Sectorial resistance of plum to sharka (Plum Pox Virus)

AB  - One unusual aspect of resistance to sharka (Plum Pox Virus, PPV) has been discovered and studied in plum trees in the six consecutive years. The absence of sharka symptoms in some of primary branches of infected plum trees characterized as resistance. Symptoms appearance was observed and PPV presence was checked serologically by DAS- ELISA test as well as by grafting transmission. That aspect of resistance mainly appears in a portion of the tree crown, i.e. on some primary branches of young plum trees and it is diminishing in following years. That distinct resistance to sharka (PPV) was found in trees of Čačanska rodna, Stanley and Crvena ranka cvs. In selected plum trees in the parts with such resistance no sharka symptoms appeared nor PPV was proved. The absence of sharka symptoms as well as PPV in a portion of a plum tree otherwise diseased by sharka is considered as a sign of resistance. For such aspect of resistance the name “Sectorial Resistance” is proposed. It is continues more and detail research on sectorial resistance of plum to PPV are in progress.
AB  - Kod stabala šljive sorti Čačanska rodna, Stenli i Crvena ranka obolelih od virusa šarke šljive (VŠŠ, Plum pox virus - PPV) uočeno je prisustvo tipičnih simptoma u određenim delovima krune, a odsustvo u drugim. Izostank simptoma i virusa šarke šljive u delu krune obolelog stabla je nazvana “sektorijalna otpornost”. Okuliranje pupoljaka ovih sorti iz zaraženih delova krune u podlogu-džanariku rezultiralo je prisustvom virusa već u jednogodišnjim prirastima i manifestovanjem bolesti preko tipičnih simptoma u prvoj godini po kalemljenju. Međutim, okuliranje pupoljaka istih sorti iz sektorijalno zdravih delova krune, dalo je potpuno suprotan rezultat. Odsustvo VŠŠ proveravano je serološki DAS-ELISA testom. Na osnovu iznetog može se zaključiti da se kalemljenjem sektorijalna otpornost uspešno prenosi na nove serije sadnica i buduće voćke.

Izostank simptoma i virusa šarke šljive u delu krune obolelog stabla je nazvana “sektorijalna otpornost”. AB  - Otkrivena je i istraživana jedna posebna otpornost šljive na virus šarke šljive (VŠŠ, Plum pox virus - PPV) za koju se predlaže naziv 'sektorijalna otpornost'. Ta otpornost se ispoljava u delu krune, odnosno na pojedinim skeletnim granama obolelih stabala. U tom delu krune, odnosno na skeletnim granama koje pokazuju sektorijalnu otpornost nema simptoma oboljenja, niti je prisutan VŠŠ. Odsustvo VŠŠ proveravano je serološki DAS-ELISA testom i kalemljenjem. Sektorijalna otpornost je nađena kod stabala Čačanska rodna, Stenley-a i Crvene ranke. Sektorijalna otpornost šljive na VŠŠ je u negativnoj korelaciji sa starošću stabala na kojima se javlja. Broj skeletnih grana sa sektorijalnom otpornošću kod obolelih stabala se smanjuje iz godine u godinu, a povećava broj skeletnih grana sa simptomima oboljenja. U toku su dalja i svestranija proučavanja te novootkrivene otpornosti šljive prema VŠŠ.

