, total number of sulfite-reducing clostridia, the presence of Salmonella spp., total numbers of fungi and yeasts and the presence of Clostridium botulinum) in honey (honeydew, blossom, sunflower, acacia, and linden) produced in an urban environment in Serbia. We analyzed 19 apiary samples of honey, collected during the 2011 harvesting season, by using recommendation methods. Physicochemical parameters of the examined honey produced in the urban environment indicated the honeys were of acceptable quality. Bacillus spp. were detected in four honeys, yeasts were detected in three honeys, and Clostridium botulinum type E was detected in one honey using PCR. The current study also showed the presence of diverse honey varieties in Serbia.

AB  - The presence of Clostridium botulinum spores in 59 honey samples originating from different regions of the Republic of Serbia was studied. In addition to microbiological methods, after enrichment, centrifugation and membrane filtration, molecular methods (PCR methods) were utilized. The number of spores in PCR positive samples was estimated by the most probable number (MPN) method. PCR confirmed C. botulinum spores in 5 (8.47%) honey samples. MPN of spores varied from 20/kg to 204/kg honey. PCR was more sensitive than cultural methods. Natural honey contamination with C. botulinum spores is low-level and not homogeneous, and therefore, PCR methods require multiple sub-sampling.

Colony strength in the spring inspection and its impact on the amount of foraged pollen at the time of red clover pollination

AB  - In this study, the impact of honey bee colony strength in the spring inspection on the colony strength at the time of pollination, the amount of foraged pollen and on the colony strength in autumn was observed. The honey bee colonies were, after the spring inspection, divided into two groups, based on the amount of bees. The weak colonies, in spring inspection, had an average up to 4 frames occupied by bees and the strong colonies, in the spring inspection, had more than 6 frames occupied by bees. In addition to the amount of bees, the amount of brood and food supplies were assessed in the inspections. It was determined that the stronger colonies had more pollen foragers in all three year of observation. The quantity of foraged pollen, in addition to strength in the spring inspection, was influenced by year. In two years (first and third) more pollen and larger quantity of red clover pollen was collected by the strong colonies, while in the second year, more pollen and a large quantity of red clover pollen was collected by weak colonies. In the fall inspections was found that the strong colonies still had more bees and brood, more pollen and, also, more honey in relation to the weak colonies.
AB  - U radu je praćeno kako, snaga društava utvrđena na prolećnom pregledu, utiče na snagu društava u vreme oprašivanja crvene deteline, na količinu sakupljenog polena, količinu polena crvene deteline i na snagu društva na jesenjem pregledu. Društva su nakon prolećnog pregleda, a na osnovu količine pčela, svrstana u dve grupe, od po 10 društava. Slaba društva su na prolećnom pregledu imala prosečno do 4 rama zaposednuta pčelama, a jaka društva su prosečno na prolećnom pregledu imala više od 6 ramova sa pčelama. Pored količine pčela na pregledima je praćena i količina legla, i zaliha hrane. Praćenje je trajalo tri godine, utvrđeno je da jaka društva imaju više izletnica i više polenarica u vreme oprašivanja crvene deteline u sve tri godine posmatranja. Na količinu sakupljenog polena pored snage društva na prolećnom pregledu uticaj ima i godina. U dve godine (prvoj i trećoj) više polena ukupno i veću količinu polena crvene deteline sakupila su jaka društva, dok su u drugoj godini nešto više polena ukupno i veću količinu polena crvene deteline sakupila slaba društva. Na jesenjem pregledu je ustanovljeno da su jaka društva i dalje imala veću snagu (više pčela i legla), više polena, ali i više meda u odnosu na slaba društva. The effect of carbohydrate additive and inoculation on quality of red clover silage

