AB  - In this study, the polyphenolic profile of 11 Morus alba fruits grown in the Vojvodina region was investigated. Ultra high performance liquid chromatography (UHPLC) coupled with Linear Trap Quadrupole and OrbiTrap mass analyzer, and UHPLC coupled with a diode array detector and a triple-quadrupole mass spectrometer were used for the identification and quantification of the polyphenols, respectively. A total of 14 hydroxycinnamic acid esters, 13 flavonol glycosides, and 14 anthocyanins were identified, in the extracts with different distributions and contents according to the sampling. The total phenolic content ranged from 43.84 to 326.29 mg GAE/100 g frozen fruit. The radical scavenging capacity (50.18-86.79%), metal chelating ability (0.21-8.15%), ferric ion reducing power (0.03-38.45 mu M ascorbic acid) and superoxide anion radical scavenging activity (16.53-62.83%) were assessed. The findings indicated that mulberry polyphenolics may act as potent superoxide anion radical scavengers and reducing agents.

Peroxidase isoenzyme polymorphism in the genus Prunus, subgenus Cerasus

AB  - The polymorphism of peroxidase was studied in 31 cherry accessions, representing six following species Prunus cerasus, Prunus avium, Prunus fruticosa, Prunus mahaleb, Prunus serrulata, Prunus gondouinii and two widely-used standard cherry rootstocks 'Gisela 5' and 'Colt'. Six 'Oblačinska' sour cherries, four wild sweet cherries, five ground and one mahaleb genotypes were selected from the natural populations of Serbia. Inner barks from one-year-old shoots and young actively growing leaves were used for protein extraction. The polymorphism of peroxidase was obtained both for leaf and inner bark tissues. The analysis of the leaf material showed the unique zymograms for all six species and two interspecies hybrids. Higher numbers of polymorphic loci and banding patterns were detected when protein was extracted from the leaves, than from inner bark. Obtained results indicate that the polymorphism determination of genus Prunus, subgenus Cerasus can be done on the basis of peroxidase, but it would not be useful for discrimination of different genotypes and clones. .
AB  - Polimorfizam peroksidaza proučavan je kod 31 genotipa roda Prunus u okviru šest vrsta podroda Cerasus: Prunus cerasus, Prunus avium, Prunus fruticosa, Prunus mahaleb, Prunus serrulata i Prunus gondouinii, kao i kod dva interspecies hibrida koji se koriste kao standardne podloge za trešnju: Gisela 5 i Colt. Šest genotipova Oblačinske višnje, četiri divlje trešnje, pet genotipova stepske višnje i jedan genotip magrive su selekcionisani iz prirodnih populacija u Srbiji. Za ekstrakciju proteina korišćena je unutrašnja kora jednogodišnjih grančica i mlado lišće. Polimorfizam peroksidaza je uočen u oba tkiva. Analizom lista dobijeni su jedinstveni peroksidazni zimogrami za svaku od ispitivanih vrsta i oba interspecijes hibrida. Veći broj polimorfnih lokusa i tipova zimograma utvrđen je kada je za proteinski ekstrakt korišćeno lišće, u poređenju sa unutrašnjom korom jednogodišnjih grančica. Dobijeni rezultati ukazuju da je na osnovu polimorfizma peroksidaza moguće determinisati vrste roda Prunus, podrod Cerasus, ali ne i utvrditi razlike među genotipovima i klonovima. .

AB  - Dehydrogenase polymorphism was studied in 36 sour cherry (Prunus cerasus L.), sweet cherry (Prunus avuim L.), mahaleb (Prunus mahaleb L.), ground cherry (Prunus fruticosa Pall.), duke cherry (Prunus gondounii Redh.), Japanese flowering cherry (Prunus serrulata Lindl.) and four iterspecific hybrids (standard cherry rootstocks ‘Gisela 5’, ‘Gisela 6’, ‘Max Ma’ and ‘Colt’). Inner bark of one-year-old shoots, in dormant stage, was used for enzyme extraction. Vertical PAGE was used for isoenzyme analysis: alcohol dehydrogenase (ADH), formate dehydrogenase (FDH), glutamate dehydrogenase (GDH), isocitrate dehydrogenaze (IDH), malate dehydrogenase (MDH), phosphogluconate dehydrogenase (PGD), and shikimate dehydrogenase (SDH). All studied systems were polymorphic at 10 loci: Adh -1 (3 genotypes) and Adh-2 (5 genotypes), Fdh-1 (2 genotypes), Gdh-1 (3 genotypes), Idh-1 (4 genotypes) i Idh -2 (5 genotypes), Mdh-1 (3 genotypes), Pgd-1 (4 genotypes), Sdh-1 (1 genotype) i Sdh-2 (3 genotypes). Cluster analysis was used to construct dendrogram on which four groups of similar genotypes were separated. Obtained results indicate that studied enzyme systems can be used for determination of genus Prunus, subgenus Cerasus. Among studied enzyme systems ADH, IDH and SDH were the most polymorphic and most useful to identify genetic variability. Polymorphism of FDH and GDH in genus Prunus, subgenus Cerasus was described first time in this work. First results for dehydrogenase variability of Oblačinska indicate that polymorphism of loci Idh-2 and Sdh-2 can be useful for discrimination of different clones.