Identification of Rhizoctonia spp. using polyclonal antiserum in ebia

AB  - A polyclonal antiserum (As-1) raised against total mycelial antigens of binu-cleate Rhizoctonia AG-I was used in EBIA in order to determine antigenic properties and serological relationship of some binucleate and multinucleate Rhizoctonia spp. After separation by electrophoresis and based on the reactions with antiserum As-1, it has been established that a total of 18 protein fractions show antigenic properties (14.4-94kDa). The number and patterns of protein bands differed in binucleate and multinucleate Rhizoctonia isolates and their anastomosis groups (AG). Protein fraction designated as number 8 was common to most of the isolates but the profiles of the remaining bands pointed out serological differences. The results show that the use of polyclonal antiserums can be helpful in detection of different Rhizoctonia spp., but, what is more important, when a polyclonal antiserum is used in EBIA, it is possible to differentiate anastomosis groups of binucleate or multinucleate Rhizoctonia spp.
AB  - Antigeni kompleks i serološki međuodnosi odabranih dvojedarnih i višejedarnih izolata Rhizoctonia spp. ispitivani su, primenom EBIA metode, na osnovu reaktivnosti sa antiserumom pripremljenim na ukupne proteine micelije izolata dvojedarne Rhizoctonia AG-I. Elektroforetskim razdvajanjem utvrđeno je da antigena svojstva poseduje ukupno 18 različitih proteinskih komponenti ispitivanih izolata (14,4 - 94 kDa). Ustanovljenje različit broj i raspored proteinskih traka kako kod dvojedarnih i višejedarnih izolata, tako i kod ana-stomoznih grupa (AG). Proteinska frakcija obeležena sa 8 bila je zajednička za najveći broj ispitivanih izolata Rhizoctonia spp., dok je profil ostalih traka ukazao na značajne serološke razlike. Obavljena ispitivanja su pokazala da se primenom poliklonalnog antiseruma mogu detektovati različite Rhizoctonia spp., kao i da se primenom poliklonalnog antiseruma u EBIA metodi mogu identifikovati anastomozne grupe dvojedarnih odnosno višejedarnih Rhizoctonia spp.

The role of relative humidity in hypersentive (shock) symptoms in sorghum infected with maize dwarf mosaic virus

AB  - Based on the results of these experiments we concluded that: The necrotic red stripe disease on MDMV-A infected sorghum plants, in addition to genotype sensitivity, is influenced by plant age, temperature and relative humidity of air; Young sorghum plants are more likely to develop necrotic red stripe symptoms than older ones; In 100% R.H. necrotic red stripe symptoms appear earlier, they are more severe and develop on a larger number of plants.
AB  - Nekrotična crvena prugavost spada među najznačajnije bolesti sirka. Izaziva potpunu ili delimičnu nekrozu lišća. Obolele biljke se najčešće delimično oporavljaju, posle čega usporeno rastu i ostvaruju umanjen prinos slabijeg kvaliteta. Nekrotičnu crvenu prugavost lišća sirka izaziva soj A virusa mozaične kržljavosti kukuruza (VMKK-A). Ta se bolest, uglavnom, javlja pri određenim ekološkim uslovima. Prema rezultatima koje su objavili prethodni istraživači, nekrotična crvena prugavost se javlja kada se mozaične biljke sirka zaražene VMKK nađu u uslovima nižih tempepratura, odnosno kada u uslovima u polju dođe do zahlađenja. Ispitivanjima koja su obavljena u toku ovog rada, utvrđeno je sledeće: - mlađe biljke sirka, koje su zaražene VMKK podložnije su nekrotičnoj (šok) reakciji (tab.l), - nekrotična (šok) reakcija kod biljaka sirka zaraženih VMKK češće se javlja na nižim temperaturama, visoka relativna vlažnost, odnosno zasićenost vazduha vodenom parom, je najznačajniji faktor koji uslovljava nekrotičnu (šok) reakciju kod biljaka sirka za raženih VMKK, nekrotična (šok) reakcija se ispoljava i kod mladih biljaka sirka zaraženih VMKK i na temperaturama iznad 20°C, ali u uslovima 100% relativne vlažnosti, - najčešća i najintenzivnija nekrotična (šok) reakcija kod biljaka sirka za raženih VMKK, utvrđena je kada su niže temperature bile istovremeno sa visokom relativnom vlažnošću vazduha (100% R.H) (tab.l).

Ispitivanjima koja su obavljena u toku ovog rada, utvrđeno je sledeće: - mlađe biljke sirka, koje su zaražene VMKK podložnije su nekrotičnoj (šok) reakciji (tab.l), - nekrotična (šok) reakcija kod biljaka sirka zaraženih VMKK češće se javlja na nižim temperaturama, visoka relativna vlažnost, odnosno zasićenost vazduha vodenom parom, je najznačajniji faktor koji uslovljava nekrotičnu (šok) reakciju kod biljaka sirka za raženih VMKK, nekrotična (šok) reakcija se ispoljava i kod mladih biljaka sirka zaraženih VMKK i na temperaturama iznad 20°C, ali u uslovima 100% relativne vlažnosti, - najčešća i najintenzivnija nekrotična (šok) reakcija kod biljaka sirka za raženih VMKK, utvrđena je kada su niže temperature bile istovremeno sa visokom relativnom vlažnošću vazduha (100% R.H) (tab.l).",