AB  - In this experiment, wilted masses of red clover of cultivar K-17 from the first cut was ensiled in three treatments: a) no additives, b) with the addition of corn (6% of biomass) and c) with the addition of inoculant BioStabil Plus. The experiment design was according to the method of a completely random plan (single factorial trial) in triplicates. Based on the results it can be concluded that the wilted biomass of red clover can be successfully ensiled without additives. However, the inoculation of red clover biomass achieves the most favourable pH value (4.20), the lowest level of degradation of the protein expressed in the amount of NH3-N (107.7 gkg-1 N), the largest production of lactic acid (91.3 gkg-1 DM) and acetic acid (42.6 gkg-1 DM), in the absence of butyric acid. Adding maize meal in the amount of 6% contributed to somewhat more favourable fermentation and increase of the energy value of silage. When using the DLG and Weissbach methods for assessing the quality of silage, all silages were classified into the first class. Contrary to this, according to the Zelter method, control and inoculated silages were evaluated as class III, because of the large amounts of acetic acid. In practices inoculants based on homo-and hetero-fermentative bacteria of lactic acid fermentation are recommended for use, because the increased production of acetic acid contributes positively to te aerobic stability of silage.
AB  - U eksperimentu je silirana provenula masa crvene deteline sorte K-17 iz prvog otkosa u tri tretmana: a) bez aditiva, b) sa dodatkom kukuruzne prekrupe (6% od biomase) i c) sa dodatkom inokulanta BioStabil Plus. Eksperiment je postavljen po metodi potpuno slučajnog plana (monofaktorijalnog ogleda) u tri ponavljanja. Na osnovu utvrđenih rezultata može se zaključiti da se provenula biomasa crvene deteline može uspešno silirati bez aditiva. Međutim, pri inokulaciji biomase crvene deteline postiže se najpovoljnija pH vrednost (4.20), najmanji stepen degradacije proteina izražen kroz količinu NH3-N (107.7 gkg-1 N), najveća produkcija mlečne kiseline (91.3 gkg-1 DM) i sirćetne kiseline (42,6 gkg-1 DM), uz istovremeno odsustvo buterne kiseline. Dodavanje kukuruzne prekrupe u količini od 6% doprinosi nešto povoljnijoj fermentaciji i povećanju energetske vrednosti silaže. Pri korišćenju DLG i Weissbach metode za ocenu kvaliteta sve silaže su svrstane u I klasu. Nasuprot tome, pri korišćenju Zelter metode, kontrolna i inokulisana silaža su ocenjene III klasom, zbog velike količine sirćetne kiseline. Za praksu se preporučuje upotreba inokulanata na bazi homo- i heterofermentativnih bakterija mlečnokisleinskog vrenja, jer povećana produkcija sirćetne kiseline pozitivno doprinosi aerobnoj stabilnosti silaža. Based on the results it can be concluded that the wilted biomass of red clover can be successfully ensiled without additives. However, the inoculation of red clover biomass achieves the most favourable pH value (4.20), the lowest level of degradation of the protein expressed in the amount of NH3-N (107.7 gkg-1 N), the largest production of lactic acid (91.3 gkg-1 DM) and acetic acid (42.6 gkg-1 DM), in the absence of butyric acid. Adding maize meal in the amount of 6% contributed to somewhat more favourable fermentation and increase of the energy value of silage. When using the DLG and Weissbach methods for assessing the quality of silage, all silages were classified into the first class. Contrary to this, according to the Zelter method, control and inoculated silages were evaluated as class III, because of the large amounts of acetic acid. In practices inoculants based on homo-and hetero-fermentative bacteria of lactic acid fermentation are recommended for use, because the increased production of acetic acid contributes positively to te aerobic stability of silage., U eksperimentu je silirana provenula masa crvene deteline sorte K-17 iz prvog otkosa u tri tretmana: a) bez aditiva, b) sa dodatkom kukuruzne prekrupe (6% od biomase) i c) sa dodatkom inokulanta BioStabil Plus. Eksperiment je postavljen po metodi potpuno slučajnog plana (monofaktorijalnog ogleda) u tri ponavljanja. Na osnovu utvrđenih rezultata može se zaključiti da se provenula biomasa crvene deteline može uspešno silirati bez aditiva. Međutim, pri inokulaciji biomase crvene deteline postiže se najpovoljnija pH vrednost (4.20), najmanji stepen degradacije proteina izražen kroz količinu NH3-N (107.7 gkg-1 N), najveća produkcija mlečne kiseline (91.3 gkg-1 DM) i sirćetne kiseline (42,6 gkg-1 DM), uz istovremeno odsustvo buterne kiseline. Dodavanje kukuruzne prekrupe u količini od 6% doprinosi nešto povoljnijoj fermentaciji i povećanju energetske vrednosti silaže. Pri korišćenju DLG i Weissbach metode za ocenu kvaliteta sve silaže su svrstane u I klasu. Nasuprot tome, pri korišćenju Zelter metode, kontrolna i inokulisana silaža su ocenjene III klasom, zbog velike količine sirćetne kiseline. Za praksu se preporučuje upotreba inokulanata na bazi homo- i heterofermentativnih bakterija mlečnokisleinskog vrenja, jer povećana produkcija sirćetne kiseline pozitivno doprinosi aerobnoj stabilnosti silaža. Uzorci pčela radilica su prikupljeni sa 7 različitih lokacija čime su pokriveni glavni pčelarski regioni na teritoriji Republike Srbije. Morfometrijska istraživanja pokazala su da pčele iz Zapadno Bačkog regiona u Vojvodini imaju najveću vrednost uglova B4 (113,17º), D7 (100,10º), E9 (24,28º), K19 (79,60º) i najveći kubitalni indeks (2,92). Iste pčele se na osnovu multivarijantne diskriminantne analize najviše razlikuju od grupe pčela sa Kosova. Na osnovu analize varijanse, utvrđeno je da postoji vrlo značajna razlika (P lt 0.01) u 7 morfoloških karakteristika između grupa pčela sa izabranih lokaliteta. Može se zaključiti da utvrđene vrednosti za ispitivane karaktere ukazuju na postojanje značajne varijabilnosti i impliciraju na postojanje subpopulacija uslovljenih delovanjem čoveka.

