Characterization and population diversity of Erwinia amylovora strains originating from pome fruits in Serbia

AB  - The diversity of 30 Erwinia amylovora strains, isolated from quince, pear and apple trees on 14 localities in Serbia, was studied using bacteriological and molecular methods. In pathogenicity tests, all strains caused necrosis and oozing of bacterial exudate on inoculated immature pear, cherry and plum fruits, and induced hypersensitive reaction in tobacco leaves. The studied strains were Gram and oxidase negative, non-fluorescent, levan and catalase positive and facultatively anaerobic. The strains did not reduce nitrates, but utilized citrate and produced acid from sorbitol, hydrolyzed gelatine, produced reducing substances from sucrose and grew in the presence of 5% NaCl, but not at 36oC. Identity of the strains was confirmed by conventional and nested PCR methods. Rep-PCR with REP, ERIC and BOX primers resulted in amplification of several DNA fragments respectively, but showed no variation within the strains. However, different genetic profiles were obtained with RAPD-PCR by using six primers which enabled differentiation of the strains into four groups. Genetic differences between the studied strains did not correlate with the host plants, geographical origin or year of isolation.
AB  - Primenom standardnih bakterioloških i molekularnih metoda proučen je diverzitet 30 sojeva Erwinia amylovora izolovanih iz dunje, kruške i jabuke, poreklom iz 14 lokaliteta u Srbiji. Svi proučavani sojevi izazvali su nekrozu i pojavu bakterijskog eksudata na nesazrelim plodovima kruške, trešnje i šljive, kao i hipersenzitivnu reakciju duvana. Proučavani sojevi bili su Gram i oksidaza negativni, fakultativno anaerobni, levan i katalaza pozitivni i nisu stvarali fluorescentni pigment na Kingovoj podlozi B. Svi sojevi hidrolizuju želatin, koriste citrate i stvaraju kiselinu iz sorbitola, proizvode redukujuće supstance iz saharoze, ne redukuju nitrate, razvijaju se u prisustvu 5% NaCl, ali ne i pri 36°C. Identitet sojeva potvrđen je konvencionalnim PCR i nested PCR metodama. Rep-PCR metodom korišćenjem REP, ERIC i BOX prajmera umnoženo je više fragmenata DNK čiji broj i veličina su se podudarali kod svih proučavanih sojeva. Za razliku od Rep-PCR, primenom RAPD-PCR metode uz korišćenje šest prajmera došlo je do izdvajanja različitih genetičkih profila i diferencijacije sojeva u četiri grupe. Genetičke razlike među proučavanim sojevima nisu bile u korelaciji sa domaćinima iz kojih su izolovani, niti sa njihovim geografskim poreklom i godinom izolacije.

Proteinase PrtP impairs lactococcin LcnB activity in Lactococcus lactis BGMN1-501: new insights into bacteriocin regulation

AB  - Proteinases and bacteriocins are of great importance to the dairy industry, but their interactions have not been studied so far. Lactococcus lactis subsp. lactis BGMN1-5 is a natural isolate from homemade semi-hard cheese which produces two bacteriocins (Lactococcin B and LsbB), as well as proteinase PrtP. A medium-dependent increase in the bacteriocin LcnB activity of L. lactis BGMN1-501, a derivate of L. lactis subsp. lactis BGMN1-5, was shown to be accompanied by a decrease in its promoter activity. A similar effect of media components on gene expression was reported for proteinase PrtP whose gene is co-localized on the same plasmid as the IcnB gene. Thus, the PrtP-LcnB interplay was investigated. Single gene knockout mutants were constructed with disrupted prtP or IcnB genes. PrtP mutants showed higher bacteriocin activity that had lost its growth medium dependence, which was in contrast to the original strain. When LcnB from this mutant was combined with proteinase from the LonB(-) mutant in vitro, its activity was rendered to the original level, suggesting that proteinase reduces bacteriocin activity. We propose a new model of medium dependent expression of these genes with regard to the effects of their interaction in vivo.

Characteristics of lactobacilli growth and activity in UF milk

AB  - The application of ultrafiltration has an increasing importance in the manufacture of dairy products, primarily of soft cheeses. However, in semi-hard and hard cheeses, manufac­ture ultrafiltration has not been applied at industrial level yet. Considering that UF milk is characterised by increased buffer capacity, lactic acid bacteria, that could be used as starter cultures, must have an expressed acidogenic ability. Also, practice has shown that cheeses manufactured of UF milk ripen slowly, because it is necessary that starter cultures have an expressed and proteolytic activity. The buffer capacity of milk equivalent (CF=1) and retentates (CF=3 and CF=4) were studied. Furthermore, the influence of milk equivalent and these retentates on the growth and activity of two strains of lactic acid bacteria: Lactobacillus lielveticus HD-50 and Lactobacillus acidophilus MC. The growth and production of lactic acid of the tested strains in UF milk were being observed during eight hours of fermentation. Proteolytic activity of these strains was examined by determination of tyrosine value and electrophoretic detection prt+ strains which were capable to hydrolyse p casein. In strain iMctobacillus helveticus HD-50 was detected better acidogenic ability, but in strain Lactobacillus acidophilus MC was detected better proteolytic activity.
AB  - Primena ultrafiltracije ima sve veći značaj u proizvodnji mlečnih proizvoda, u prvom redu mekih sireva, dok je u proizvodnji polutvrdih i tvrdih sireva zastupljena u manjem obimu. S obzirom da se UF mleko karakteriše povećanim puferskim kapacitetom, bakterije mlečne kiseline koje bi se mogle koristiti kao starter kulture, moraju se odlikovati izraženom acidogenom sposobnošću. Takodje, praksa je pokazala da sirevi dobijeni iz UF mleka sporije sazrevaju, zbog čega je neophodno da starter kulture poseduju izražena i proteolitička svojstva. U radu je prikazan puferski kapacitet UF mleka različitog stepena koncentrovanja i to mlečnog ekvivalenta (CF=1) i retentata CF=3 i CF=4. Zatim je ispitivan uticaj UF mleka navedenih koncentracija na aktivnost i rast dva soja bakterija mlečne kiseline i to : Lactobacillus helveticus HD-50 i Lactobacillus acidophilus MC. Rast i acidogena sposobnost ispitivanih sojeva je praćena tokom osam sati fermentacije u mlečnom ekvivalentu i retenlatima. Proteolitička aktivnost ispitivanih sojeva je determinisana određivanjem tirozinske vrednosti i elektroforetskom detekcijom prt+ sojeva koji hidrolizuju P kazein. Ispitivanja su pokazala da soj Lactobacillus helveticus HD-50 ima bolju acidogenu sposobnost, a soj Lactobacillus acidophilus MC je pokazao bolju proteolitičku aktivnost.
