Effect of dicamba on wheat haploid embryo development

AB  - An interspecies zygote derives after wheat x maize crosses. This zygote aborts in absence of auxin stimulation. Treatment with dicamba is widely used to promote wheat haploid embryo development. We analyzed the effect of dicamba on wheat haploid embryo development. Four treatments related to the time of dicamba treatment were carried out: I day before pollination and 1, 5 and 9 days after pollination. It was concluded that wheat haploid embryos developed only in spikes treated with dicamba one or five days after pollination with maize. Dicamba. treatment of wheat spikes prior to pollination hinders zygote formation and embryo development. Dicamba treatment 9 days after pollination is too late because young embryos have already aborted.

Uniparental chromosome elimination at mitosis and interphase in wheat and pearl millet crosses involves micronucleus formation, progressive heterochromatinization, and DNA fragmentation

AB  - Complete uniparental chromosome elimination occurs in several interspecific hybrids of plants. We studied the mechanisms underlying selective elimination of the paternal chromosomes during the development of wheat (Triticum aestivum) x pearl millet (Pennisetum glaucum) hybrid embryos. All pearl millet chromosomes were eliminated in a random sequence between 6 and 23 d after pollination. Parental genomes were spatially separated within the hybrid nucleus, and pearl millet chromatin destined for elimination occupied peripheral interphase positions. Structural reorganization of the paternal chromosomes occurred, and mitotic behavior differed between the parental chromosomes. We provide evidence for a novel chromosome elimination pathway that involves the formation of nuclear extrusions during interphase in addition to postmitotically formed micronuclei. The chromatin structure of nuclei and micronuclei is different, and heterochromatinization and DNA fragmentation of micronucleated pearl millet chromatin is the final step during haploidization.

Influence of time of auxin application on wheat haploid embrio formation

AB  - A hybrid interspecies zygote appears after crosses between wheat and maize Zygote derived after usual self-fertilization in wheat is dividing by mitotic divisions into embryo. However, interspecies zygote aborts soon. Auxin treatment is widely used to promote its development. Growth hormones auxins have stimulative ortoxic effects on plant tissue sin relation to its concentration and the time of application. In this paper the effect of time of auxin dicamba application on embryo in wheat x maize crosses was investigated. Chromosomes of pollen donor parent are eliminated quickly in cells of such embryos and they become haploid. It was concluded that for the production of haploid embryos the best time for auxin application is one day after pollination with maize.
AB  - Posle ukrštanja pšenice sa kukuruzom nastaje hibridni interspecies zigot Zigot nastao uobičajenom samooplodnjom pšenice deli se mitotičkim deobama i formira embrion. Međutim, interspecies zigot relativno brzo abortira. Da bi se podstakao njegov razvoj koristi se auksinski tretman. Auksini su hormoni rasta koji u zavisnosti od koncentracije i vremena primene imaju stimulativni ili toksični efekat na razvoj biljnih tkiva. U ovom radu analiziranje efekat vremena primene auksina dikamba u koncentraciji od 100 ppm na formiranje embriona pšenice dobijenih posle ukrštanja sa kukuruzom. U ćelijama takvih embriona tokom razvoja brzo se gube hromozomi donora polena, te oni postaju haploidni embrioni. Zaključeno je da za proizvodnju haploidnih embriona optimalno vreme tretiranja klasova pšenice je jedan dan posle oprašivanja polenom kukuruza.