Variability of production characteristics of distinguished lines of bees in western Serbia

AB  - Condition for achieving high performance results in beekeeping is focused on the use of high-quality breeding queen bee. Because that a large amount of research are based on the examination of properties of selected lines of bees and choosing the best queen mothers in the aim of selection and improving the production characteristics of honey bees from the territory of Serbia. The amount of bees, brood, honey and pollen was investigated in two spring and one autumn survey and productivity controlled in the pasture of accacia. Line 1 had the greatest amount of brood (1.23, 4.04 and 5.39), while line 3 had the best spring development. The best quality of brood in all three exams had the lines 1 and 3 (3.00, 3.00 and 2.95) as well as the best productivity compared to the average yield of all other three lines. All tested lines were exhibited variability, which provides enough possibility for their further improvement and single out the mother queenbees with best results.
AB  - U dvogodišnjem istraživanju ispitivano je četiri odabrane linije medonosne pčele na području Zapadne Srbije. Proizvodne karakteristike praćene su prema standardnoj metodologiji. Na osnovu rezultata istraživanja utvrđeno je da je najveću površinu pčela imala linija 1 (u jesenjem i prvom prolećnom pregledu: 2,10 i 3,27 rama), dok je u drugom prolećnom pregledu najviše pčela imala linija 3 (6,19 rama). Utvrđene razlike u površini pčela nisu bile statistički značajne (P>0.01). Najbolji prolećni razvoj legla između prva dva prolećna pregleda utvrđen je u liniji 3 (povećanje za 1,4 rama) i liniji 1 (povećanje za 1,35 rama). Najveću površinu meda i polena imala su društva linije 1. Najbolji kvalitet legla, po pregledima utvrđen je u linijama 1 i 3. Na osnovu rezultata istraživanja, utvrđeno je da su linija 1 i 3 imale najbolje proizvodne karakteristike. Postojeća varijabilnost ispitivanih karakteristika pčela pruža mogućnost daljeg usavršavanja linija i širenja genetskog napretka na području Srbije, pri čemu prednost treba dati umnožavanju matica na bazi biološkog materijala pčela linija 1 i 3.