Metoda vertikalne poliakrilamidne gel elektroforeze (PAGE) korišćena je za razdvajanje proteina za analizu enzimskih sistema: ADH (alkohol dehidrogenaze), FDH (format dehidrogenaze), GDH (glutamat dehidrogenaze), IDH (izocitrat dehidrogenaze), MDH (malat dehidrogenaze), PGD (fosfoglukonat dehidrogenaze) i SDH (šikimat dehidrogenaze). Polimorfizam je utvrđen za sve proučavane sisteme. Prvi rezultati o varijabilnosti dehidrogenaza kod Oblačinske višnje ukazuju da polimorfizam lokusa Idh-2 i Sdh-2 može biti koristan za identifikaciji klonova.

Objectives, methods and accomplishments in fruit and grape breeding

AB  - The most important objectives, methods and accomplishments in fruit and grape breeding are showed in this study. Yield, quality and resistance to plant diseases and pests are underlined among numerous breeding aims. Besides, review of specific targets was also listed. The most conventional and new biotechnological breeding methods were described. A lot of accessory methods, which in combination with the conventional ones are contributing faster achievement of set goals, are also mentioned. Accomplishments of many breeding programs showed that planned work contributed creating numerous important cultivars and rootstocks in all fruit and grapevine species. Those cultivars are pushing out the standard ones and replacing them in new plantations. All of this is showing that breeding work throughout the world and in our country should go on, while only new cultivars and rootstocks with high genetic potential can provide, with certain conditions, high income in fruit production and viticulture.
AB  - U radu su prikazani najvažniji ciljevi, metode i dostignuća u oplemenjivanju voćaka i vinove loze. Među ciljevima oplemenjivanja posebno su istaknuti prinos, kvalitet i otpornost prema izazivačima bolesti i štetočinama. Pored njih dat je i prikaz pojedinih specifičnih ciljeva. Od metoda oplemenjivanja u prvom redu obrađene su konvencionalne i nove metode biotehnologije. Određena pažnja usresređena je i na brojne pomoćne metode, koje u kombinaciji sa konvencionalnim, doprinose bržem ostvarivanju postavljenih ciljeva. Dostignuća mnogih oplemenjivačkih programa pokazala su da su planskim radom na oplemenjivanju stvorene brojne privredno značajne sorte i podloge kod svih vrsta voćaka i vinove loze. Mnoge od njih potiskuju iz proizvodnje standardne sorte i zauzimaju odgovarajuća mesta u novim zasadima. Sve ovo ukazuje da oplemenjivački rad kako u svetu tako i u našoj zemlji treba dalje nastaviti, jer samo nove sorte i podloge velikog genetičkog potencijala mogu, uz odgovarajuće druge uslove, obezbediti visok dohodak u voćarskoj i vinogradarskoj proizvodnji.

abstract = "The most important objectives, methods and accomplishments in fruit and grape breeding are showed in this study. Yield, quality and resistance to plant diseases and pests are underlined among numerous breeding aims. Besides, review of specific targets was also listed. The most conventional and new biotechnological breeding methods were described. A lot of accessory methods, which in combination with the conventional ones are contributing faster achievement of set goals, are also mentioned. Accomplishments of many breeding programs showed that planned work contributed creating numerous important cultivars and rootstocks in all fruit and grapevine species. Those cultivars are pushing out the standard ones and replacing them in new plantations. AB  - The exciting challenge for all breeders is to release cultivars which the market wants and which can produce a profit for the growers. They have done it in the past and they will continue to do it in the future. Today's fruit breeding program are interrelated with other research disciplines. The complex, dynamic and interdisciplinary interactions require innovative approaches, including modern tools and techniques. The most important areas of collaboration are: database management, genetics, phytopathology entomology, ecology, physiology, molecular biology, fruit quality and sensory perception. Breeders also need to be aware of modern fruit marketing and consumer requirements. The complexity of tasks in modern fruit breeding sometimes leads to the specialization of particular breeding program and a dynamic interaction between complementary programs. Responses from fruit breeders about genetic transformation ranged from strongly negative to actively positive. Fruit breeders are constantly challenged to consider new tools and technology to increase the efficiency of their programs.
AB  - Generalni, najvažniji izazov oplemenjivačkog rada je stvaranje novih sorti koje će prihvatiti tržište i koje donose profit proizvođačima. Selekcioneri su uspešno ispunjavali ove najviše ciljeve do sada i nema sumnje da će nastaviti da to rade i ubuduće. Današnji oplemenjivački programi su interdisciplinarni. Kompleksnost, dinamičan razvoj i interdisciplinarna interakcija zahtevaju nove prilaze, inventivne i dobro obučene kadrove savremene metode i opremljenost laboratorija. Najvažnije oblasti saradnje predstavljaju baze podataka, genetika, fitopatologija, entomologija fiziologija, ekologija, molekularna biologija, mehanizacija proizvodnje kvalitet ploda i senzorsko ocenjivanje. Oplemenjivači moraju biti svesni modernih zahteva tržišta i potrošača. Kompleksnost ciljeva vrlo često vodi ka specijalizaciji pojedinih oplemenjivačkih programa i dinamičkoj interakciji komplementarnih programa. Odnos oplemenjivača prema genetički modifikovanim organizmima ide od negativnog do pozitivnog. Stalan izazov pred naučnim radnicima je spoznaja novih metoda i tehnologija kako bi unapredili efikasnost svojih oplemenjivačkih programa.