AB  - State of mutual or reciprocal relationship between traits is determined by the correlation coefficients. Correlation of morphological and production traits was determined in two generations of honey bee colonies. In the first generation, mother queen bees were studied, and in the second generation, their offspring. The study of one generation lasted two years. Morphological traits were determined in the beginning of the study and production traits were determined in the spring and autumn inspections of every year. In the first generation, the positive and mostly high correlation between morphological traits, and honey yield and hygienic behavior was determined. Correlation of morphological traits, colony strength and food supplies per colony ranged from highly positive to slightly negative values. It was found that there was a high correlation between some production traits (colony strength in the spring and autumn inspections and honey yield per colony). In the analysis of offspring (second generation), most traits that were positively correlated in mother generation now, also, showed a similar relationship. The only difference, in contrast to mother generation, was that, for most traits, the intensity of interaction increased.
AB  - Stanje uzajamne ili recipročne povezanosti jedne osobine od druge utvrđuje se na osnovu koeficijenta korelacije. Korelacija morfoloških i proizvodnih osobina utvrđivana je kroz dve generacije društava medonosne pčele. U prvoj generaciji praćene su majke matice, a u drugoj njihovo potomstvo. Ispitivanje po jednoj generaciji trajalo je dve godine. Morfološke osobine su utvrđene na početku praćenja generacija, a proizvodne na prolećnom i jesenjem pregledu svake godine. U prvoj generaciji utvrđena je pozitivna i mahom visoka korelacija između morfoloških osobina, prinosa meda i higijenskog ponašanja. Stepen korelacije morfoloških osobina, snage društva i zaliha hrane po društvu bio je od visoko pozitivne do slabo negativne vrednosti. Ustanovljeno je da postoji i visok stepen korelacije između pojedinih proizvodnih osobina (snaga društava na prolećnom i jesenjem pregledu i prinosa meda po društvu). Prilikom testiranja potomstva (II generacija) većina osobina koje su bile u pozitivnoj korelaciji kod roditelja i sada su pokazivala uzajamni odnos. Jedina razlika u odnosu na roditelje je u tome da se, za većinu osobina, intezitet međusobnog uticaja povećao.

AB  - For apicultural practice the productive traits of bee colonies are of great significance, since it is related to different geographical bee breeds. For the purpose of preservation of existing populations and natural diversity of honey bee and for the purpose of their further selection, researches upon the productive features of certain individual populations of choice at the territory of Republic of Serbia. During three years of research productive traits of six selected lines of honey bee from three different localities in Serbia (East, West and South Serbia) were studied. Through the survey of productive traits we have come to conclusion that there are significant variations among separate lines, which can be used to improve bee breeding program of the domestic honey bee and pick up the best lines for next generations.
AB  - U trogodišnjem ispitivanju, istraživane su najvažnije produktivne osobine izabranih linija medonosne pčele sa teritorije Republike Srbije. Najveća površina pčela utvrđena je u liniji 2 (u prvom i drugom prolećnom pregledu je iznosila: 1,89 i 5,24 rama, dok je u jesenjem bila 3,21 rama). Najveća površina legla u oba prolećna pregleda bila je najveća u liniji 2 i 5 (1,61 : 1,25, odnosno 5,06 : 5,15 rama), dok su u jesenjem pregledu sve linije imale približno istu površinu legla. U liniji 1 i 2 utvrđena je najveća prosečna površina meda i iznosila je 3,74, odnosno 3,68 rama. Površina polena u prolećnim pregledima bila je najveća u liniji 2 (0,54 i 0,42 rama).

For the purpose of preservation of existing populations and natural diversity of honey bee and for the purpose of their further selection, researches upon the productive features of certain individual populations of choice at the territory of Republic of Serbia. During three years of research productive traits of six selected lines of honey bee from three different localities in Serbia (East, West and South Serbia) were studied. Through the survey of productive traits we have come to conclusion that there are significant variations among separate lines, which can be used to improve bee breeding program of the domestic honey bee and pick up the best lines for next generations., U trogodišnjem ispitivanju, istraživane su najvažnije produktivne osobine izabranih linija medonosne pčele sa teritorije Republike Srbije. Najveća površina pčela utvrđena je u liniji 2 (u prvom i drugom prolećnom pregledu je iznosila: 1,89 i 5,24 rama, dok je u jesenjem bila 3,21 rama). Najveća površina legla u oba prolećna pregleda bila je najveća u liniji 2 i 5 (1,61 : 1,25, odnosno 5,06 : 5,15 rama), dok su u jesenjem pregledu sve linije imale približno istu površinu legla. U liniji 1 i 2 utvrđena je najveća prosečna površina meda i iznosila je 3,74, odnosno 3,68 rama. Površina polena u prolećnim pregledima bila je najveća u liniji 2 (0,54 i 0,42 rama).",

AB  - The dependence between traits is determined by the correlation coefficients. The experiment was conducted using two generations of honey bee. The first generation consisted of the mother queen bees, and the second were their offspring. The experiment lasted for two years per generation. In the first generation, the correlation between colony strength in the both spring and autumn inspection and the honey yield was high. Considering food supply, the high correlation between food supplies in autumn and honey yield was determined. In the offspring colonies, the high correlation was determined between colony strength in autumn and honey yield. The colony strength in the spring inspection and honey yield were in medium correlation. Also, the medium correlation was determined between honey yield and brood area in the spring inspection. Other studied characteristics in the offspring colonies were not significantly correlated with honey yield.
AB  - Stepen zavisnosti jedne osobine od druge utvrđuje se na osnovu koeficijenta korelacije. Ogled je sproveden na dve generacije društava medonosne pčele. U prvoj generaciji praćene su majke matice, a u drugoj njihovo potomstvo. Ispitivanje po svakoj generaciji trajalo je po dve godine. U prvoj generaciji utvrđena je visoka korelacija između snage društava na prolećnom jesenjem pregledu i prinosa meda po društvu. Kod zaliha hrane utvrđena je visoka korelacija između prinosa meda i zaliha hrane na jesenjem pregledu. Kod potomstva utvrđena je visoka korelacija između snage društava na jesenjem pregledu i prinosa meda. Snaga društva na prolećnom pregledu i prinos meda pokazuju srednji stepen korelacije. Takođe, utvrđen je i srednji stepen korelacije između prinosa meda i površine legla na prolećnom pregledu. Ostale praćene karakteristike potomačkih društava nisu pokazivale značajan stepen korelacije sa prinosom.

Nutrition of the honey bee

AB  - Many plants are excellent sources of nectar and pollen. However, when this food is not available, or short in supply, beekeepers have to supply artificial food to beehives. Nectar and honey are main carbohydrate sources and are used mainly for the production of energy. If there are shortfall suggests substitutes preferably as invert sugars or candies, produced by enzyme hydrolysis. Naturally collected pollen is the best source of protein for honey bee nutritional requirements and the best is collecting pollen from a number of different floral sources. Pollen is the sole source of lipids in the diet of honey bee. Sterols, especially 24-methylene chloresterol, has very important role in honey bee nutrition. Vitamins are required for optimum growth and young adult bees need the vitamin B complex for brood rearing. Water is collected by bees and used primarily as diluents for thick honey. Minerals are required by honey bees and the present elements in bee collected pollen was K, P and S.
AB  - Ishrana medonosne pčele je specifična i u uslovima proizvodnje važan činilac uspešnog pčelarenja. Veliki broj autora, istraživao je kompleksnost hraniva i mogućnost njihove adekvatne zamene, kao i različite načine dodavanja. Medonosna pčela sakuplja veliki broj supstanci i to: nektar koji odrasle jedinke skladište u ćelije saća i polen koji predstavlja izvor belančevina masti, vitamina i mineralnih materija. Ukoliko zalihe hrane u košnici nisu dovoljne, pčelama se dodaju različite zamene od kojih su najčešće šećernomedno testo, šećerni sirup obogaćen vitaminima, zamena polena i sl. ali se preko hrane može delovati i na parazite pčela, u prvom redu na varou.
